In Vivo LncRNA Inhibition in Human Tumor Xenografts Using Antisense Locked Nucleic Acid GapmeRs.

Long noncoding RNAs (lncRNAs) have emerged as significant regulators of many physiological and pathological processes and play a key role in the onset and progression of a variety of diseases, including cancer. Currently, the functions of the majority of lncRNAs in carcinogenesis are still unclear. Functional characterization of specific lncRNAs is often challenging using the conventional RNA interference method, due to their specific and restricted subcellular localization. Antisense locked nucleic acid (LNA) GapmeRs, which induce RNase H1-mediated RNA cleavage, provide a promising strategy t

Pathway and Kinase Analysis in Proteomics and Phosphoproteomics with rROMA.

Advances in data acquisition tools, particularly mass spectrometry-based techniques, have made high-throughput methods like proteomics and phosphoproteomics increasingly accessible. However, the volume and complexity of the data require specialized computational tools for analysis and interpretation to extract meaningful insights. Here, we describe the use of rROMA software to analyze pathway and kinase activity in proteomic and phosphoproteomic datasets. rROMA is an open-source package available on Github at: www.github.com/sysbio-curie/rROMA . As a case study, we applied rROMA to breast canc

Lipid Composition in the Breast of Premenopausal BRCA1/2 Genetic Mutation Carriers Using MRI.

Deregulation of lipid composition in the breast has been suggested as an indicator of the risk of developing breast cancer, and chemical shift-encoded imaging (CSEI) in MRI allows rapid lipid composition mapping of the whole breast. We set out to examine if breast lipid composition in BRCA1 and BRCA2 gene carriers differed from women with breast cancer and controls. Eighty-two premenopausal female participants, 30 BRCA1/2 carriers, 37 women with breast cancer, and 15 controls, participated in this study. Monounsaturated, polyunsaturated, and saturated fatty acids (MUFAs, PUFAs, SFAs) in the br

The Catalytically Inactive KDM5B-NTT Isoform Associates with Chromatin and Increases H3K4 Trimethylation in Breast Cancer Cells.

Histone lysine demethylases are mainly known as transcriptional regulators for their demethylase activity, but catalysis-independent functions are increasingly recognized. This work focuses on lysine-specific demethylase 5B (KDM5B), targeting H3K4me3. The canonical functional isoform is PLU-1. A catalytically inactive N-terminally truncated isoform (NTT) was previously identified in human cell lines. NTT lacks the Jumonji N-terminal (JmjN) domain, responsible for demethylase activity, and part of the AT-Rich Interaction Domain (ARID), but retains the plant homeodomains (PHDs) involved in chrom

Design, Synthesis, and Biological Evaluation of Quinolin-2-One-Indole Schiff Base Hybrids as Potential EGFR-Targeted Anticancer Agents Against Breast and Ovarian Cancer.

A series of new quinolin-2-one-indole Schiff base hybrids was designed, synthesized, and tested as potential anticancer agents targeting the epidermal growth factor receptor. The designed compounds were synthesized via a multistep synthetic route comprising phenylhydrazone formation, Fischer indole cyclization, Vilsmeier-Haack formylation, and subsequent Schiff base condensation. Structural elucidation was performed using IR, 1H NMR, 13C NMR, and mass spectral analysis. In silico studies demonstrated that the synthesized compounds exhibited strong binding affinities to both wild-type (PDB ID:

Evaluation of the radiomodifying effects of Ginkgo biloba in radioresistant triple negative breast cancer cells.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype characterized by limited therapeutic options and intrinsic resistance to ionizing radiation. Ginkgo biloba (GB) possesses documented anticancer, antioxidant and pro-apoptotic properties; however, its potential role as a radiomodifying agent remains insufficiently investigated. The present study evaluated the effects of GB on the radiation response of the radioresistant MDA-MB-231 TNBC cell line. Cells were treated with increasing concentrations of GB (10-350 μg/mL) either alone or in combination with a clinically relev

Exosomes derived from itaconate-treated macrophages promote epithelial-mesenchymal transition, migration, and invasion in MCF-7 breast cancer cells.

Itaconic acid is an immunometabolite that regulates macrophage polarization within the tumor microenvironment and influences tumor progression and invasion. Although its anti-cancer and tumor-promoting effects remain controversial, its role in modulating macrophage-derived exosomes and their impact on breast cancer cells is not well understood. This study investigated the effects of exosomes derived from dimethyl itaconate (DMI)-treated macrophages on epithelial-mesenchymal transition (EMT), apoptosis, migration, and EMT-related miRNAs in MCF7 breast cancer cells. U937 macrophages were treated

Comment on "Associations of neighborhood deprivation with breast cancer tumor genomics, targeted treatment use, and survival".

INTRODUCTION: Podany et al. examined associations between Area Deprivation Index (ADI) and tumor genomics, PI3K inhibitor use, and survival in 1,127 patients with metastatic breast cancer. While their multi-institutional, genomically annotated dataset is a valuable contribution, several methodological issues warrant scrutiny. METHODS: We critically reviewed the study's statistical approach, focusing on multiple-comparison handling, potential confounding from ctDNA shedding variability, and the reliability of subgroup survival estimates. RESULTS: Key findings (TP53, AKT1, overall survival, PI

Breast Microbiome Dysbiosis in Plasma Cell Mastitis: A Ribosomal RNA Gene Sequencing Study.

Plasma cell mastitis (PCM) is a chronic breast condition with limited effective treatment options. Surgical intervention is a standard treatment approach but may be associated with postoperative complications. Recent studies suggest that alterations in the microbiome may be associated with the onset, progression, and recurrence of PCM. In this study, high-throughput 16S ribosomal RNA (rRNA) gene sequencing was used to compare the breast tissue microbiota of patients with PCM with that of the non-PCM control (NC) group. A total of 86 participants were enrolled. No significant differences were o

Engineering Poly(ε-caprolactone)-Based Maleimide-Enediyne Micelles for Next-Generation Anticancer Therapies.

Inspired by the DNA-cleaving potency of natural enediynes such as calicheamicin, we report the design and synthesis of maleimide-enediyne conjugates integrated into poly(ε-caprolactone) (PCL) as a next-generation platform for anticancer drug delivery. Maleimide-based enediynes typically suffer from poor aqueous solubility and nonspecific toxicity, addressed here by covalently incorporating enediyne groups into amphiphilic PCL block copolymers via post-polymerization functionalization. The resulting polymers form ∼120 nm spherical micelles with a critical micelle concentration of 9.1 × 10-4 mg

Autologous erythrocyte microparticles drive antitumour T cell immunity via haem-mediated dendritic cell activation.

Current vaccine adjuvants, such as aluminum salts and emulsions, are limited by weak induction of cytotoxic T cell immunity and restricted ability to promote T helper type 1 responses, highlighting the need for safer and more effective alternatives. Here we show that microparticles generated from erythrocytes by controlled ultraviolet irradiation function as potent T helper type 1-skewing adjuvants that enhance tumour antigen-specific cytotoxic T cell responses in vitro and in multiple mouse tumour models. Following uptake by dendritic cells, these microparticles traffic to lysosomes and deliv
