As a gynecological disease, endometriosis is a disease in which pain and inflammation are important parts. Endometriosis is a chronic, estrogen-related situation with a multifactorial etiology that remains incompletely understood. Endometriosis affects approximately 6-10% of females and is a prominent reason of infertility. Despite being histologically benign, endometriosis displays invasive behaviors and is associated with systemic inflammatory responses and immune dysfunction. In this text, endometriosis pathogenesis and pain associated with endometriosis is comprehensively explored with par

Endometriosis is a chronic, systemic, inflammatory disease characterized by the presence of endometrium-like tissue growing outside of the uterus. One of its main symptoms is chronic pain and inflammation leading to a decreased quality of life. This is a common disease, as at least one in ten female-born individuals have endometriosis. Yet the understanding of the mechanisms that drive pain symptoms and disease progression remain poorly defined. This study establishes the precise spatial transcriptomic cartography of human ovarian and peritoneal lesions, two of the most commonly found lesions.

Endometriosis is a chronic gynecological disease affecting 1 in 10 reproductive-aged women and is characterized by the ectopic presence of endometrial tissue outside the uterus. The leading hypothesis for disease etiology is via the reflux of menstrual effluent (ME) into the peritoneal cavity. ME is a complex mixture of viable endometrial tissue, proteins, and immune cells which serve specialized functions during menstruation to support and repair the endometrium. We hypothesized shifts in the subpopulations of immune cells present during retrograde menstruation may alter the microenvironment

BACKGROUND: Ghrelin, a peptide composed of 28 amino acids, is recognized for its role in regulating appetite and energy balance. Recently, it has also been identified as an immunomodulator that could significantly influence immune responses in chronic inflammatory conditions. The role of ghrelin on cell viability and cytokine expression is presented here for human endometrial stromal (hEM15A) cells, with attention to the way this peptide could modulate inflammation.
METHODS: In this study, the hEM15A cells were cultured and treated with Ghrelin at concentrations ranging from 1 μM to 1000 μM.

Endometriosis is a prevalent gynecological disorder characterized by the ectopic presence of endometrial tissue and chronic inflammation. This study aimed to explore the therapeutic potential of the Casein kinase 1 alpha (CK1α) agonist pyrvinium in endometriosis. Human endometrial stromal cells and an in vivo mouse model of endometriosis were employed. Quantitative PCR was used to assess the mRNA levels of CK1α and autophagy-related genes, while Western blot analysis measured their protein expression. Immunohistochemical staining was performed to detect CK1α and Atg7 in ectopic lesion tissues.

BACKGROUND: Pelvic endometriosis is an estrogen-driven inflammatory syndrome of unknown origin that alters the peritoneal microenvironment and likely impairs endometrial receptivity, adversely affecting fertility. Chronic endometritis (CE) may be a potential contributing factor to reduced endometrial receptivity in endometriosis. The aim of the study was to analyze the correlation between pelvic endometriosis and CE.
METHODS: The study included women undergoing laparoscopy for suspected pelvic endometriosis, and each underwent endometrial aspiration biopsy for CE. The stage of endometriosis w

Chronic endometritis (CE) is a persistent inflammatory disorder of the endometrium, associated with infertility, recurrent pregnancy loss, and implantation failure. Diagnosis primarily depends on hysteroscopy and immunohistochemistry, while microbial dysbiosis and antibiotic resistance pose significant challenges to effective management. The pathogenesis of CE involves microbial infections that induce immune dysregulation through TLR/NLR signaling pathways, metabolic reprogramming of immune cells, miRNA-mediated inflammatory responses, and DNA methylation alterations. The activation of pro-inf

Macrophage dysregulation is a feature of ulcerative colitis (UC), a chronic inflammatory bowel disease (IBD). Exosomes generated from endometrial regenerative cells (ERCs) (ERC-Exos) may influence the immunological dysfunction associated with IBD. In immunological diseases, Sirtuin 6 (SIRT6) reduces NF-κB signalling due to its protective benefits. Building on these findings, this study investigates the function and mechanism of SIRT6 derived from ERC-Exos in mediating the relief of UC. Lentiviral transduction was used to achieve SIRT6-specific knockdown ERC-Exos (SIRT6-KD-ERC-Exos). Dextran su

Endometriosis is an estrogen-dependent, chronic inflammatory disorder characterized by the presence of endometrium-like tissue outside the uterus, affecting approximately 10% of individuals of reproductive age. It contributes to chronic pelvic pain, dysmenorrhea, and subfertility, resulting in substantial societal economic burdens. Genetic and environmental risk factors have been identified, and recent research suggests that endometriosis functions as a systemic disease affecting nonreproductive systems and increasing susceptibility to other health conditions. Various phenotypes-superficial pe

BACKGROUND: Endometriosis is a chronic inflammatory disease with a prevalence of approximately 10% in women of childbearing age. Metabolic pathways have been demonstrated by previous studies to be potential avenues for the development of new therapeutic strategies and may be used for early diagnosis of the disease. This study aimed to investigate the potential causal relationships between 1400 metabolites and various endometriosis subtypes using Mendelian randomisation (MR) analysis.
METHODS: Data from a genome-wide association study were analysed. MR analysis was performed using the inverse-

Endometriosis is a prevalent chronic inflammatory condition impacting 5-10% of reproductive-age women, commonly resulting in debilitating pelvic pain and infertility. Despite extensive research efforts, the precise underlying pathophysiology remains largely unclear. Emerging evidence increasingly suggests that cellular senescence, iron overload, and fibrosis collectively form a critical pathological axis that significantly contributes to the persistence and severity of the disease. However, the intricate mechanistic interplay between the immune system's failure to effectively clear senescent c

Uterine fibroids (UFs) are benign smooth-muscle tumors of the uterus that commonly affect women of reproductive age and can influence systemic hormonal and inflammatory states. Osteoarthritis (OA) is a chronic degenerative disease of weight-bearing joints (e.g., hip and knee) and a leading cause of pain and disability. Shared hormonal and immune pathways plausibly link UFs and OA; however, whether a genetic or biological association exists remains unclear. The present study utilized Mendelian randomization (MR) analysis combined with experimental validation to investigate a potential genetic a

Endometriosis (EM) is a chronic inflammatory disorder characterized by the ectopic growth of endometrial-like tissue outside the uterine cavity. Its pathogenesis is closely linked to an imbalanced immune–inflammatory–angiogenic microenvironment. Core pathological features include immune suppression (e.g., M2 macrophage polarization, regulatory T cell increase), chronic inflammation (e.g., elevated IL-6, TNF-α, NF-κB/NLRP3 activation), and hypoxia-driven aberrant angiogenesis via the VEGF/HIF-1α axis. These components interact through shared molecular pathways—such as HIF-1α and NF-κB synergist

Endometriosis is a chronic systemic disease characterized by the presence of endometrium-like glands and/or stroma outside the uterus, usually with an associated inflammatory process. It affects around 5-10% of women worldwide, with chronic pelvic pain and infertility being the predominant symptoms. Although suspected clinically, transvaginal ultrasound (TVUS) and magnetic resonance imaging (MRI) are the primary imaging tools for diagnosing pelvic endometriosis, as well as helping in pre-surgical mapping. Computed tomography (CT) is not the preferred imaging modality for detection or depiction
