Early Versus Late Enzyme Replacement Therapy in Siblings With Morquio A Syndrome: Insights Into Therapeutic Timing.

Enzyme replacement therapy (ERT) with elosulfase alfa is the only approved treatment for mucopolysaccharidosis type IVA. This case report delineates the 5-year outcomes of ERT in two Korean siblings with mucopolysaccharidosis type IVA, with the younger sibling initiating treatment at 0.8 years of age and the older one at 5.4 years. Both patients exhibited a progressive decline in their height standard deviation scores, with trajectories paralleling the natural history curves of the disease. At 5.2 years of age, persistent skeletal dysplasia was evident in both siblings. However, the younger si

Utility of PAX1/JAM3 methylation analysis for triage of high-risk HPV-positive individuals.

INTRODUCTION: We sought to assess the clinical utility of methylation detection of paired box gene 1 (PAX1) and junctional adhesion molecule 3 (JAM3) in the triage of individuals testing positive for high-risk human papillomavirus (HPV). METHODS: Cervical secretions from 312 high-risk HPV-positive patients were analyzed for dual-gene methylation of PAX1 and JAM3 (PAX1m/JAM3m). Methylation levels were compared across histologically confirmed cervical lesions. Using histopathology as the reference standard, the triage performance of PAX1m/JAM3m was evaluated against cytology and HPV-16/18 genot

Association Between HPV Vaccination and Cervical Dysplasia Severity in HPV-Positive Women.

Background: Although HPV vaccination is highly effective in the primary prevention of cervical cancer, its potential role in women already diagnosed with HPV-associated cervical dysplasia remains uncertain. This study aimed to evaluate the association between post-diagnosis HPV vaccination and short-term clinical outcomes in HPV-positive women with cervical dysplasia. Methods: Women aged ≥18 years with abnormal cervical screening results suggestive of squamous intraepithelial lesions and high-risk HPV positivity were retrospectively evaluated. High-grade disease was defined as histologically c

Negative Xpert HPV Test (Cepheid) With Late Amplification Signals: Is There a Clinical Significance? (the PaPCR Study).

Cervical cancer is the fourth most common cancer among women worldwide, with higher prevalence in low-resource countries due to limited access to screening and HPV vaccination. Nucleic acid amplification tests (NAATs) such as Xpert HPV (Cepheid), enable rapid detection of high-risk HPV (HR-HPV) genotypes. However, the clinical significance of weak and late amplification signals (cycle threshold ≥ 38), interpreted as negative, remains unclear. In this study, a comparison with cytological results aimed to assess their significance. Out of 6846 cervical samples analyzed at Brest University Hospit

Balancing diagnostic accuracy and cost in the follow-up of histological low-grade squamous intraepithelial lesions.

INTRODUCTION: Primary cervical screening with testing for human papillomavirus (HPV) brings increased detection of women with low-grade squamous intraepithelial lesion recommended for follow-up with colposcopy. This study evaluates whether co-testing (HPV + cytology) or testing for HPV alone can safely replace colposcopy as follow-up for histologically confirmed low-grade squamous intraepithelial lesions. It estimates these strategies' diagnostic performance in detecting histological high-grade squamous intraepithelial or more severe lesions at follow-up and analyzes their relative cost-effect

Mechanism of activation of an ancestral TEC kinase by PIP3.

BACKGROUND: The introduction of second-generation antipsychotics (SGAs) over the past 2 decades generated considerable optimism that better antipsychotic treatments for schizophrenia and bipolar disorder were possible. SGAs offer several tolerability benefits over first-generation antipsychotics (FGAs), particularly with respect to extrapyramidal symptoms. However, SGAs can induce serious metabolic dysregulations, especially in drug-naive, first-episode, and child and adolescent populations, with olanzapine and clozapine having the highest propensity to cause these abnormalities. In this conte

Analysis of the clinical utility of p16/Ki-67 dual staining in screening cervical lesions among women positive for high-risk HPV.

Cervical cancer is closely associated with persistent infection by high-risk human papillomavirus (hrHPV). Although HPV testing offers high sensitivity, its limited specificity may result in excessive colposcopy referrals, and the triage performance of traditional liquid-based cytology (TCT) remains suboptimal among hrHPV-positive women. As a biomarker reflecting HPV-mediated cell-cycle dysregulation and aberrant proliferation, p16/Ki-67 dual staining has shown potential advantages in identifying clinically significant high-grade cervical lesions. This study aimed to evaluate the clinical util

Agreement and Diagnostic Performance of Urinary HPV Testing Versus Clinician-Collected Cervico-Vaginal Sampling: A Prospective Cross-Sectional Study in a Romanian Cohort.

Background: Cervical cancer screening uptake in Romania remains below 20%, with invasive sampling cited as a major participation barrier. Urinary HPV testing is a non-invasive alternative, but data from Eastern European populations are scarce and reporting stratified by histological grade is inconsistent. Objectives: To evaluate the agreement between paired urinary and clinician-collected cervico-vaginal HPV testing, and to assess the diagnostic performance of urinary HPV testing for biopsy-confirmed cervical intraepithelial neoplasia grade 2 or worse (CIN2+). Methods: Prospective cross-sectio

A paper-based HPV E7 oncoprotein assay for cervical precancer detection at the point of care.

Cervical cancer, while preventable through screening and treatment of cervical precancer, remains a global challenge with a disproportionately high burden of disease in resource-limited settings, especially in low- and middle-income countries (LMICs). Lack of affordable, easy-to-use screening and diagnostic tests contributes to this disparity. Most commercially available tests are not appropriate for use in LMICs due to resource constraints. Specifically, HPV mRNA and oncoprotein tests that have high specificity for cervical precancer and cancer require complex sample preparation protocols and

Analysis of HPV-16 viral load, integration status, and p16 expression in relation to EBV co-infection and cervical lesion severity.

Cervical cancer, one of the most common cancers in women, is primarily driven by high-risk human papillomaviruses (HPV) infections, particularly HPV-16. Co-infection with Epstein-Barr virus (EBV) has been reported to exacerbate disease progression by influencing HPV genome integration. This study examines HPV-16 integration status, p16INK4a expression, and their relationship with EBV co-infection and viral load in cervical cancer cases. In this study, 134 HPV-16-positive formalin-fixed, paraffin-embedded cervical samples were collected and analyzed for HPV-16 viral load, genome integration and
