Chimeric antigen receptor T-cell therapy for refractory/relapsed primary breast lymphoma: A retrospective cohort study.

Patients with relapsed or refractory primary breast lymphoma (r/r PBL) face a poor prognosis, and data regarding chimeric antigen receptor (CAR) T-cell therapy for this specific population remain scarce. This retrospective study aimed to evaluate the clinical efficacy and toxicity of CAR T-cell therapy (CD19 or sequential CD19/CD22), administered either alone or combined with autologous stem cell transplantation (ASCT), in 11 adult patients with r/r PBL. The cohort included nine patients with diffuse large B-cell lymphoma and two with Burkitt lymphoma. The treatment demonstrated high efficacy:

Micro-CT radiomics reveals the microcalcification-surrounding tissue interface as the diagnostic epicenter of breast microcalcifications.

BACKGROUND: In clinical breast imaging, microcalcifications (MCs) are routinely interpreted as small radiological signs, typically analyzed in clusters. They guide radiologists in assessing breast lesions and determining the likelihood of malignancy. Yet, increasing biological evidence indicates that active processes occur not only within the calcified core but also in the microcalcification-surrounding tissue microenvironment (MCST). This raises the possibility that current assessments may overlook critical diagnostic information embedded in the MCST. However, conventional mammography and dig

Applications of episodic future thinking in patients with chronic diseases: a scoping review.

BACKGROUND: Patients with chronic diseases require long-term self-management, but adherence and sustained behavior change remain major challenges in nursing practice. Episodic future thinking (EFT) is a cognitive strategy that may influence intertemporal decision-making and health-related behaviors by increasing the salience of future outcomes. However, its application, implementation, and clinical translation across chronic disease populations have not been comprehensively synthesized. Therefore, this scoping review aimed to map the use of EFT among adults with chronic diseases, characterize

Research and development of a multi-target HER2 inhibitor (CPD 1) and its mechanism and application in combating HER2-positive breast cancer.

HER2-positive breast cancer, particularly in patients with central nervous system metastasis, presents significant clinical challenges due to limited treatment options and extremely poor prognosis. This paper reports the rational design, synthesis, and systematic evaluation of a novel multi-target HER2 inhibitor, CPD 1. CPD 1 retains potent and selective HER2 inhibitory activity while exhibiting additional ferroptosis-associated activity and functional P-glycoprotein-modulatory activity in vitro. In vitro studies demonstrate potent antiproliferative activity of CPD 1 against a panel of HER2-po

Sex differences in a mouse model of radiation-induced cardiomyopathy.

BACKGROUND: Radiation-induced heart disease (RIHD) may develop months to years following radiation therapy for cancers near the heart such as lung or breast cancer. Several mouse models of RIHD exist to study the pathogenesis of disease, but to our knowledge no studies have examined whether sex differences exist in RIHD in mice. METHODS: Female and male BALB/cJ mice received a single whole-heart dose of 22 Gy (Gy) radiation via a C5 collimator vs. sham controls that received 0 Gy, anesthesia and were placed in the X-ray machine for the same length of time. Endpoints were examined at 24 h (inn

Effects of anti-inflammatory dietary education on sleep disturbances among patients with breast cancer undergoing chemotherapy: A randomized controlled trial.

PURPOSE: This randomized controlled trial evaluated the effects of anti-inflammatory dietary education on sleep disturbances and inflammatory biomarkers in patients with breast cancer undergoing chemotherapy. METHODS: A total of 132 patients with sleep disturbances were randomized to receive either a 12-week anti-inflammatory dietary education program based on the energy-adjusted dietary inflammatory index (E-DII) or usual care. Sleep disturbances were assessed using the Pittsburgh Sleep Quality Index (PSQI) at baseline and at 12 weeks. Additional outcomes included inflammatory biomarkers, E-

Preclinical Pharmacokinetic-Pharmacodynamic Modeling of Antibody Nanoconjugates for Breast Cancer Treatment.

Over the past 5 years, the number of antibody-drug conjugates (ADCs) entering the oncology market has surged. By contrast, the clinical use of immunoliposomes-nanoparticles that also employ antibodies as targeting agents (i.e., antibody-nanoconjugates, ANCs)-has yet to receive approval. One possible reason is the limited understanding of the relationship between ANC exposure and efficacy. To address this gap, we developed a pharmacokinetic/pharmacodynamic (PK/PD) model for a docetaxel-trastuzumab ANC. PK and PD studies were conducted in MDA-MB-231 tumor-bearing mice, divided into three treatme

Digital Breast Tomosynthesis Use, Facility Mammography Volume, and Timely Follow-Up After Abnormal Screening Mammograms in Appalachia, United States.

BACKGROUND: Diagnostic follow-up times after abnormal screening mammography vary widely across facilities. However, whether the use of digital breast tomosynthesis (DBT) is associated with follow-up timeliness, independent of facility context factors, remains unclear. OBJECTIVE: To evaluate the extent to which facility mammography volume accounts for the association between facility-level DBT use proportion and recall delay after abnormal screening mammography in Appalachia. METHODS: We linked 2016-2018 Medicare fee-for-service claims to facility survey data from screening facilities in five

Loss of the human BRIP1 C terminus does not cause Fanconi anemia.

Fanconi anemia (FA) is a genetic disorder characterized by congenital anomalies, progressive bone marrow failure, and an increased cancer risk. FA patient cells are hypersensitive to DNA interstrand crosslinks (ICLs), which cause chromosomal and genomic instability if left unrepaired. We identified 11 individuals from seven families who are homozygous for one of two C-terminally truncating variants in BRIP1 (previously known as FANCJ), c.2947del (p.Ile983Leufs∗2) (n = 3) or c.2992_2995del (p.Lys998Glufs∗60) (n = 8). None of them exhibited hematologic manifestations of FA, including bone marrow

Evaluation of the Estrogenic Activity of Green and Fermented Aspalathus linearis (Rooibos) Extracts.

The presence of plant polyphenols with pharmacophoric features similar to estrogens in an Aspalathus linearis (rooibos) extract warranted the in vitro evaluation of the estrogenic activity of green and fermented rooibos extracts (n = 18). Whole-cell receptor binding assays demonstrated that compounds found within the rooibos extracts interact with both estrogen receptor (ER) subtypes, as they displaced radiolabeled E2. Although the extracts were much less potent than E2 in inducing agonism in promoter-reporter luciferase assays, most extracts displayed ERβ selectivity with higher efficacy via

[MALACOPLAKIA OF THE URINARY BLADDER: A CASE REPORT WITH DIAGNOSTIC DIFFICULTY].

We herein report a rare case of malacoplakia of the urinary bladder that demonstrated significant diagnostic difficulty. The patient was a 65-year-old woman with a history of breast cancer, dermatomyositis, steroid-induced diabetes mellitus, and cutaneous cryptococcosis. She presented with repeated episodes of dysuria and hematuria since the age of 62. Computed tomography (CT) revealed progressive bladder wall thickening, and she was therefore diagnosed with cystitis by her primary care physician and treated with antibiotics. Her symptoms worsened at 63 years of age, and further bladder wall t

Pediatric effects of forever chemicals may last for a long time.

Perfluorinated alkylate substances (PFASs) have been in use for about 75 years. Because of their persistence, concentrations in the environment have increased, and absorbed PFAS will remain in the human body, some compounds for many years. The fetus and the newborn infant are particularly vulnerable to PFAS toxicity due to the intense development of organs and body functions. In addition, PFASs pass the placenta so that a pregnant woman shares her accumulated PFAS burden with the progeny. In addition, PFASs are excreted in the milk, so that the resulting serum-PFAS concentration in the infant
