The interactive relationship between Fusobacterium necrophorum, Bacteroides pyogenes, and Porphyromonas levii in driving inflammatory uterine disease.

BACKGROUND: Bovine metritis is associated with uterine microbiota dysbiosis, characterized by the proliferation of Gram-negative anaerobes, including Fusobacterium necrophorum, Bacteroides pyogenes, and Porphyromonas levii. However, the mechanisms by which these opportunistic pathogens proliferate together and contribute to disease remain unclear. This study aimed to identify the interactions among these bacteria and to elucidate their role in the development of metritis. METHODS: F. necrophorum, B. pyogenes, and P. levii were isolated from the uteri of cows with metritis and cultured anaerob

Puerarin and DHEA combination therapy alleviates primary dysmenorrhea via inhibition of the Hsp90ab1/p38/JNK pathway.

Primary dysmenorrhea (PD), a prevalent gynecological disorder, is fundamentally driven by chronic uterine inflammation, leading to severe lower abdominal pain and recurrent cramping. Given the known anti-inflammatory properties of Puerarin (Pue) and Dehydroepiandrosterone (DHEA), we investigated their individual and combined therapeutic effects on PD using an estradiol benzoate/oxytocin-induced mouse model. Our findings revealed that both Pue and DHEA alleviated PD symptoms, as evidenced by reduced writhing frequency, prolonged inter-writhe intervals, diminished uterine edema, lower uterine PG

Targeting the Wnt signaling network in endometriosis: mechanistic insights and translational opportunities†.

Endometriosis is a chronic gynecological disorder defined by the growth of endometrial-like tissue outside the uterus, driven by estrogen-dependent inflammation and progressive fibrosis. According to available data, remodeling and persistent abnormal activation of the Wnt/β-catenin signaling network constitute one of the main molecular bases underlying its progression. Pathological characteristics of this disease include increased proliferation and anti-apoptotic potential of ectopic endometrial cells, as well as invasive phenotypes and progressive fibrosis. The Wnt/β-catenin pathway contribut

Neutrophil-to-lymphocyte ratio (nlr) as a predictor of endometriosis occurrence and prognosis: Evidence from a single-center study in Indonesia.

INTODUCTION: Endometriosis is a chronic inflammatory condition characterized by the presence of endometrial tissue outside the uterus, leading to pelvic pain and infertility. It affects approximately 10% of women of reproductive age. Endometriosis is associated with a chronic inflammatory process that results in pain, tissue remodelling, fibrosis, adhesions, and infertility. Numerous chronic inflammatory indicators can serve as practical prognostic and diagnostic biomarkers for the severity of the disease, one of which is the neutrophil-lymphocyte ratio (NLR). The normal range of NLR is 1-3, w

Endometrial expression of T-cell immunoreceptor with Ig and ITIM domains and cluster of differentiation 155: A case-control study of a novel immunomodulatory axis in endometriosis.

BACKGROUND: Endometriosis is a chronic inflammatory disorder affecting about 10% of females in their reproductive years, characterized by endometrial tissue growing outside the uterus. Immune checkpoints play a crucial role in regulating the immune system and preserving homeostasis. T-cell immunoreceptor with immunoglobulin and immunoreceptor tyrosine-based inhibitory motif domains(TIGIT), a newly discovered immune checkpoint, interacts with its ligand, cluster of differentiation 155(CD155), to exert inhibitory effects on immune responses. OBJECTIVE: Though numerous studies have explored the

Mesenchymal Stem Cells Therapy for Intrauterine Adhesions and Endometriosis: Potential, Mechanisms, and Future Directions.

Intrauterine adhesions (IUA) and endometriosis are debilitating gynecological disorders that impair endometrial function and fertility. IUA, typically caused by iatrogenic trauma to the basal endometrium, leads to fibrosis and infertility, whereas endometriosis, characterized by ectopic endometrial growth, induces chronic inflammation, pain, and subfertility. Current treatments, such as surgical adhesiolysis for IUA and hormonal suppression for endometriosis, frequently fail to address underlying pathological mechanisms, including aberrant fibrosis, inflammatory cascades, and impaired tissue r

Interactions Between Neurotrophins and Ovarian Steroids in Endometriosis and Their Implications for Neuroangiogenesis: A Narrative Review.

Endometriosis is a long-term gynecological condition marked by the growth of endometrial-like tissue outside the uterus, which undergoes proliferation, bleeding, and regeneration. This disease is associated with disrupted steroid hormone signaling, notably progesterone (P4) resistance and estradiol (E2) dominance. P4 resistance has been associated with impaired activation of the progesterone receptor (PR) and reduced transcription of P4 target genes, while elevated E2 levels induce estrogen receptor (ER)-mediated signaling, enhancing estrogen-dependent lesion growth. This hormonal imbalance co

RNF34 restrains endometriosis through SREBP1-dependent metabolic-immune crosstalk.

Endometriosis is a chronic inflammatory disease with cancer-like features, yet the mechanisms linking metabolic dysregulation to immune remodeling during lesion progression remain poorly understood. Here, we identify the ubiquitin E3 ligase RNF34 as a central suppressor of endometriosis that integrates cell-intrinsic metabolic control with macrophage-mediated immune regulation. Mechanistically, RNF34 directly interacts with SREBP1 and promotes its K48 and K63-linked ubiquitination and proteasomal degradation, thereby restraining lipogenic gene expression and fatty acid synthesis in endometrial

Stage-dependent sphingosine-1-phosphate pathway modulation as a therapeutic strategy in a murine model of adenomyosis.

Adenomyosis is characterized by chronic inflammation, myometrial remodeling, and subfertility, yet therapeutic options that preserve reproductive potential remain limited. Sphingosine-1-phosphate (S1P) signaling regulates immune-cell trafficking, vascular remodeling, and tissue invasion, but its contribution to adenomyosis remains poorly defined. This study investigated the temporal regulation of this pathway in a tamoxifen-induced murine model of adenomyosis and evaluated whether its pharmacological inhibition alters uterine immune profiles and lesion severity. A total of 122 female CD1 mice
