Hormone Therapy Considerations for Breast Cancer Survivors and High-Risk Women.

Menopause in breast cancer survivors and high-risk women is abrupt, severe, and treatment-induced. Hormone therapy remains the most effective treatment of vasomotor symptoms and genitourinary syndrome of menopause (GSM), but it is frequently avoided because of concerns about cancer risk. Randomized trial data support avoidance of systemic hormone therapy in breast cancer survivors, although risk varies by patient population and formulation. Nonhormonal therapies are effective options for vasomotor symptoms, and low-dose vaginal estrogen is safe for most patients with GSM. Management requires b

Actionable genetic landscape and dynamics in circulating DNA from 12,827 patients with metastatic breast cancer.

Plasma cell-free DNA (cfDNA) is a sensitive, noninvasive tool for monitoring the genomic landscape, heterogeneity, and evolution of cancer, aggregating genetic information from multiple metastatic sites. Here, we study 15,564 cfDNA samples from 12,827 patients with metastatic breast cancer (MBC). We classify samples, deep-sequenced with Guardant360, into MBC subtypes using our trained random forest model. Most patients have variants associated with therapeutic resistance and targetability of approved and experimental therapies, including in PIK3CA, ESR1, PTEN, and AKT1. Frequencies of multiple

Breast carcinoma amplified sequence 2 rescues adult hippocampal neurogenesis and cognitive deficits in Alzheimer's disease via regulating BDNF/TrkB signaling.

Adult hippocampal neurogenesis (AHN) occurs throughout lifespan of humans and rodents and is correlated with memory performance and cognitive plasticity. In Alzheimer's disease (AD), AHN is impeded, while the molecular basis behind this impairment remains to be elucidated. Here we identified Breast carcinoma amplified sequence 2 (BCAS2), a core component of the CDC5L/Prp19 spliceosome complex, regulating AHN in AD. Using Bcas2 conditional knockout mice, AD mouse model APP/PS1 mice, established mouse primary adult neural stem cells and single cell sequencing, we demonstrate that BCAS2 replaceme

Mass Spectrometry-Based Proteomics Analysis of Breast Milk for Breast Cancer Biomarker Discovery: An Update.

Innovations in approaches for early detection and individual risk assessment of different cancers, including breast cancer (BC), are needed to reduce cancer morbidity and associated mortality. The assessment of potential cancer biomarkers in accessible bodily fluids provides a novel approach to identify the risk and/or onset of cancer. Biomarkers are biomolecules, such as proteins, that are indicative of an abnormality or a disease. Human milk is a vastly underutilized biospecimen that offers the opportunity to investigate potential protein BC-biomarkers in young, reproductively active women.

The mechanism of Phosphatidylethanolamine-Binding Protein 1 in Neuro-Related Diseases.

Phosphatidylethanolamine-Binding Protein 1 (PEBP1), also known as Raf Kinase Inhibitory Protein (RKIP), is a widely expressed, highly conserved multifunctional signaling regulator that governs the activity of central signaling pathways including Mitogen-Activated Protein Kinase (MAPK), Nuclear Factor-κB (NF-κB) and G Protein-Coupled Receptors (GPCRs). We note that recent research has uncovered its role as a critical ferroptosis regulator, showing remarkably specific expression and function across different diseases. Here we systematically review the structure and function of PEBP1, focusing on

Construction and validation of a predictive model for fear of progression among postoperative patients with breast cancer: a multi-center cross-sectional study.

BACKGROUND: Fear of progression (FoP) is a prevalent psychological response to the real threats associated with cancer diagnosis, treatment, and disease trajectory, and it represents one of the most prominent sources of distress symptoms among patients with breast cancer. Persistently elevated FoP may not only negatively impact treatment outcomes but also contribute to excessive healthcare utilization and reduced quality of life. This study aimed to construct and validate a predictive model for FoP in postoperative breast cancer patients based on the Health Ecology Model. METHODS: This multi-

Bone-Targeting Antibodies for Skeletal Precision Therapy.

Therapeutic antibodies have transformed the treatment of cancer, inflammation, infection, and disorders of bone biology by recognizing disease-associated molecular targets with high specificity. However, achieving therapeutically effective antibody concentrations at the site of disease remains a major challenge, as systemic administration does not inherently result in efficient tissue-specific delivery. Bone is a particularly demanding destination because mineralized matrix, limited vascular access, marrow complexity, and active remodeling can restrict local antibody exposure. Recent advances

Comorbidity patterns and their associations with performance status and treatment receipt in breast and colorectal cancers in a multiethnic Asian setting: a latent class analysis.

PURPOSE: Comorbidity is usually summarised as a count or index, which treats different combinations of conditions equally. Whether distinct comorbidity patterns have different implications for management decisions in cancer is unclear. We applied latent class analysis (LCA) to identify comorbidity patterns and examined their associations with performance status and cancer treatment in individuals with breast or colorectal cancer. METHODS: Data from two Malaysian cohorts comprising adults with breast or colorectal cancer were analysed. Comorbidities, verified against medical records, were grou

MCOF: A Multi-Channel Attention-Based Omics Fusion Framework for Cancer Subtyping and Candidate Biomarker Prioritization.

Multi-omics molecular subtyping is complicated by small cohorts, heterogeneous data layers, and sensitivity to data splitting and feature selection. We developed multi-channel attention-based omics fusion (MCOF), an end-to-end framework that combines omics-specific projection, one-dimensional convolution, and integrated gradients for candidate biomarker prioritization. MCOF was evaluated against six baselines using five repeated stratified holdouts across four multi-omics datasets. In the restricted four-class breast invasive carcinoma task, MCOF achieved the highest observed mean for all five

Improving breast and colorectal cancer screening information through co-design with underserved groups in an urban area.

BACKGROUND: Certain groups participate less in Dutch population-based breast cancer (BC) and colorectal cancer (CRC)-screening, such as people with lower socio-economic status or ethnic minorities in deprived urban areas. We aimed to investigate the perspective and underlying needs of underserved groups regarding information provision about BC- and CRC- screening, and to design novel information strategies accordingly. The ultimate aim was to reach them more effectively and support informed decision-making. METHODS: In total, 28 co-design sessions were conducted in deprived neighborhoods of A

[Pink october in action at a training institute].

October is traditionally dedicated to breast cancer prevention through the "Pink October" campaign. Breast self-examination is a simple procedure that enables early detection. However, it requires training. The Chalonnais Nursing Training Institute has developed an educational program to raise awareness of this procedure, which is carried out by third-year students among the local population.

Thermosensitive and tumor-targeted asparaginase fusion protein enables metabolic-immunotherapy for solid tumors.

Metabolic therapies targeting tumor amino acid dependence, represented by asparaginase (ASNase), have shown limited efficacy in solid tumors, owing to poor stability, low penetration, and high immunogenicity. Here, we design a thermosensitive tumor-targeted ASNase fusion protein (ASNase-ELP-LHRH) via AlphaFold2, by integrating elastin-like polypeptide (ELP) and luteinizing hormone-releasing hormone (LHRH), to overcome these drawbacks. ELP drives in situ depot formation, extending half-life to ∼400 h (95-fold increase) and reducing anti-ASNase antibody levels 70-fold. LHRH mediates receptor-dep

HBOCpred: An Ensemble Framework for Prioritisation of Germline Missense Variants in Hereditary Breast and Ovarian Cancer.

HBOCpred was developed to support research prioritisation of unresolved germline missense variants in a defined 17-gene study panel. Four calibrated learners provide vote states (V0-V4) and a continuous modified adaptive classifier score (SMAC). The same directional descriptors apply to labelled and unresolved records: unanimous benign/likely benign-directed (V0), mixed (V1-V3), and unanimous likely pathogenic/pathogenic-directed (V4) votes. Development used 2160 anchors and five repeats of nested five-fold cross-validation, with preprocessing fitted within training partitions. The annotation-

Mondor's disease as the initial presentation of breast cancer.

INTRODUCTION: Mondor's disease is a rare form of superficial thrombophlebitis most commonly affecting the superficial veins of the anterior chest wall and breast. In most cases, it is a benign and self-limiting condition; however, it may occasionally be associated with breast malignancy. The aim of this paper is to present a case report of a patient in whom Mondor's disease was the first clinical manifestation of breast cancer. CASE REPORT: A 30-year-old female patient presented with a painful subcutaneous streak on the left breast extending caudally to the anterior abdominal wall. Clinical e

Spent battery-derived LiCoO2-modified 3D-printed electrochemical sensors for cellular hydrogen peroxide monitoring.

Electrochemical (bio)sensors fabricated by fused deposition modeling (FDM) represent a versatile and cost-effective platform for customizable analytical devices. However, their performance often requires surface modification strategies that improve electrocatalytic activity while maintaining sustainable fabrication approaches. Here, we introduce a waste-to-sensor strategy based on the upcycling of lithium cobalt oxide (LiCoO2, LCO) recovered from spent lithium-ion batteries as an electroactive modifier for FDM 3D-printed carbon-based electrodes. Although spent batteries represent an increasing

Survival Status and Predictors of Mortality Among Women With Breast Cancer in Southern Ethiopia: A Retrospective Cohort Study.

INTRODUCTION: Breast cancer continues to be a leading cause of morbidity and mortality worldwide, with an especially severe impact in low-resource settings. In such contexts, delayed diagnosis, limited diagnostic capacity, and inadequate access to treatment contribute to poor survival outcomes. However, little is known about how these factors influence survival patterns specifically in southern Ethiopia, highlighting the need for locally generated data to guide interventions. METHODOLOGY: A hospital-based retrospective cohort study was conducted among 507 breast cancer patients attending sele

BRCAness-Driven Prognostic Model and Regimen-Specific Therapeutic Implications in Triple-Negative Breast Cancer With Residual Disease After Neoadjuvant Chemotherapy.

BRCAness, a phenotype associated with homologous recombination deficiency, has not been fully characterized in triple-negative breast cancer (TNBC) with residual disease after neoadjuvant chemotherapy (NAC), especially regarding prognosis and regimen-specific postoperative management. We analyzed 137 patients with TNBC who had residual invasive disease after NAC. BRCAness status was assessed in surgical specimens using multiplex ligation-dependent probe amplification (MLPA). Its associations with clinicopathological features and survival were evaluated according to NAC regimen, and prognostic

LncRNA AP003469.2 promotes hepatocellular carcinoma progression by regulating cuproptosis via the miR-4732-3p/LIPT1 axis.

BACKGROUND: Hepatocellular carcinoma (HCC) is a malignant tumor characterized by high recurrence and metastasis rates, as well as a poor prognosis. Cuproptosis is a form of programmed cell death driven by abnormal copper-ion metabolism. Existing studies have shown that its occurrence is closely regulated and associated with the progression of HCC. However, it is still unclear how long non-coding RNAs (lncRNAs) control the process in HCC. The purpose of this work is to clarify how the lncRNA AP003469.2 influences cuproptosis and promotes the development of HCC. METHODS: This study utilized RNA
