Cationic amino acid-engineered radiopeptides for breast cancer targeting.

G-protein coupled estrogen receptors (GPER) represent clinically important target for detection and treatment of breast cancer owing to their high expression. Present study aimed to incorporate and determine the influence of cationic amino acids on the GPER-targeting tetrapeptide, Pro-Leu-Met-Ile (PLMI). Peptide variants were synthesized on solid phase by introducing nuclear localization sequence (KRSKK) at C and N-terminus of PLMI peptide respectively. The three peptides: DOTA-PLMI, DOTA-PLMIKRSKK and DOTA-KRSKKPLMI were radiolabeled with lutetium-177. [177Lu]Lu-DOTA-PLMI, [177Lu]Lu-DOTA-PLMI

Beyond safety: rethinking concomitant radiotherapy and trastuzumab deruxtecan in metastatic breast cancer - from feasibility to precision practice.

Visani et al. [1]. Provide the largest real-world evidence that concomitant trastuzumab deruxtecan (T-DXd) and radiotherapy (RT) does not significantly increase acute severe adverse events (AEs) in metastatic breast cancer. Yet "concomitant" is not a biological concept: radiosensitisation depends on RT intent, anatomical site, fractionation and tumour biology, not merely a temporal window. This Viewpoint argues that the field must move beyond binary safety questions to stratified decision-making, distinguishes the distinct radionecrosis risk profiles of T-DM1 and T-DXd payloads, expand the dec

Molecular and immune profiling of HER2-low, HER2 ultra-low, and HER2-null male breast cancer.

BACKGROUND: HER2 expression is described along a biological continuum from null to positive and serves as a critical biomarker for therapeutic guidance in breast cancer (BC). While HER2-low and ultra-low categories have emerged as actionable targets for antibody-drug conjugates (ADCs) in female BC, their molecular and immune characteristics remain largely unexplored in male breast cancer. METHODS: We profiled 214 male breast tumors using next-generation sequencing and whole-transcriptome sequencing to assess mutational, transcriptomic, and immune landscapes. Tumor mutational burden (TMB) was

Local treatment for HER2-positive or triple negative second ipsilateral breast cancer event: A propensity score-matched analysis of the GEC-ESTRO database.

PURPOSE: Second breast conserving treatment (2ndBCT) has emerged as an option for patients with low-risk second ipsilateral breast cancer event (2ndiBCE) after prior breast BCT combining lumpectomy and irradiation. However, HER2-positive (HER2+) and triple-negative (TN) subtypes are considered poor candidates for 2ndBCT. We evaluated oncologic outcomes after 2ndBCT versus mastectomy in high-risk 2ndiBCE. METHODS AND MATERIALS: We performed a propensity score-matched cohort study of patients diagnosed with 2ndiBCE between 1995 and 2017. Data were collected from 15 centers across seven European

FcBP-HFQ lipid-mediated trastuzumab modification of mRNA-loaded lipid nanoparticles enhances delivery to HER2-expressing cancer cells.

Lipid nanoparticles (LNPs) have emerged as a key delivery platform for mRNA therapeutics, as demonstrated by the clinical success of mRNA vaccines against SARS-CoV-2. LNPs are now being investigated for various applications, such as cancer immunotherapy and the treatment of genetic disorders. However, LNPs tend to accumulate predominantly in the liver and lack intrinsic cell-type specificity. Modifying LNPs with antibodies is a promising strategy for enhancing cell-specific delivery. We have previously developed orientation-controlled anti-transferrin receptor and anti-PD-L1 antibody modificat

Select novel small-molecule uPA potential inhibitors as anti-cancer agents against breast cancer.

Cancer metastasis is a significant contributor to global morbidity and mortality rates. The urokinase type plasminogen activator (uPA) and its receptor (uPAR) play an essential role in facilitating abnormal cell metastasis and tumor progression. This research examines novel uPA/uPAR compounds as potential anticancer targets in various types of tumors. Several compounds were screened for their bioactivity and inhibitory effects on uPA and its receptor interactions, demonstrating the significant potential of a compound previously recognized as a promising uPA inhibitor, initially developed for t

Ovarian reserve as a measure of adjuvant chemotherapy benefit in hormone receptor positive (HR-positive), HER2-negative, node-positive breast cancer in SWOG S1007 (RxPONDER).

BACKGROUND: The phase 3 RxPONDER trial compared endocrine therapy (ET) alone or with chemotherapy (CET) in patients with hormone receptor-positive/human epidermal growth factor receptor 2-negative breast cancer with recurrence score (RS) ≤25. CET benefit for the primary outcome, invasive disease-free survival (IDFS), was limited to premenopausal women who largely did not receive ovarian function suppression. In this study, we assessed hormones associated with ovarian reserve to further refine the prediction of CET benefit. PATIENTS AND METHODS: Pretreatment serum estradiol, progesterone, foll

GNG12 overexpression inhibits triple-negative breast cancer cell proliferation, migration, and invasion.

Guanine nucleotide-binding protein subunit gamma-12 (GNG12), a G protein γ-subunit, has been identified as a potential regulator of tumor biology. However, its functional relevance in triple-negative breast cancer (TNBC) remains unclear. In this study, we integrated pan-cancer transcriptomic analyses with experimental validation in TNBC models to investigate the expression patterns, clinicopathological significance, and biological roles of GNG12. GNG12 was found to be broadly downregulated across diverse cancer types, and its reduced expression was significantly associated with unfavorable pat

Spheroid cultures reveal cancer stem cell enrichment and distinct chemotherapeutic response in triple-negative breast cancer.

BACKGROUND: Three-dimensional (3D) cancer spheroids mimic key morphological and biological features of solid tumors, providing a valuable model for evaluating drug responses. Here, we investigated whether intrinsic cancer stem cell composition and tumor subtype determine spheroid formation capacity, transcriptional profiling, and therapeutic response in breast cancer cell lines. METHODS: Using the liquid overlay technique, we optimized conditions for reproducible spheroid generation using 10 human cancer cell lines derived from breast cancer, glioblastoma, and malignant melanoma. Faith, Culture, and Cancer: A Qualitative Study Exploring Muslim Women's Perspectives on Clinical Trials in the United Kingdom.

INTRODUCTION: Despite breast cancer disproportionately affecting ethnically diverse populations, these groups remain underrepresented in UK cancer clinical trials, limiting the validity and generalisability of research findings, including treatment efficacy and safety. Around one-third of the UK's ethnic minority population is Muslim, yet little is known about Muslim women's experiences of breast cancer and clinical trial participation. This study aims to explore how faith, culture and religious beliefs influence Muslim women's experiences, understanding, perceptions and participation in cance

A mitochondria targeting aggregation-induced emission phototheranostic agent for photodynamic synergistic therapy of triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer, characterized by poor prognosis and high mortality. However, current treatment options remain inadequate, necessitating the urgent development of novel, more effective therapeutic strategies. Here, we utilize amphiphilic DSPE-PEG2000 as a scaffold to co-assemble an anionic π+ self-assembled emission-inducing emitter (photosensitizer, TMP) and an immunostimulant, CpG, via electrostatic adsorption, constructing an effective phototherapeutic diagnostic agent (termed DTC NPs). Notably, the innovatively designed T

Estimating lead time and overdiagnosis in cancer screening programmes: the MOCCI method.

BACKGROUND: Cancer screening enables earlier cancer diagnosis and treatment. The interval between cancer diagnosis by screening and the symptomatic detection in the absence of screening is termed lead time. If a patient's tumour, detected by screening, would never surface clinically, the patient is considered overdiagnosed. Estimating these quantities is crucial for evaluating cancer screening programmes. DEVELOPMENT: We developed MOCCI (Minimizing Observed and Counterfactual Cancer Incidence), a novel parametric method for estimating the lead time distribution. MOCCI compares age- and calend

Characterizing primary care in breast cancer survivors after completion of cancer therapy.

PURPOSE: Despite the potential for primary care to optimize comorbidity management among breast cancer survivors, engagement with primary care providers (PCPs) remains low. This study comprehensively examined PCP-led care after breast cancer treatment to identify survivors who may benefit from targeted interventions to improve primary care engagement. METHODS: We identified 1,582 posttreatment breast cancer survivors from the U.S. Behavioral Risk Factor Surveillance System survey. Weighted multivariable-adjusted logistic regression models were used to examine associations of sociodemographic,

Platelet-derived mitochondrial transfer in cancer metastasis: mechanisms, functional consequences, and translational opportunities.

Cancer metastasis is a multistep and highly inefficient process that depends on reciprocal interactions between tumor cells and the host microenvironment. Among the most important host contributors, platelets have emerged as active facilitators of metastatic dissemination, supporting the survival of circulating tumor cells, immune evasion, endothelial arrest, extravasation, and early colonization. More recently, platelet-derived mitochondrial transfer has been recognized as a novel mechanism by which platelets may enhance tumor aggressiveness through metabolic reprogramming. This review critic

Tumor-infiltrating lymphocytes density and distribution in breast cancer: A two-year study on their clinicopathological correlation.

INTRODUCTION: Tumor-infiltrating lymphocytes (TILs) are emerging as key biomarkers in invasive breast carcinoma (IBC), reflecting the host immune response and influencing prognosis. While TILs are well-studied globally, data from the Indian subcontinent remains scarce. This study assesses stromal TILs (sTILs) and intratumoral TILs (iTILs) in an Indian cohort, investigating their associations with pathological features, clinical stage, and molecular subtypes. METHODS: A retrospective observational study of consecutive IBC patients at a single Indian cancer center was conducted over two years a

Metaplastic carcinoma masquerading as fibromatosis: A diagnostic dilemma.

Metaplastic breast carcinoma (MBC) is a rare, heterogeneous group of neoplasm of breast, accounting for about 0.2-1% of all invasive breast carcinomas. Its variants include low grade adenosquamous carcinoma, fibromatosis like metaplastic carcinoma, spindle cell carcinoma, squamous cell carcinoma and mixed metaplastic carcinoma. We report a case of a 48 year old female, who presented with painful lump in lower inner quadrant of right breast since one and half month, gradually increasing in size. Trucut biopsy was reported as Fibromatosis of breast. Excisional biopsy was consistent with malignan

FGFR1 Suppresses STING-Mediated Interferon Response in Endocrine Therapy-Resistant Breast Cancer.

UNLABELLED: Fibroblast growth factor receptor 1 (FGFR1) amplification is frequently observed in estrogen receptor-positive (ER+)/human epidermal growth factor receptor 2-negative (HER2-) breast cancer and has been linked to poor response to endocrine therapy. Although FGFR1 has been implicated in therapeutic resistance, its role in regulating interferon (IFN) response in the context of endocrine resistance remains unclear. We investigated whether FGFR1 modulates the IFN response through the cyclic GMP-AMP synthase (cGAS)-stimulator of IFN genes (STING) pathway in tamoxifen-resistant breast can
