An application of nowcasting methods: Cases of norovirus during the winter 2023/2024 in England.

BACKGROUND: Norovirus is a leading cause of acute gastroenteritis, adding to strain on healthcare systems. Diagnostic test reporting of norovirus is often delayed, resulting in incomplete data for real-time surveillance. METHODS: To nowcast the real-time case burden of norovirus a generalised additive model (GAM), semi-mechanistic Bayesian joint process and delay model "epinowcast", and Bayesian structural time series (BSTS) model including syndromic surveillance data were developed. These models were evaluated over weekly nowcasts using a probabilistic scoring framework. RESULTS: Using the

Norovirus GI.5 [P4]: first report of the rare norovirus recombinant variant in Northeastern Mexico and its global epidemiological context.

Norovirus is the primary cause of acute gastroenteritis outbreaks, considerably impacting children under 5 years, followed by older adults and immunocompromised individuals. As an RNA virus, norovirus exhibits high genetic variability, driven by recombination events at the ORF1-ORF2 junction. This study reports the first detection of the rare norovirus GI.5 [P4] variant in Northeastern Mexico, identified in a single positive isolate (MTY0115; GenBank: PQ369661) from a sample group of 386 individuals, with a prevalence of 0.25%. Notably, norovirus GII was not detected. Phylogenetic analysis of

Multi-Province Outbreak of Acute Gastroenteritis Linked to Potential Novel Lineage of GII.17 Norovirus in Argentina in 2024.

Noroviruses represent one of the leading causes of outbreaks and sporadic acute gastroenteritis (AGE) cases across all age groups. Although the GII.4 norovirus has been identified as the primary cause of most AGE outbreaks, the transient predominance of other genotypes has been reported globally. In this study, we describe a multi-province AGE outbreak caused by a potential new lineage of norovirus GII.17[P17], which has been recently detected at a high incidence in the United States and Europe. An amino acid analysis of the major viral capsid protein revealed several substitutions in the hype

Endoscopic findings among healthy adults with asymptomatic norovirus infection in Japan: A single-center cross-sectional study.

INTRODUCTION: Norovirus infection, a major cause of acute viral gastroenteritis, is commonly reported during the winter season. Norovirus invades the upper intestinal tract, causing inflammation. In severe cases, endoscopic findings and complications, including duodenal perforation, have been reported. However endoscopic findings in patients with asymptomatic infections have rarely been reported. Therefore, this study aimed to investigate the upper endoscopic findings of patients with asymptomatic norovirus infection. METHODS: Healthy adults (aged ≥18 years) who participated in voluntary heal

Proteomics as a complementary approach to measure norovirus infection in clinical samples.

Norovirus (NoV) is a leading cause of global acute gastroenteritis, particularly in young children, with no current licensed vaccine. Epidemiological studies have shown that asymptomatic cases are common, and infected patients may test positive for prolonged periods; however, the impact of these phenomena on transmission and public health measures remains unclear. A major limiting factor is our ability to measure infection, which is constrained to real-time reverse transcription polymerase chain reaction or antibody-based assays, both of which are susceptible to loss of detection by rapid NoV

Initial Establishment of Warning Model for Epidemic Intensity of Norovirus GII Associated with Acute Gastroenteritis in Beijing Based on Synthetic Index Method.

At present, there is no research that classifies the epidemic intensity of acute gastroenteritis (AGE) caused by Norovirus (NoV) GII combined cases and environmental surveillance data at the same time. With reference to the experience of the epidemiological-level classification of infectious disease and the actual epidemiological status of NoV AGE in Chaoyang District, Beijing, China, the epidemic intensity of NoV GII was divided into five grades with increasing intensity from grade 1 to grade 5, which corresponds to non-epidemic risk, general risk, moderate risk, high risk, and ultra-high ris

Seasonality of medically attended norovirus gastroenteritis and its association with climatic factors within an US integrated healthcare system, 2016-2019.

BACKGROUND: While acute gastroenteritis (AGE) occurs year-round, norovirus has a winter seasonality in the United States. OBJECTIVE: We analyzed norovirus seasonality within a US integrated healthcare delivery system from 2016-2019. METHODS: Electronic medical records were collected for acute gastroenteritis (AGE) encounters with specific ICD-9/10 codes or clinical stool testing. Norovirus percent positivity was calculated as the 8-week centered rolling average. Temperature and absolute humidity data were measured via weather station. The relationship between these factors and weekly norovir

Integrated in-silico design and in vivo validation of multi-epitope vaccines for norovirus.

BACKGROUND: Norovirus (NoVs) is a foodborne pathogen that causes acute gastroenteritis. The diversity of its principal antigenic protein poses a significant challenge to vaccine development and the prevention of large-scale outbreaks globally. Currently, no licensed vaccines against norovirus have been approved. METHODS: We developed a novel pipeline that integrates multiple bioinformatics tools to design broad-spectrum vaccines against NoVs. Specifically, broad-spectrum T-cell epitope vaccines were designed based on consensus sequences and optimized epitope screening, while broad-spectrum B-

Manipulation of immunodominant variable epitopes of norovirus capsid protein elicited cross-blocking antibodies to different GII.4 variants despite the low potency of the polyclonal sera.

Norovirus is a major cause of viral acute gastroenteritis. The predominant genotype, GII.4, presents a continuous emergence of new variants that could escape immunity developed in previous infections. The emergence of GII.4 variants is associated with multiple mutations on five antigenic sites (A, C, D, E, and G) of the major viral capsid protein. Antigenic site A includes most immunodominant epitopes, while antigenic site G shifted its immunodominance during the evolution of GII.4 noroviruses. This interrelation of immunodominance and high variability challenges the development of universal v

Integrating Enzyme-DNA Complex and CRISPR/Cas12a for Robust Norovirus Detection.

Human norovirus (NoV) is a primary cause of acute gastroenteritis in children, making accurate and rapid detection essential for effective disease prevention and control. In this study, we developed a sensitive and efficient platform for pathogen nucleic acid detection by integrating asymmetric nucleic acid sequence-based amplification (asymmetric NASBA), enzyme-DNA molecular complex, and the clustered regularly interspaced short palindromic repeats (CRISPR) system, namely an A-enDMC platform. The target recognition capability of the enzyme-DNA complex operates independently from the signal am

Adaptation and validation of a gastrointestinal panel to detect diarrheal virus pathogens on a high-throughput qPCR system.

With around 2 billion cases each year, infectious gastroenteritis remains a worldwide health problem. A major cause of acute gastroenteritis is infection with enteric viruses, which often leads to hospitalization in children and immunocompromised people. We adapted and validated a gastrointestinal qPCR panel, which simultaneously detects the most common enteric viruses: Norovirus GI and GII, Rotavirus, Adenovirus, Sapovirus, Astrovirus and Enterovirus in stool samples on a fully automated, high-throughput system (Roche cobas5800/6800/8800). Limits of detection (LOD), linear range and precision

The saga to monitor and control norovirus: the rise of GII.17.

Norovirus is a major cause of acute gastroenteritis in all age groups, with recent surges of cases in Europe and the USA reinforcing the influence of this virus on human health. Despite its societal impact, no vaccine or antiviral drug is available. The development of these countermeasures has been impaired at least in part by the extreme genetic and antigenic diversity of noroviruses. Here, we reviewed historical norovirus outbreaks, including the pandemics of GII.4 norovirus that were first documented in the mid-1990s, sporadic increases of non-GII.4 norovirus (e.g. GII.17 and GII.2) during
