Vaccination Coverage and Prevention Counselling for Vaccine-Preventable STIs Among HIV PrEP Users in São Paulo, Brazil: A Retrospective Cohort Study.

INTRODUCTION: HIV pre-exposure prophylaxis (PrEP) users may be disproportionately vulnerable to sexually transmitted infections (STIs) in general, including several that are vaccine-preventable. Understanding immunization patterns in this population is, therefore, crucial. However, data on vaccination coverage among Brazilian PrEP users remains limited. METHODS: We conducted a retrospective single-centre study of adults using HIV PrEP at an STI clinic in São Paulo, Brazil, between 2017 and 2024, to assess vaccination adequacy for vaccine-preventable STIs among PrEP users, as well as other STI

Immunogenicity and safety of a dose-sparing inactivated poliovirus vaccine in infants in India: a phase 2/3, double-blind, randomised controlled trial.

BACKGROUND: Several countries are using fractionated or limited-dose regimens of full dose inactivated polio vaccine (IPV) in infants in addition to oral poliovirus vaccine (OPV), due to procurement cost of IPV and delivery challenges for its campaign use. An adjuvanted dose-sparing IPV (ds-IPV) with around a one-fourth antigen content of the full dose of IPV was developed in India. A non-inferiority trial was conducted to compare the immune response of ds-IPV with IPV in infants. METHODS: A phase 2/3, double-blind, randomised controlled trial was conducted at nine tertiary care hospitals in

Post-pandemic resurgence of Mycoplasma pneumoniae and its emerging link with Rhinovirus in children.

BACKGROUND: Mycoplasma pneumoniae (MPP) infections occur regularly every 1 to 3 years. A wave of infections was anticipated around late 2022 to early 2023. However, our data revealed a delayed onset, with a notable increase in cases in 2023/24. This study describes pediatric MPP detections during this resurgence and explores clinical differences between MPP mono-detection and MPP with rhinovirus/enterovirus (RV/EV) co-detection. METHODS: We retrospectively analyzed PCR-confirmed MPP detections in children aged 0-17 years presenting to a tertiary pediatric hospital between 2017 and 2024. Cases

Structured missingness in biomedical research.

Antipathogen defense; b-caryophyllene; vector-virus mutualism Plants respond to insect herbivory by producing and releasing distinctive blends of volatile compounds. These herbivore-induced plant volatiles (HIPVs) serve several roles, including direct defense through toxic or repellent effects on herbivores and indirect defense by attracting the herbivores' natural enemies (Bleeker et al. 2009; Veyrat et al. 2016). HIPVs also act as warning signals, informing undamaged tissues within the same plant and nearby plants of a pending insect attack (Heil and Bueno 2007). HIPVs are mostly composed of

BEV 2C protein inhibits the NF-κB signalling pathway to promote viral replication by targeting IKBKB and p65.

Bovine enterovirus, a member of the Enterovirus genus in the Picornaviridae family, causes severe digestive and respiratory illnesses in cattle. These illnesses threaten the healthy development of the cattle industry. Innate immunity plays a critical role in resisting viral infections, but viruses also use various strategies to evade or counteract the host's immune system. The mechanisms by which bovine enteroviruses evade the host immune response and promote their replication remain unclear. This study used the HY12 strain of enterovirus as a model to investigate its interaction with both bov

Mitigating murine acute and chronic Coxsackievirus B3-induced myocarditis with human right atrial appendage-derived stromal cells.

We previously have shown the potential of human endomyocardial biopsy (EMB)-derived cardiac adherent proliferating cells (CardAPs) as a new cell-therapeutic treatment option for virus-induced myocarditis. To overcome the limited cell yield per EMB, CardAPs have been isolated from the human right atrial appendage (RAA) in view of allogeneic application and off-the-shelf use. We aimed to investigate the cardioprotective and immunomodulatory potential of RAA-CardAPs in experimental acute and chronic Coxsackievirus B3 (CVB3)-induced myocarditis upon injection in the viral and inflammatory phase. Computational discovery of natural inhibitors targeting enterovirus D68 3C protease using molecular docking pharmacokinetics and dynamics simulations.

Enterovirus D68 (EV-D68) is a significant global health threat, responsible for severe respiratory and neurological complications, with no FDA-approved antiviral treatments currently available. The 3C protease of EV-D68, an essential enzyme involved in viral replication, represents a key target for therapeutic development. In this study, we employed a comprehensive computational approach, including molecular docking, pharmacokinetic predictions, and molecular dynamics simulations, to identify potential natural inhibitors of the EV-D68 3C protease. Screening a library of natural compounds, With

Association of Respiratory Pathogen Panel Testing on the Proportion of Emergency Department Revisits in Children With Symptoms of Upper Respiratory Illness.

OBJECTIVE: Acute respiratory tract infections are the main cause of morbidity and emergency department visits for children. Recent advances in testing have brought about the ability to quickly diagnose many of the pathogens contributing to viral respiratory illness. The purpose of the study is to evaluate respiratory pathogen panel testing versus clinical diagnosis on the proportion of emergency department (ED) same-site revisits in an uncomplicated, healthy pediatric population. METHODS: This was an IRB-approved, retrospective cohort study. Patients discharged from the ED between the ages of

Noncanonical Poly(A) Polymerase TENT4 Drives Expression of Subgenomic Hepatitis A Virus RNAs in Infected Cells.

Both hepatitis B virus (HBV), an hepadnavirus with a DNA genome, and hepatitis A virus (HAV), a picornavirus, require the TRAMP-like host ZCCHC14-TENT4 complex for efficient replication. However, whereas HBV requires the nucleotidyltransferase activity of TENT4 to extend and stabilize the 3' poly(A) tails of mRNA transcribed from its genome, the role played by TENT4 in HAV replication is uncertain. HAV proteins are synthesized directly from its genomic RNA, which possesses a 3' poly(A) tail, with its length and composition presumably maintained by 3Dpol-catalyzed RNA transcription during its r

A Fluorescent Reporter Virus Toolkit for Interrogating Enterovirus Biology and Host Interactions.

Enteroviruses are a group of highly prevalent human pathogens responsible for a wide range of illnesses, ranging from common cold symptoms to life-threatening diseases. A deep understanding of enterovirus biology, evolution, and host interaction is required for the development of effective vaccines and antivirals. Recombinant reporter viruses encoding luminescent or fluorescent proteins have been developed to facilitate such investigation. In this work, using coxsackievirus B3 as our model, we analyze how the insertion of fluorescent reporter genes at three distinct sites within the viral poly

A Pan-Enterovirus Natural Product Inhibitor Targeting a Unique Allosteric Site on the Viral 3C Protease.

Infections caused by Enterovirus like rhinoviruses, coxsackieviruses, and polioviruses represent a significant public health concern, for which there are no antivirals available yet. The highly conserved viral 3C protease has been the primary target for antiviral development, but competitive inhibitors targeting its active site does not meet expectations in clinical studies. Previously, an unconventional allosteric site is identified on human rhinovirus 14 (HRV14) 3C, representing novel opportunities for pan-enterovirus antivirals development. Here, in silico screening of 143,621 natural produ

[Genomic characterization of a case of enterovirus D68 infection in a child from Tongzhou District, Beijing City].

A throat swab sample from a pediatric case in Tongzhou District, Beijing was identified as enterovirus; the patient was a 1-year-and-8-month-old male sporadic case. Whole-genome sequencing revealed a viral genome length of 7 436 bp. BLAST alignment confirmed the serotype as EV-D68. Phylogenetic analysis of the whole genome indicated that this strain belongs to the B3 clade, showing closer genetic proximity to the 2018 Shanghai strain MW697453 with 99.53% whole-genome nucleotide homology. Genetic and amino acid variation analysis demonstrated that the B3 subclade to which this strain belongs ex

The multifaceted role of the viral 2A protease in enterovirus replication and antagonism of host antiviral responses.

Enteroviruses dramatically remodel the cellular infrastructure for efficient replication and curtailing host antiviral responses. The roles of viral proteins in these processes have been studied mostly in vitro, by ectopic overexpression, or by surrogate infection systems, all of which have shortcomings. Here, we replace the essential 2A cleavage site at the P1-P2 junction with an internal ribosome entry site (IRES), 3CD cleavage site, or T2A sequence, allowing us to catalytically inactivate 2Apro in the virus context. Viruses with an inactive 2Apro are hampered in replication in cell lines an

RAA-CRISPR/Cas12a-based visual field detection system for rapid and sensitive diagnosis of major viral pathogens in calf diarrhea.

Calf diarrhea is a complex digestive disorder in cattle that imposes significant economic losses in terms of calf mortality, growth impairment, and treatment costs. Both infectious and non-infectious agents contribute to its aetiology; however, most of the infectious cases are caused by viruses, often accompanied by severe co-infections. To identify viral culprits, we performed viral metagenomic sequencing on three pooled samples from the 150 diarrheal samples from Xinjiang, China, which helped with identification of the following four predominant agents: bovine nepovirus (BNeV), bovine corona

Classification of hepatitis A virus in clades based on VP1/2A region allows higher geographic and temporal resolution than conventional genotyping: a global sequences analysis.

Infections caused by the Hepatitis A virus (HAV) occur worldwide. Molecular epidemiology is crucial to understanding the transmission dynamics; however, the current genotype assignment fails to provide useful epidemiologic information. We classified HAV sequences available in GenBank based on 460 bp of the VP1/2A region in clades. Additionally, we associated the site of isolation of the strains with the country's HAV endemicity. We analyzed 1369 sequences available in GenBank up to December 2023 which fulfilled inclusion criteria for our study. The collection date ranged from 1957 to 2023, and

Subacute Thyroiditis-Is it Really Linked to Viral Infection?

CONTEXT: Subacute thyroiditis (SAT) is a painful inflammatory disorder of the thyroid gland, which-after a phase of thyrotoxicosis-leads to transient, or less frequently permanent hypothyroidism. Apart from a strong association with specific human leukocyte antigen alleles, the causes are uncertain. Viral disease has been hypothesized as a trigger, with enteroviruses, namely echovirus and coxsackievirus, showing a seasonal distribution that coincides with the incidence of SAT. OBJECTIVE: In the first year of the COVID-19 pandemic, strict hygiene measures led to a sharp decline in infections a
