rs2357322 and rs17726078 Contribute to Breast Cancer Risk by Regulating DLX2 Expression.

BACKGROUND AND AIMS: Breast cancer is the most abundant cancer type in female. Genome-wide association study suggests that rs2016394, one SNP at the intron of DLX2 divergent transcript (DLX2-DT), is significantly associated with this disease. Through 1000 genomes project data analysis, it is observed that another three SNPs, rs743605, rs2357322, and rs17726078, show strong linkage disequilibrium with rs2016394. However, the functional SNP(s) and mechanism are still unknown. METHODS: Functional genomics effort was performed for this locus. RESULTS: Through luciferase assay, it is disclosed th

B cell receptor-based hybridoma enrichment enables discovery of antibodies against intracellular tumor antigens.

Tumor-associated B cell responses are frequently observed in tumors and their draining lymph nodes, yet their antigen specificities and functional potential remain incompletely defined. Historically, intracellular antigens have been largely disregarded in tumor antigen mining and discovery efforts. In particular, the extent to which intracellular tumor antigens can be effectively targeted by antibodies and the cellular immune system has remained unclear, poorly characterized, and therapeutically underexploited. We developed a high-throughput platform combining hybridoma generation with unbiase

Neoadjuvant chemotherapy in women with triple-negative breast cancer: real world results from Western Kenya.

BACKGROUND: Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype for which neoadjuvant chemotherapy (NACT) is used for locally advanced (LA) disease. Patients with TNBC who experienced a pathologic complete response (pCR) following NACT have been found to have extended overall survival (OS) compared to patients with residual disease. Sparse data exist on the frequency and impact of pCR following NACT for TNBC in East Africa. METHODS: We retrospectively reviewed patients diagnosed with LA TNBC who received NACT between 2018 and 2022 at Moi Teaching and Referral Hospital

[Effect of ESR1 Somatic Mutations on SERM/SERD Binding: Insights from Molecular Docking and Molecular Dynamics Simulations].

Acquired resistance to endocrine therapy poses a major limitation in the treatment of estrogen receptor (ER)-positive breast cancer (BC). Somatic mutations of the ESR1 gene, which encodes ERα, are one of the key resistance mechanisms. However, the effect of mutations and especially double mutations on the binding of novel drugs remains insufficiently understood. To evaluate the effect of the mutations at the molecular level, a comprehensive computational approach was employed, including molecular docking, binding free energy calculations (MM-GBSA), and molecular dynamics simulations. Climate-sensitive exposures and cancer outcomes: a systematic review and meta-analysis.

Climate change is intensifying environmental exposures - wildfire smoke, ambient air pollution, extreme heat, and weather-driven disasters - with plausible but incompletely quantified consequences for cancer, and no prior synthesis has jointly examined them across cancer incidence, mortality, survival, and recurrence. We systematically searched five databases for observational studies up to May 2026 of wildfire, ambient temperature, hurricane or multi-hazard disaster, precipitation anomaly, or ambient PM₂.₅ exposure and cancer outcomes, and pooled 25 effect estimates from 14 studies within ten

Comment on "Effects of a sensorimotor-based, movement quality-focused intervention on functional mobility in breast cancer survivors: a multicenter randomized controlled trial".

This correspondence comments on the multicenter randomized trial of the Rehabilitation through Movement and Perception (ReMAP) program in breast cancer survivors. Although the study highlights the potential value of sensorimotor-focused rehabilitation, clarification is needed regarding participant accounting, the intention-to-treat analysis, the clinical interpretation of the Timed Up and Go outcome, control-group exposure, and the proposed mechanism linking movement quality to functional mobility.

MIF-Induced CD74+ Microglia and Macrophages Promote Progression of Brain Metastasis and Are Clinically Relevant across Central Nervous System Disorders.

UNLABELLED: The upregulation of CD74, a chaperone involved in MHC-II antigen processing, has been mostly interpreted as indicative of antigen presentation in multiple brain disorders. However, CD74 expression has also been described in cancer cells across multiple tumor types and in the tumor microenvironment, notably in glioma. In this study, we found that the presence of CD74+ microglia/macrophages, which was induced by increased levels of interferon γ in brains affected by metastases, did not relate to its canonical pathway. Instead, the alternative function of CD74 as a cytokine receptor w

A Three-Dimensional Hyaluronic Acid-Based Scaffold Model for Electroporation-Enhanced Resveratrol Therapy in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) is an aggressive malignancy defined by the absence of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor 2 (HER2). These molecular characteristics render targeted therapies less effective and contribute to poor clinical outcomes. Towards this, we studied the electroporation-mediated delivery of resveratrol, a polyphenolic compound with pleiotropic anticancer activities, on MDA-MB-231, human TNBC cell line. Both conventional two dimensional (2D) monolayer cell culture and three-dimensional (3D) hyaluronic acid HA-E

Structure-Function Regulation of Proteolytically Stable Amphiphilic Peptide-Based Soft Materials With Antimicrobial Efficacy Against Drug-Resistant Strains and Anticancer Activity in Triple-Negative Breast Cancer Cells.

The rapid progress of multidrug-resistant (MDR) bacterial infections and the clinical aggressiveness of triple-negative breast cancer (TNBC) possess a great problem to human beings. It necessitates the discovery and development of multifunctional therapeutic biomaterials. Herein, we report two enzymatically stable short cationic peptide hydrogelators comprising of N-terminal lysine headgroups, non-coded amino acid spacers, aromatic phenylalanine residues, and dodecylamine tails. These amphiphilic peptides are rapidly self-assembled in Tris-HCl buffer (pH 7.4) to form hydrogels with nanofibrill

Editorial: Toward Prognostic Precision in the Management of Gastroenteropancreatic Neuroendocrine Neoplasms.

Gastroenteropancreatic neuroendocrine neoplasms (GEP-NENs), which encompass well-differentiated neuroendocrine tumors (NETs) and poorly differentiated neuroendocrine carcinomas (NECs), are a clinically and pathologically heterogeneous group of diseases with an increasing incidence, presenting significant challenges in diagnosis and treatment. This Special Collection brings together studies examining GEP-NEN management from four distinct perspectives: clinicopathological prognostic factors, sex-related survival differences, comparative real-world use and outcomes of long-acting somatostatin ana

Bidirectional phenotypic transition in macrophages influences triple-negative breast cancer cell immune evasive patterns and response to chemotherapies in a co-culture spheroid model.

Tumor-associated macrophage (TAM) infiltration is a critical characteristic of triple-negative breast cancer (TNBC) related to drug resistance and poor prognosis. Integrating macrophages into TNBC spheroids is crucial to improve the accuracy of 3D in vitro models that mimic the complexity of the tumor microenvironment (TME) and assess treatment response. However, this remains challenging since the reciprocal effects of these two cell types on each other are not fully understood. In this study, we used the TNBC cell line, MDA-MB-231, and polarized M1-like or M2-like macrophages derived from THP

The American Joint Committee on Cancer Prognostic Staging System for Breast Cancer: Sustained Relevance of Anatomic Criteria on 5-Year Survival.

PURPOSE: In 2018, the American Joint Committee on Cancer (AJCC) revised the TNM staging system for breast cancer (BC), incorporating biological factors into anatomical staging. This study evaluated the clinical impact of this integration and the relative prognostic contribution of anatomical and biological factors to 5-year overall survival (OS). METHODS: A retrospective analysis was conducted on a cohort of 1075 patients diagnosed with BC at the Breast Centre of São João University Hospital (2015-2020), using clinicopathological data from a EUSOMA-certified database. The primary endpoint was

Time-dependent prognostic impact of tumor grade in stage I-III breast cancers.

BACKGROUND: Breast cancer (BC) mortality risk extends over decades; yet, the temporal prognostic patterns of histological grade across subtypes and stages remain unclear. PATIENTS AND METHODS: Women with stage I-III BC diagnosed between 2000 and 2018 were identified from Surveillance, Epidemiology, and End Results registries (N = 767 218). Breast cancer-specific mortality (BCSM), as the primary endpoint, was analyzed using cumulative incidence functions including competing risk and annual hazard rates stratified by grade, estrogen receptor (ER) status, stage, and adjuvant chemotherapy. Phenotype-driven screening of semi-synthetic terpene analogs identifies TRX-41 as a lead compound active in chemotherapy-resistant breast cancer organoids.

Natural products remain an important source of anticancer agents, yet identifying pharmacological vulnerabilities in chemotherapy-refractory tumors remains a major challenge. Natural terpenes (NT) represent one of the most structurally diverse classes of bioactive compounds, but their native scaffolds often display limited potency and restricted translational potential. Here, we applied a phenotype-driven screening strategy using a library of semi-synthetic terpene analogs to identify compounds capable of targeting therapy-resistant tumor states. A first-generation library of 137 semi-syntheti

Awake endoscopic mastectomy in one-day-surgery: Preliminary results.

BACKGROUND: While enhanced recovery after surgery (ERAS) and awake monitored anaesthesia care (MAC) have been described to promote same-day discharge (SDD) in conventional mastectomy, their application in reverse-sequence endoscopic nipple-sparing mastectomy (R-E-NSM) has not yet been established. We present our protocol and early experience with awake R-E-NSM. METHODS: In this retrospective pilot case series, from February 2026 to April 2026, all consecutive awake R-E-NSM performed at Tor Vergata University Hospital were retrieved from a prospectively maintained database (Approval number 62.

Peri-operative CDK4/6 inhibitors in DIEP flap breast reconstruction: Early safety data and proposed management strategy.

The introduction of cyclin-dependent kinase 4/6 (CDK4/6) inhibitors has significantly altered the treatment paradigm for hormone receptor-positive, HER2-negative breast cancer. Increasingly, surgeons encounter patients receiving these agents at the time of breast reconstruction, yet there is no published evidence on the safety of free tissue transfer in this context. We present the first series examining post-operative outcomes in patients exposed to CDK4/6 inhibitors undergoing deep inferior epigastric perforator (DIEP) flap breast reconstruction and outline a pragmatic management approach. W

The GCN5L2-SIX1 complex drives glycolysis to promote progression of triple-negative breast cancer.

Lactylation, a lactate-dependent posttranslational modification, is involved in cancer development. Here, we showed that GCN5L2 physically interacts with nuclear SIX1 and acts as a key regulator of SIX1 lactylation. GCN5L2 depletion promotes APC/Cdh1 binding to SIX1, increases SIX1 ubiquitination, and reduces SIX1 protein stability. Functionally, the GCN5L2-SIX1 axis promotes glucose uptake, glycolysis process, and energy supply in TNBC cells. GCN5L2 knockdown attenuates the growth and migration of TNBC cells, and increases the sensitivity of TNBC cells to paclitaxel. Overexpression of SIX1 re
