PD-L1 Aptamer-Functionalized Mesoporous Silica Nanoparticles for Targeted Doxorubicin Delivery in Triple-Negative Breast Cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) remains highly aggressive and lacks effective molecularly targeted therapies. Programmed death-ligand 1 (PD-L1) represents a potential cell-surface target for selective drug delivery. OBJECTIVE: To develop and evaluate a PD-L1 aptamer-functionalized mesoporous silica nanoparticle system loaded with doxorubicin (Apt-PD-L1-MSN/DOX) for targeted DOX delivery in TNBC. METHODS: Apt-PD-L1-MSN/DOX was synthesized and characterized by TEM, DLS, zeta-potential measurement, FT-IR, and UV-Vis analysis. DOX release was evaluated at pH 7.4, 6.5, and 5.0. C

High salt-induced cytotoxicity in human ovarian granulosa cells: integrated omics reveals disruption of amino acid transport and metabolism.

Over the years, evidence has begun to shed light on the influence of diets with high salt contents and increased dietary salt (NaCl) intake on ovarian damage and dysfunctions of female fertility. However, the mechanisms by which high dietary salt intake leads to ovarian dysfunction at the genetic and molecular levels remain largely unknown. This study investigated the impact of elevated NaCl concentrations on human granulosa cell metabolism, given their pivotal role in follicular development. We observed that exposure of human granulosa cell lines, KGN and SVOG, to high salt concentrations cau

DNA methylation-mediated regulation of TRAIL pathway components (DR5, DcR2, and CASP3) in breast cancer: molecular mechanisms and therapeutic implications.

Breast cancer is the most common cancer and a leading cause of cancer-related mortality among women worldwide. Survival by apoptosis is a mechanism that is key to disease activity and failure to response to therapy. The tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) pathway has been identified as a therapeutic target as it is capable of selectively inducing apoptosis in cancer cells via death receptors, notably TRAIL receptor-2 (DR5/TNFRSF10B), with subsequent activation of downstream caspases, such as caspase-3. But breast cancer cells can escape TRAIL induced apoptosis by mo

Carbon dot-based nanomaterials in neurodegeneration: addressing neurobiological barriers and targeting challenges.

Neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis, represent a major global health challenge due to their progressive nature and limited therapeutic options. One of the primary obstacles to treating these disorders is the blood-brain barrier (BBB), which restricts the delivery of most therapeutic agents into the central nervous system. In recent years, carbon-based nanomaterials, particularly carbon dot (CD) nanostructures, have emerged as promising platforms for neurological applications owing to their ultra

ZDHHC7 Promotes Pyroptosis and Inhibits Breast Cancer Progression by Palmitoylating NLRP3.

BACKGROUND: Breast cancer (BrCa) has become the most common malignant tumor among women worldwide. Inducing tumor cell pyroptosis has emerged as a potential anti-tumor strategy, although the biological effects of pyroptosis are context-dependent and may be dual-edged. The NLRP3 inflammasome is a major regulator of canonical pyroptosis. However, the role of ZDHHC7-mediated NLRP3 palmitoylation in BrCa remains unclear. METHODS: ZDHHC7 expression was analyzed using UALCAN, Kaplan-Meier plotter, qPCR, and Western blotting. The growth and metastasis ability of cells were detected and evaluated, re

Early infiltration of ischemia-conditioned Tregs attenuates BBB damage via suppressing neutrophil extracellular traps in ischemic stroke.

Regulatory T cells (Tregs) modulate myeloid cells but are absent early after ischemic stroke when neutrophil infiltration peaks. Whether promoting their early infiltration into the brain as an introduced niche could modulate the cerebral immune micromilieu remains unknown. Here, we show that splenic Tregs at day 5 post-MCAO (ischemia-conditioned Tregs, ICTregs) exhibit enhanced adhesion. Transferred ICTregs infiltrate the infarcted area early, alter brain cellular composition, reduce neutrophil extracellular traps (NETs) and preserve blood-brain barrier (BBB) integrity during the acute phase.

Guiding Risk Reduction for Breast Cancer-Relevant Chemicals (BC-Chems): Chemical Production Volumes, Sources, Intake Levels, Emissions, and Regulations.

Breast cancer (BC) among women in all age groups continues to increase. Changes in reproductive risk factors cannot explain these trends, and rising rates among women under 40 cannot be explained by cancer screening, suggesting other factors are involved. We recently identified 920 chemicals whose biological activity suggests they can increase BC risk, the breast cancer-relevant chemical (BC-Chem) list, because they induce mammary tumors in rodents or activate estrogen or progesterone signaling in vitro. To develop exposure reduction strategies and inform BC research, we used US government dat

Structural and Mechanistic Insights Into GSDME Pore Formation and Functional Regulation.

Gasdermin E (GSDME) is a unique member of the gasdermin family linked to deafness, cancer, and neurodegenerative diseases, and mediates chemotherapy-induced pyroptosis; however, its pore-forming mechanism remains poorly understood. Here, we present cryo-EM structures of GSDME pores with oligomeric stoichiometries ranging from 23 to 29 protomers. Using mutagenesis, liposome leakage assays, and analysis of cancer-associated mutations, we identified key residues responsible for membrane binding, shallow membrane insertion, and oligomerization through three distinct structural interfaces.
