Household Transmission and Genomic Diversity of Respiratory Syncytial Virus in the United States, 2022-2023.

BACKGROUND: Household transmission of respiratory viruses may drive community spread. Few recent studies have examined household respiratory syncytial virus (RSV) transmission in the United States. METHODS: We conducted a prospective community-based cohort study from 1 June 2022 to 31 May 2023. Participants had blood samples collected and completed nasal swabs and surveys at least weekly, irrespective of symptoms. We tested serum for RSV antibody, nasal swabs by quantitative reverse transcription polymerase chain reaction (RT-qPCR), and performed whole genome sequencing. We evaluated secondar

Self-reported reactogenicity after COVID-19 vaccination with and without influenza vaccine coadministration during pregnancy in Canada.

BACKGROUND: Despite consistent evidence of safety and efficacy of immunization against COVID-19, vaccine hesitancy remains prevalent among pregnant individuals. Reactogenicity (post-vaccination inflammatory symptoms) has been cited as a significant factor in decreasing vaccine acceptance. The seasonal overlap in the vaccination schedule for COVID-19 and influenza provides an opportunity to increase vaccine accessibility and uptake while decreasing healthcare burden through coadministration. METHODS: The Canadian COVID-19 Vaccine Registry for Pregnant Women and Pregnant People (COVERED) is a n

Computational Investigation of Natural Substances as SARS-CoV-2 Main Protease Inhibitors: A Virtual Screening Method.

INTRODUCTION: The coronavirus disease 2019 (COVID-19) pandemic, caused the by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has had a profound impact on public health, overburdening healthcare systems, and disrupting global economies. Moreover, the absence of specific antiviral drugs remains a major challenge in COVID-19 treatment. The SARS-CoV-2 main protease (Mpro) is a crucial therapeutic target due to its essential role in viral replication. The objective of this study was to identify natural compounds with potential inhibitory activity against SARS-CoV-2 Mpro, which could

A lossless one-shot distributed algorithm for addressing heterogeneity in multi-site generalized linear models.

OBJECTIVE: We propose Heterogeneity-aware Collaborative One-shot Lossless Algorithm for Generalized Linear Model (COLA-GLM-H), a novel one-shot lossless distributed algorithm that enables the integration of heterogeneous multi-institutional data while relying solely on instituion-level summary information rather than patient-level data. MATERIALS AND METHODS: Generalized Linear Models (GLMs) are widely used in medical research for analyzing diverse outcome types. In multi-institution settings, we demonstrated that the global likelihood can be reconstructed using only institution-level summary

Assessing SARS-CoV-2 transmission in African households from the reanalysis of serosurveys.

Household transmission studies provided key insights on SARS-CoV-2 transmission in high-income countries but were rarely implemented in Africa. To help fill this gap, we analyzed SARS-CoV-2 seroprevalence studies with a household-based recruitment, focusing on households with ≤7 members, in four Sub-Saharan African cities: Kinshasa (82 households, 370 individuals), Lubumbashi (225 households, 970 individuals), Conakry (149 households, 649 individuals), and Yaoundé (311 households, 1183 individuals), between late 2020 and mid-2021. Using an extended chain-binomial model accounting for missing s

Occupational determinants of Long COVID in the population-based COVICAT cohort.

OBJECTIVES: Occupational factors affect SARS-CoV-2 infection risk, but the occupational factors associated with Long COVID (LC) are unknown. We aimed to address this issue using individual data in a population-based cohort. METHODS: In the prospective COVICAT study, 2020-2023, Catalonia, Spain, we examined the association between occupational determinants and LC. Among subjects with previous SARS-CoV-2 infection, those employed in the pandemic and with occupational information were analysed. Different metrics, including four job-exposure matrices, were used to evaluate individual occupational

Atherosclerosis licenses for an exceeding immune response in COVID-19 disease by interferon priming in circulating myeloid cells.

AIMS: Patients with cardiovascular disease (CVD) have an increased risk of developing severe respiratory infections, including COVID-19. However, the underlying molecular mechanisms are not completely understood. It has been previously shown that CVD predisposes to an altered responsiveness to subsequent inflammatory triggers by an imprinted epigenetic memory in innate immune cells. Therefore, we hypothesized that patients with pre-existing atherosclerotic cardiovascular disease (ASCVD) and COVID-19 display a dysregulated inflammatory response compared to patients without ASCVD due to epigenet

Charged scanning mutagenesis as a high-throughput approach for epitope mapping.

Identifying neutralizing epitopes is important for developing vaccines and inhibitors against viral pathogens. We describe a rapid method for epitope mapping, employing barcoded charged scanning mutagenesis libraries displayed on the yeast surface, and screening using flow cytometry coupled with deep sequencing. Prior scanning mutagenesis data suggest that mutations to a charged residue, such as Aspartic acid or Arginine, will be well tolerated at exposed positions of an antigen and minimally affect protein stability and expression. Yet such substitutions at epitope residues strongly perturb b

Adverse Events Following Sinopharm Covid-19 Vaccination among Medical Students.

To develop immunity against Covid-19, vaccination was prioritized by World Health Organization (WHO). According to WHO, at least 70% of a population should be vaccinated to develop herd immunity. For this purpose, several kinds of vaccination had been introduced. One of them was Sinopharm inactivated Covid 19 vaccine, that was administered for a trial purpose to the medical students from 16th august 2021. So our purpose was to see the side effects following vaccination to sweep out the fear regarding Sinopharm vaccination. This cross-sectional analytical study was carried out from June to Sept

Experimental intranasal infection reveals broad tissue tropism of bovine coronavirus.

Bovine coronavirus (BCoV) causes neonatal calf diarrhea, winter dysentery, and respiratory disease worldwide. However, its primary replication site remains uncertain, and systemic studies on tissue tropism are limited. We, therefore, intranasally inoculated colostrum-fed Holstein calves (6-13-day-old) with a BCoV isolate derived from diarrheic calf, and monitored clinical signs, gross and histopathological lesions, antigen distribution, and viral replication in multiple tissues. All calves were seropositive before challenge. Infected animals developed diarrhea without respiratory manifestation

Choroid plexus alterations in long COVID and their associations with IL-6.

SARS-CoV-2 disrupts the choroid plexus (ChP) epithelium by binding to the ACE-2 receptor, causing blood cerebrospinal fluid barrier leakage and permitting interleukin (IL)-6 and pathogens into the brain, subsequently leading to demyelination, white matter (WM) damage in long COVID, and clinical worsening. The role of the ChP in long COVID and its relationships to WM integrity, IL-6, clinical symptoms, and ACEIs/ARBs medications remains unclear. Fifty-two long COVID individuals, 21 COVID-19 survivors, and 26 healthy controls (HCs) completed Montgomery-Asberg Depression Rating Scale (MADRS), Mon

Metal-Organic Framework@Gold Nanoparticles Enables Point-of-Care Diagnostics To Have Comparable Analytical Performance to Chemiluminescent Immunoassays.

Lateral flow immunoassay (LFIA) is a vital point-of-care testing (POCT) technique that is widely used for on-site detection and in vitro diagnosis. Many diseases, particularly cardiovascular diseases (CVDs), require the quantitative detection of multiple biomarkers across a broad dynamic range. Recurrent viral infections like COVID-19 elevate CVDs risk, necessitating early warning and prognostic monitoring, especially based on POCT of multiple blood biomarkers of D-dimer, NT-proBNP, and cTnI across a broad range from pg/mL to μg/mL. Traditional LFIAs lack ultrasensitivity and broad-range quant

Two-year persistence of MERS-CoV-specific antibody and T cell responses after MVA-MERS-S vaccination in healthy adults.

MVA-MERS-S, a vaccine candidate against Middle East respiratory syndrome (MERS), was recently evaluated in a randomized, placebo-controlled, double-blind phase 1b clinical trial to assess its safety, immunogenicity, and optimal dosing in healthy adults in Hamburg and Rotterdam. A three-dose regimen was safe and elicited robust spike-specific antibody responses. We extended this trial to assess the two-year durability of MERS-CoV-specific antibody and T cell responses in 48 study participants of the Hamburg cohort. Our findings show that immune responses remain detectable for at least 24 months

Acceptability of and Implementation Supports for Video Directly Observed Treatment to Enhance Methadone Dosing Flexibility in a Multisite Opioid Treatment Program: Qualitative Rapid Needs Assessment Study.

BACKGROUND: Methadone is a first-line treatment for opioid use disorder, which is delivered in federally regulated opioid treatment programs (OTPs). Federal policies require directly observed dosing of methadone followed by graduated provision of nonobserved doses to take at home (ie, "take-home" dosing) after demonstrated stability is achieved. Policy changes since the COVID-19 pandemic have greatly expanded take-home dosing. Video directly observed treatment (video DOT) is an approach in which patients submit videos of themselves taking medications, which are asynchronously reviewed to verif
