Detection and Molecular Characterisation of Protoparvovirus carnivoran1 in Golden Jackals (Canis aureus) in Croatia.

Protoparvoviruses are highly contagious pathogens that cause severe, often fatal diseases in both domestic and wild carnivores. Golden jackal (Canis aureus) populations have experienced expansion in recent years, increasingly occupying urban and peri-urban areas. Despite this, they remain largely overlooked in scientific research. This study aimed to detect and characterise Protoparvovirus carnivoran1 circulating in a golden jackal population in Croatia and to assess their role in the epidemiology of parvovirus infections in companion animals. Small intestines from 55 jackals hunted in 2024 an

Ultrastructural pathology and temporal patterns of viral replication and lesion development following mouse kidney parvovirus infection in B6, CD1, and NSG mice.

Mouse kidney parvovirus (MKPV) causes inclusion body nephropathy, resulting in clinical signs and mortality in immunodeficient mice and subclinical infection in immunocompetent mice. While late-stage renal lesions and viral replication have been characterized, a comprehensive multisystemic investigation of MKPV infection from the initial to the late stages of infection has not been conducted. Our goal was to investigate lesions and viral replication in all major organs at multiple stages of MKPV infection in immunocompetent C57BL/6NCrl (B6) and Crl: CD1(ICR) (CD1) mice and immunodeficient NOD.

Eco-evolutionary dynamics of a shifting porcine parvoviruses (PPV1-PPV8) ecosystem reveal dichotomous selection pressures.

Porcine parvoviruses (PPVs) constitute a complex viral ecosystem. However, the genetic structure and evolutionary dynamics of currently circulating PPV populations remain poorly characterized. A large-scale genomic survey in China (1,055 genomes; 863 samples; 2021-2024) reveal a profound ecosystem shift, with PPVs detected in 212/863 samples (24.57%) Novel species (PPV7, 16.45%; PPV6, 9.62%; and PPV2, 8.82%) now dominate the niche previously occupied by classical PPV1 (3.13%). This new landscape is defined by pervasive multi-species co-infection (129/212, 60.85%), including 61 cases with ≥ 3 s

Antibody responses to inactivated (adjuvanted) and attenuated (non-adjuvanted) feline panleukopaenia virus vaccination, including in different injection sites.

ObjectivesVaccines have traditionally been administered to cats into the interscapular subcutaneous region ('scruff') for ease of restraining the cat while the vaccine is injected. Concern about feline injection-site sarcomas (FISSs) has prompted recommendations to use distal sites such as the hindlimb or tail, but evidence of adequate immune response at these sites is limited. This field study evaluated the antibody response to feline panleukopaenia virus (FPV) in kittens vaccinated with one of two commercial vaccines, injected at different sites.MethodsAn inactivated (killed), adjuvanted pen

A rapid on-site diagnostic method for goose parvovirus disease based on recombinase polymerase amplification and CRISPR/AsCas12a.

Goose parvovirus (GPV) is a highly pathogenic and lethal virus responsible for Derzsy's disease in goslings and ducklings, significantly influencing the economic viability of waterfowl farming. This necessitates the development of rapid diagnostic techniques for effective disease management. In this study, an optimized clustered regularly interspaced short palindromic repeats (CRISPR)/Acidaminococcus sp. CRISPR associated nuclease 12a (AsCas12a) system was developed for the diagnosis of GPV. The study determined that the optimal conditions for the CRISPR/Cas12a-based fluorescence assay were 20

Phylogenetic analysis and molecular structure of NS1 proteins of porcine parvovirus 5 isolates from Mexico.

Porcine parvovirus 5 (PPV5) is an unclassified member of the family Parvoviridae with no reported pathogenicity, although it is associated with multisystemic, reproductive, and respiratory diseases. Its open reading frame 1 (ORF1) encodes non-structural protein 1 (NS1), which is predicted to have helicase activity that is essential for viral replication. This protein contains a C-motif with an invariant asparagine residue that forms the core of the enzyme's active site, in conjunction with the Walker A and B motifs. The aim of this study was the phylogenetic and molecular characterization of t

Structures and functions of the limited natural polyclonal antibody response to parvovirus infection.

Host antibody responses are key components in the protection of animals against pathogens, yet the defining properties of viral antigens and induction of B cell responses that result in varied protection are still poorly understood. Parvoviruses are simple molecular structures that display 60 repeated motifs on their capsid surface, and rapidly induce strong antibody responses that protect animals from infection. We recently showed that following canine parvovirus infection of its natural host, the polyclonal response in the sera contained only two or three dominant antibodies that bound two e

Discovery of miRNAs unique to actively transcribed erythroparvovirus infection in heart failure patients.

AIMS: miRNAs, small non-coding RNAs, play key roles in gene regulation, cell differentiation and tissue development. They influence viral infection outcomes by directly interacting with viral genomes or modifying the host microenvironment. This study demonstrates miRNAs' ability to selectively suppress transcriptionally active erythroparvovirus, highlighting their potential in antiviral therapies. METHODS AND RESULTS: Seventy-five endomyocardial biopsy (EMB) specimens from patients with unexplained heart failure were analysed. The samples included 19 with dilated cardiomyopathy and inflammati

Capsid Structure of the Fish Pathogen Syngnathus Scovelli Chapparvovirus Offers a New Perspective on Parvovirus Structural Biology.

Chapparvoviruses (ChPVs) comprise a divergent lineage of the Parvoviridae ssDNA virus family and evolved to infect vertebrate animals independently from the Parvovirinae subfamily. Despite being pathogenic and widespread in environmental samples and metagenomic assemblies, their structural characterization has proven challenging. Here, we report the first structural analysis of a ChPV, represented by the fish pathogen, Syngnathus scovelli chapparvovirus (SsChPV). We show through the SsChPV structure that the lineage harbors a surface morphology, subunit structure, and multimer interactions tha

Genetic characteristics of human bocavirus in children with acute respiratory tract infections during 2023 in Beijing, China.

Human bocavirus (HBoV) is an emerging pathogen associated with pediatric respiratory and gastrointestinal infections, yet its genetic diversity remains understudied. The aberrant global resurgence of other respiratory viruses after the COVID-19 pandemic has raised concerns regarding significant evolutionary shifts in viral genetic profiles. Accordingly, this study characterized HBoV isolates that circulated in Beijing during 2023 by analyzing 1,442 pediatric respiratory samples associated with acute respiratory tract infections. Among 43 HBoV-positive cases with a 2.98% detection rate, 11 comp

A combination of recombinase polymerase amplification with CRISPR technology rapidly detects goose parvovirus with high accuracy and sensitivity.

BACKGROUND: Goose parvovirus (GPV) poses a significant threat to the waterfowl industry, necessitating reliable detection methods. However, conventional techniques are often time-consuming, equipment-dependent, or lack sufficient sensitivity for detecting early-stage infection. In contrast, emerging CRISPR/Cas12a-based systems offer a promising alternative for rapid, sensitive, and on-site diagnostics. METHODS: We developed and optimized a recombinase polymerase amplification (RPA)-CRISPR/Cas12a assay targeting the conserved VP3 gene of GPV. The analytical and diagnostic performance of this a

Residue 301-dependent epitope mapping reveals the molecular basis for GPV/MDPV serotype discrimination by neutralizing monoclonal antibody D1.

Waterfowl parvoviruses (goose parvovirus, GPV; Muscovy duck parvovirus, MDPV; Muscovy duck-origin goose parvovirus, MDGPV; short beak and dwarfism syndrome virus, SBDSV) are significant pathogens in waterfowl. The monoclonal antibody D1 specifically recognizes GPV-serotype parvoviruses (including GPV, SBDSV, and MDGPV) but cannot bind MDPV. Thus, Western blot and indirect immunofluorescence assays confirmed that D1 recognizes a conformational epitope, and residue 301 is critical for its discriminatory binding. A rapid, visual, ultrasensitive and highly specific method for detecting adeno-associated virus 2020 based on the LAMP-CRISPR/Cas12a system.

Avian parvovirus infection would lead to growth retardation, weight loss, physical deformities and increased mortality in poultry, causing substantial economic losses to the poultry industry. Therefore, the development of a rapid, visual, ultrasensitive and highly specific method is essential for timely diagnosis and effective control of the avian parvovirus infection. In this study, we developed a detection platform based on loop-mediated isothermal amplification (LAMP) combined with the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 12a (Cas12a)
