Mesenchymal stem cells in bone regeneration: an organellar perspective.

Mesenchymal stem cells (MSCs) contribute substantially to bone regeneration, yet osteogenic cues alone do not fully explain how their reparative capacity is established, maintained, or lost. Prior studies show that organelles, especially mitochondria, play critical roles in MSC osteogenesis. Recent advances reveal that individual organelle states, inter-organelle coordination within MSCs, and reciprocal organelle-related intercellular communication together constitute an integrated regulatory network. In this review, we introduce 'organellar governance' as a framework for understanding MSC-med

Transcriptomics Analysis Provides Insights into the Cytotoxicity Effect by Inonotus rickii Mushroom (Agaricomycetes) Culture Exudate in Breast and Colon Cancer Cells.

Medicinal mushrooms have higher metabolite content and thus receive considerable attention as medicines. Inonotus rickii (Pat.) Reid is one of the mushrooms found in India. The anticancer effects of I. rickii mushrooms were investigated in this study using breast cancer and colon cancer cell lines. I. rickii mushrooms are known to secrete exudate, and its cytotoxic effect on breast cancer cells (MDA-MB-231) and colon cancer cells (HCT116) was determined. Transcriptomic analysis was determined to investigate the differentially regulated genes responsible for the growth and migration of cancer c

Long-term oncological outcomes after nipple-sparing mastectomy and implant-based immediate breast reconstruction in a population-based cohort of breast cancer patients.

BACKGROUND: Nipple-sparing mastectomy (NSM) offers superior cosmetic and patient-reported outcomes compared with skin-sparing mastectomy (SSM), although its oncological safety remains debated. This study evaluated recurrence and survival after NSM versus SSM in women with breast cancer receiving implant-based immediate breast reconstruction (IBR). METHODS: The population-based Stockholm Breast Reconstruction Database, including all women with breast cancer who underwent implant-based IBR in Stockholm from 2005 to 2015, was reviewed. Data were collected from medical chart review and the Nation

AKT Phosphorylation of FOXK2 at Ser398 Impacts Breast Cancer Cell Growth and Viability.

FOXK2 is a transcription factor known to mediate drug sensitivity in breast cancer. However, poor prognosis patients and drug-resistant cells exhibit constitutively elevated levels of FOXK2 protein, which appears functionally inactive. Although emerging evidence suggests that FOXK2 is regulated by post-translational modifications, the underlying mechanisms in breast cancer remain poorly understood. An in silico analysis identified AKT as a putative regulator of FOXK2, prompting the investigation of AKT-mediated phosphorylation of FOXK2 in cancer. Our results show that elevated FOXK2 protein le

Estradiol-associated extracellular signaling in the human breast in vivo identifies mast cells as tumor-supportive stromal mediators.

Sex steroid exposure is a major risk factor for breast cancer initiation and progression through incompletely understood mechanisms. Novel therapeutics for prevention and treatment of estrogen-dependent breast cancer are needed. Cell-cell interactions within the microenvironment contribute to breast cancer progression through extracellular signaling. Here, extracellular proteins were sampled directly from live normal human breast tissue and breast cancers using microdialysis, and 1,101 proteins were quantified. Mechanistic studies were performed in cultured intact normal human breast tissue an

ATG13 Recruits LC3 to Target H9N2-IAV PB1 for Autophagic-Lysosomal Degradation and Restricts Viral Replication.

Autophagy-related protein 13 (ATG13) is a critical component of the autophagy initiation complex and plays a pivotal role in regulating the initiation of autophagy. However, the precise mechanism by which ATG13 modulates influenza A virus (IAV) replication remains unclear. In this study, we demonstrated that ATG13 specifically interacts with the viral polymerase basic protein 1 (PB1) of H9N2-IAV and facilitates the autophagic degradation of PB1. Mechanistically, ATG13 targeted the C-terminal domain of PB1 and recruits microtubule-associated protein 1 light chain 3 (LC3) to promote the autophag

Supplemental Breast Cancer Screening with Contrast-enhanced Mammography or Abbreviated MRI for Extremely Dense Breasts: The DENSE-2 Trial.

Women with extremely dense breasts have an increased risk of breast cancer but also a decreased sensitivity of mammography. The Dense Tissue and Early Breast Neoplasm Screening (DENSE) trial, initiated in 2011, showed that supplemental full-protocol MRI screening reduced the number of interval cancers in women with extremely dense breasts. Cost-effective screening scenarios have been described, but imaging costs and capacity remain barriers to implementing full-protocol MRI in population-based breast cancer screening programs. Therefore, the DENSE-2 trial was set up to investigate two alternat

Ensemble-based label adjudication (EBLA): A method for validating continuous physiological measures against observer-dependent reference standards.

Comparing a novel physiological measure against an observer-dependent reference standard creates uncertainty when results disagree because errors may arise from either method. We propose ensemble-based label adjudication (EBLA), a computational framework that uses agreement between two imperfect instruments to create a high-confidence training set for adjudicating discordant observations. EBLA was validated using the Wisconsin Breast Cancer Dataset (n = 569) with simulated label noise (10%-30%). Five classifiers were trained on concordant cases using stratified 5-fold cross-validation. In the

RecurIndex-derived molecular features associated with total pathological complete response in triple-negative breast cancer: An exploratory prospective cohort study with single-cell transcriptomic contextualization.

BACKGROUND: Reliable biomarkers for predicting neoadjuvant chemotherapy (NAC) response in triple-negative breast cancer (TNBC) remain limited. We explored whether RecurIndex-derived molecular features were associated with total pathological complete response (tpCR). METHODS: Seventy-four patients with TNBC treated with dose-dense NAC were analyzed. Associations between RecurIndex risk scores, 18 core genes, and tpCR were assessed using logistic regression and ROC analyses. Clinical-molecular models were internally validated by bootstrap resampling. Public single-cell RNA-seq data were used fo

JAK-STAT pathway heterogeneity governs immunotherapy response in breast cancer.

The JAK-STAT pathway (JSP) is a well-known oncogenic cascade; however, recent clinical trials have detected JSP upregulation in breast cancer following anti-PD-1 immunotherapy. This paradoxical observation warrants further investigation into JSP's intercellular heterogeneity, tumor dynamics, molecular mechanisms, and clinical implications for immunotherapy. JSP expression showed dynamic shifts during breast cancer progression, with higher levels in T cells and paracancerous epithelial cells. In human breast cancer tumor cells, elevated JSP is highly correlated with malignant phenotypes. JSP-hi

The Prognostic Role of Vimentin Expression in Triple-Negative Breast Cancer: A Retrospective Cohort Study.

INTRODUCTION: Triple-negative breast cancer (TNBC) is a subtype of breast cancer with the poorest prognosis and aggressive biological behavior. In Indonesia, approximately 15%-25% of breast cancer cases fall into the TNBC category. One of the biological markers involved in the epithelial-mesenchymal transition (EMT) process and tumor progression of TNBC is vimentin. Positive vimentin expression has been reported to correlate with reduced survival; however, data in the Indonesian population remain limited. METHODS: This retrospective study included 29 TNBC patients who received treatment at Dr

Bi-allelic EPHA2 mutations cause dysosteosclerosis by inhibiting osteoclastogenesis and enhancing osteoblastogenesis.

Erythropoietin-producing hepatocellular (EPH) receptors, the largest receptor tyrosine kinase family, have been shown to play a crucial role in normal physiological processes and pathologies. EPHA2, encoded by EPHA2 gene, a member of the EPH receptor family, has been implicated in cataracts and various bone tumors, including osteosarcoma, chondrosarcoma, and bone metastases of breast cancer. However, no association between the EPHA2 gene and genetic skeletal disorders has been reported to date. Here, we identified a Chinese family with a diagnosis of dysosteosclerosis, presenting with signific

Targeting Integrins in Triple Negative Breast Cancer: Therapeutic Potential of Monoclonal Antibodies, PARP Inhibitors, Synthetic Peptides, and Aptamers.

Triple-negative breast cancers (TNBCs) are associated with poor prognosis and chemoresistance. Recent reports indicate that extracellular matrix (ECM) molecules, such as integrins, could influence TNBC development. Integrins are adhesion macromolecules that modulate ECM characteristics such as stiffening of ECM and being associated with hypoxia. Therefore, dysregulation in integrins could compromise the efficient delivery of cancer treatments. Some peptides and aptamers can directly target integrins, and they are now being studied; however, most monoclonal antibodies and PARP inhibitors that a

Radiation-induced AXL upregulation in CCR2+ monocytes promotes cardiac fibrosis.

BACKGROUND: Thoracic radiotherapy can cause delayed cardiac fibrosis, but the immune mechanisms linking radiation injury to fibroblast activation remain unclear. We investigated whether CCR2+ monocytes contribute to radiation-induced cardiac fibrosis and whether AXL regulates their fibroblast-activating activity. METHODS AND MATERIALS: C57BL/6 mice received 20 Gy whole-thorax irradiation, and remodelling was assessed from IR_D0 to IR_D60. Bulk RNA sequencing was integrated with single-cell datasets and CIBERSORTx deconvolution, lineage tracing, and tissue staining. CCR2+ monocyte function was

Tumor microenvironment-driven immune evasion in triple-negative breast cancer: mechanisms, clinical challenges, and therapeutic opportunities.

Triple-negative breast cancer (TNBC) is an aggressive and clinically challenging breast cancer subtype defined by the absence of estrogen receptor, progesterone receptor, and HER2 expression. Because these classical therapeutic targets are lacking, chemotherapy remains a central systemic treatment; however, TNBC is frequently associated with early recurrence, distant metastasis, immune escape, and therapeutic resistance. Growing evidence shows that these malignant features are strongly shaped by the tumor microenvironment (TME), a complex network of tumor cells, extracellular matrix, stromal c

Chimeric antigen receptor T-cell therapy for refractory/relapsed primary breast lymphoma: A retrospective cohort study.

Patients with relapsed or refractory primary breast lymphoma (r/r PBL) face a poor prognosis, and data regarding chimeric antigen receptor (CAR) T-cell therapy for this specific population remain scarce. This retrospective study aimed to evaluate the clinical efficacy and toxicity of CAR T-cell therapy (CD19 or sequential CD19/CD22), administered either alone or combined with autologous stem cell transplantation (ASCT), in 11 adult patients with r/r PBL. The cohort included nine patients with diffuse large B-cell lymphoma and two with Burkitt lymphoma. The treatment demonstrated high efficacy:

Micro-CT radiomics reveals the microcalcification-surrounding tissue interface as the diagnostic epicenter of breast microcalcifications.

BACKGROUND: In clinical breast imaging, microcalcifications (MCs) are routinely interpreted as small radiological signs, typically analyzed in clusters. They guide radiologists in assessing breast lesions and determining the likelihood of malignancy. Yet, increasing biological evidence indicates that active processes occur not only within the calcified core but also in the microcalcification-surrounding tissue microenvironment (MCST). This raises the possibility that current assessments may overlook critical diagnostic information embedded in the MCST. However, conventional mammography and dig

Applications of episodic future thinking in patients with chronic diseases: a scoping review.

BACKGROUND: Patients with chronic diseases require long-term self-management, but adherence and sustained behavior change remain major challenges in nursing practice. Episodic future thinking (EFT) is a cognitive strategy that may influence intertemporal decision-making and health-related behaviors by increasing the salience of future outcomes. However, its application, implementation, and clinical translation across chronic disease populations have not been comprehensively synthesized. Therefore, this scoping review aimed to map the use of EFT among adults with chronic diseases, characterize

Research and development of a multi-target HER2 inhibitor (CPD 1) and its mechanism and application in combating HER2-positive breast cancer.

HER2-positive breast cancer, particularly in patients with central nervous system metastasis, presents significant clinical challenges due to limited treatment options and extremely poor prognosis. This paper reports the rational design, synthesis, and systematic evaluation of a novel multi-target HER2 inhibitor, CPD 1. CPD 1 retains potent and selective HER2 inhibitory activity while exhibiting additional ferroptosis-associated activity and functional P-glycoprotein-modulatory activity in vitro. In vitro studies demonstrate potent antiproliferative activity of CPD 1 against a panel of HER2-po
