Total glucosides of Paeony alleviates chronic endometritis in rats through regulating gut microbiota composition and inhibiting TLR4/NF-κB/NLRP3 pathway.

OBJECTIVE: This study aimed to elucidate the functional role and regulating mechanism of Total Glucosides of Paeony (TGP) in chronic endometritis (CE). METHODS: To assess the impact of TGP on CE, a CE rat model was established via infusion with 60 µL of 5.0 mg/mL LPS, while human endometrial stromal cells (HESCs) were pre-treated with 500 ng/mL LPS to simulate a CE cell model. Cell proliferation and apoptosis were evaluated using CCK-8, Flow cytometry and EDU staining techniques. Histological changes in uterine tissues were evaluated using H&E staining, while NLRP3 expression was analyzed via

Pre-Weaning Administration of a PGF2α Analogue (Cloprostenol) and Its Influence on Early Reproductive Indicators in Sows Under Commercial Farm Conditions.

Efficient reproductive management in commercial swine production requires minimising non-productive days and ensuring rapid return to oestrus after weaning. The present study evaluated the effect of pre-weaning administration of cloprostenol, a PGF2α analogue, on early reproductive indicators in sows under commercial farm conditions. A total of 734 clinically healthy multiparous sows (parity 2-9) from a commercial farm were randomly allocated to two groups before weaning. Control sows (n = 367) received no treatment, whereas treated sows (n = 367) received an intramuscular injection of clopros

The microbiome-inflammasome axis in endometriosis-associated chronic pelvic pain: mechanisms and therapeutic opportunities from an immunological perspective.

INTRODUCTION: Endometriosis (EMs) is increasingly recognized as a chronic inflammatory disease in which innate immune dysregulation plays an important role. Its immunopathological features include macrophage polarization imbalance, with M1-skewed activation in inflamed peritoneal fluid and lesions, reduced natural killer (NK) cell cytotoxicity, adaptive immune alterations including Th2/Treg-skewed profiles reported in some studies, and persistently elevated pro-inflammatory cytokines. The disease affects approximately 10% of women of reproductive age worldwide, and 70%-80% of these patients ex

Predicting Endometriosis Status and Menstrual Cycle Phase Using DNA Methylation.

Endometriosis is a chronic inflammatory disease associated with pelvic pain, infertility, and delayed diagnosis. Growing evidence suggests that altered DNA methylation contributes to disease development and could serve as a biomarker for disease. We developed a leakage-safe machine learning pipeline to classify endometriosis case-control status and menstrual cycle phase using genome-wide DNA methylation data from eutopic endometrial tissue. The dataset consisted of 984 samples profiled using the Illumina Infinium MethylationEPIC array, with measurements across approximately 759,000 CpG sites.

Possible role of bacterial infection in embryo implantation failure.

Embryo implantation is a critical and complex process that requires coordination between the embryo and the receptive endometrium for its success. However, a multitude of factors have the potential to contribute to implantation failure. Among these factors, bacterial infections in the uterine environment are of particular concern and are increasingly being considered as a contributing factor to implantation failure. The presence of pathogenic bacteria has been demonstrated to stimulate inflammatory responses, alter the endometrial microbiome, interfere with critical signaling pathways, and fin

Barriers to Adherence to Remote Pelvic Floor Muscle Training Among Postpartum Women: A Qualitative Study Guided by the COM-B Model.

BACKGROUND: Pelvic floor muscle training (PFMT) is recognized as the first-line conservative intervention for postpartum pelvic floor rehabilitation. Remote pelvic floor muscle training, defined as a training modality conducted at home via mobile applications, online platforms, teleconsultation, or application-based reminders, without real-time on-site supervision, has expanded the accessibility of rehabilitation services; however, patient adherence remains suboptimal. Elucidating the barriers to adherence is critical for developing effective and well-accepted intervention strategies. This stu

Establishment and characterization of a new immortalized human adenomyosis epithelial-like cell line, tAEC21†.

Adenomyosis occurs when endometrial glands and stroma grow within the uterine myometrium. As a clinically significant disease, adenomyosis causes substantial pelvic pain and heavy menstrual bleeding. It remains understudied due to a lack of translational research tools and model systems. This study aimed to develop a telomerase-transformed, epithelial-like cell line derived from the eutopic endometrium of a subject with focal adenomyosis. De-identified endometrial tissue was processed through mechanical and enzymatic digestion. Epithelial and stromal populations were separated by selective adh

Synchronized breeding and reproductive resilience in dairy cattle: mitigating metritis, heat stress, and transgenerational fertility impairment.

Heat stress profoundly compromises reproductive efficiency in dairy cattle by disrupting follicular and oocyte development, altering steroid hormone profiles, impairing estrous expression and ovulation, reducing fertilization success, hindering embryonic development, and increasing early embryonic loss. Although the molecular mechanisms underlying these disruptions remain incompletely understood, significant advances in reproductive technologies have been developed to mitigate the negative impact of heat stress on reproductive performance. Estrous cycle synchronization programs, enabling timed

Surgical site infections after cesarean delivery.

Surgical site infections represent a persistent and clinically relevant complication of cesarean delivery, contributing substantially to postpartum morbidity, hospital readmission, and pregnancy-related mortality. As cesarean delivery has escalated to the most common major operation worldwide-now comprising 21% of global births and 32% in the United States-the burden of postcesarean surgical site infections, reported in 3% to 15% of cases, remains unacceptably high despite advances in perioperative antisepsis, antibiotic prophylaxis, and prevention bundles. This review provides an updated comp
