Comparative genomic analysis of clinically relevant human skin-associated fungi.

Fungal skin infections represent a significant global health burden, affecting approximately one billion people annually. Despite their prevalence and major global health impact, the molecular mechanisms underlying pathogenicity remain largely uncharacterized. Here we present the genomes of 66 fungal strains, representing highly prevalent and clinically relevant species associated with human skin infections. Using comparative genomics, we investigate whether the taxonomic groups in this collection of skin fungi differ in their genome architecture, metabolic capacity, secreted enzyme repertoire

Malassezia isolated from patients with PV exhibits resistance features supporting its pathogenic nature.

Malassezia is a lipophilic yeast and a major commensal associated with the human skin and gut. It is responsible for several dermatological conditions and has been associated with human diseases, including certain cancers, yet the factors that trigger pathogenicity in this otherwise benign yeast remain poorly defined. Out of the 21 species of Malassezia that have been identified, 14 are associated with humans. In this study, we investigated how clinical strains of Malassezia furfur differ from the standard laboratory strain in terms of pathogenicity-associated elements and resistance features.

Pityriasis Versicolor in a Southern Ground Hornbill (Bucorvus leadbeateri).

A 20-year-old southern ground hornbill (Bucorvus leadbeateri) developed hyperpigmented macules on its gular pouch, resembling the lesions typically seen in humans with pityriasis versicolor. Cytologic examination revealed over 100 budding yeasts per oil immersion field. Fungal culture showed rare growth of smooth, cream-colored yeast colonies, identified by PCR as Malassezia slooffiae. Histopathological analysis showed lymphocytic perivascular dermatitis. Periodic acid-Schiff staining revealed rare forms of yeasts between the layers of the stratum corneum. This clinical report provides further

Managing Malassezia species and related infections: new insights into recent natural and synthetic antifungal compounds and their mechanism of action.

Malassezia spp. are lipophilic yeasts that colonize human and animal skin, being implicated in several dermatological and systemic disorders, including seborrheic dermatitis, pityriasis versicolor, and opportunistic fungemia in immunocompromised individuals. The increasing diffusion of resistant strains and the limited efficacy of conventional azole-based antifungal therapies have prompted the search for alternative treatment strategies. This review explores recent approaches to fighting Malassezia, focusing on both natural and synthetic compounds targeting key molecular pathways. Among the mo

Inverse Pityriasis Versicolor (PV).

Inverse pityriasis versicolor (PV) involving the groin and scrotum is an uncommon presentation of PV. Despite its uncommon location, PV should be suspected in patients with scaly and persistent hypo or hyperpigmented lesions, especially in regions with hot and humid environment. Early recognition allows effective antifungal therapy, preventing chronicity and recurrence.

Atrophying Pityriasis Versicolor: A Rare Presentation of a Common Dermatological Disorder and Literature Review.

Pityriasis versicolor is a superficial fungal infection of the skin caused by fungi of the genus Malassezia. Typically, patients present with well-defined, hypopigmented, scaly macules, or patches over seborrheic areas. However, rare presentations include papular, confetti-like spots, and folliculocentric, atrophic, and inverse forms. Diagnosis may be established clinically with the help of noninvasive diagnostic tools such as dermoscopy, Wood's lamp, and direct microscopic examinations. However, skin biopsies are imperative in equivocal cases. To date, 34 cases have been reported. This study

Erythematous pityriasis versicolor: a case report and review of the literature.

Pityriasis versicolor (PV) is a superficial fungal infection caused by Malassezia species, usually presenting as hypo- or hyperpigmented macules with fine scaling. We report a rare case of an erythematous variant of PV in a 28-year-old man, confirmed by potassium hydroxide examination of skin scrapings and direct microscopy. Awareness of such atypical variants is crucial in order for dermatologists and practitioners to avoid misdiagnosis and unwanted treatments. Our case also highlights the importance of considering these atypical variants in differential diagnosis of erythematous dermatoses.

Advancements in Novel Drug Delivery Systems for Topical Management of Superficial Fungal Infections: A Detailed Review.

INTRODUCTION: Superficial fungal infections, such as dermatophytosis, candidiasis, and pityriasis versicolor, are prevalent worldwide. They put a heavy burden on healthcare systems, mainly because infections often recur, linger for months, and do not always respond well to standard treatments. The conventional topical creams and ointments fail to achieve the goal of treatment as they are often unable to penetrate deeply, do not stay long enough on the site of application, and patients mostly stop using them before the completion of the course. This review explores the biology of superficial fu

Acquired epidermodysplasia verruciformis presenting as isolated genital hypopigmentation in a renal transplant recipient: a rare case report.

Epidermodysplasia verruciformis (EV) is a rare genodermatosis resulting from mutations in the EVER1/EVER2 genes, that increase vulnerability to beta-human papillomavirus (β-HPV) infection. The acquired form (AEV) develops in immunocompromised individuals, particularly organ transplant recipients, and typically manifests as pityriasis versicolor-like macules on sun-exposed areas. However, lesions confined to non-sun-exposed regions, especially the genitalia, are exceedingly uncommon and may be misdiagnosed. We report the case of a 38-year-old woman, nine years post-renal transplantation and on

Early-Onset Neonatal Pityriasis Versicolor Associated With Maternal Infection: A Case Report From Uganda.

We report a case of early-onset neonatal pityriasis versicolor (PV) in a term newborn associated with maternal infection. The mother had characteristic hypo- and hyperpigmented, finely scaly lesions over the neck and upper trunk prior to delivery. Within the first weeks of life, the neonate developed multiple hypopigmented, finely scaly macules involving the trunk and proximal thighs. The infant was otherwise clinically well, with no history of prematurity, neonatal intensive care unit admission, antibiotic exposure, or underlying medical illness. The clinical features were consistent with PV,

Clinicoepidemiological and Dermoscopic Patterns of Hypopigmented Disorders: A Cross-Sectional Observational Study.

Introduction Hypopigmented disorders comprise a heterogeneous group of dermatoses characterized by hypopigmented macules, patches, plaques, papules, and scaling lesions. These disorders often follow a chronic course and significantly affect patients' quality of life. Clinical differentiation may be challenging because of overlapping clinical presentations and atypical morphology. Dermoscopy serves as a non-invasive diagnostic tool that enhances visualization of submacroscopic morphological features and improves diagnostic accuracy. Materials and methods This cross-sectional observational study

The Vinyl Adhesive Tape Technique With Methylene Blue for Diagnosis of Superficial Fungal Skin Infections.

The Vinyl adhesive tape (VAT) technique with methylene blue (MTB) may be useful as an adjunctive microscopic method for diagnosis of superficial fungal skin infections. A cross-sectional study was conducted between 2022 and 2025 on adults with dermatophytosis, pityriasis versicolor, and cutaneous candidiasis confirmed by skin scraping with potassium hydroxide (KOH). VAT technique with MTB staining was performed at the same lesion. Data on clinical characteristics, hyphal index, and time to microscopic identification were recorded. The primary objective was the positivity rate of VAT technique

Translational evaluation of magnetized saline water combined with ciclopirox olamine and piroctone olamine against Malassezia species and scalp seborrheic dermatitis.

Yeasts of the Malassezia genus are implicated in numerous skin conditions, including seborrheic dermatitis, pityriasis versicolor, and folliculitis. While ciclopirox olamine (CPO) and piroctone olamine (OCT) are effective anti-Malassezia agents, the activity of magnetized water against pathogenic yeasts remains unclear. The aim of this study was to examine the in vitro efficacy of magnetized saline water (MSW), alone or combined with CPO plus OCT, against three Malassezia species (M. furfur, M. globosa, and M. restricta) using broth microdilution to determine minimal inhibitory concentrations
