MicroRNA-3619 is Significantly Increased During the Progression of Premature Ovarian Failure.

INTRODUCTION: Premature ovarian failure (POF) is a condition characterized by the onset of menopause before the age of 40. The progression of various disease stages, including diminished ovarian reserve (DOR) and POF, is regulated by microRNAs, which are small noncoding RNAs. This study aimed to compare the altered expression of miR-3619 during disease progression in healthy women, patients with DOR, and patients with POF. METHODS: First, bioinformatics analysis techniques were used to characterize and predict the target genes associated with miR-3619. Real-time PCR was then performed to eval

Stn1 promotes zebrafish oocyte development via amplifying Wnt/β-catenin signaling.

Wnt/β-catenin signaling regulates oocyte development in vertebrates. However, the dynamics of Wnt/β-catenin signaling in oocyte progression remain unclear. Here, we analyze oocytes across different developmental stages in zebrafish and find that the activity of Wnt/β-catenin signaling sharply increases when oocytes reach a size of 10 to 45 μm (stage IA to middle stage IB) and subsequently declines rapidly. In addition, we show that expression of the CST complex subunit Stn1 is enriched in germ cells. Stn1 interacts with the transcription factor Tcf/Lef, facilitates its association with promote

Impact of male genital tract infections on semen quality: a systematic review and meta-analysis.

Male genitourinary (GU) infections have been proposed to adversely affect semen quality and reproductive outcomes; however, existing evidence remains heterogeneous. This systematic review and meta-analysis evaluated the association between GU infections and semen parameters, reproductive outcomes, and assisted reproductive technology (ART) outcomes using control-comparator studies identified through PubMed, Web of Science, and Embase. Studies compared men with documented GU infections with noninfected controls and reported outcomes that were included consisted of semen volume, concentration, m

Loss of maternal PADI6 disrupts DNA methylation and genomic imprinting maintenance in late preimplantation mouse embryos.

BACKGROUND: The maternal-effect protein PADI6, which is part of the subcortical maternal complex, is involved in proper spindle assembly, organelle distribution, ribosome storage, and cytoplasmic lattice organization in mouse oocytes. In humans, variants of PADI6 are associated with female infertility and multilocus imprinting disturbance in offspring. Recently, it was demonstrated that PADI6 plays a role in the storage and cytoplasmic localization of epigenetic factors, including UHRF1 and DNMT1. Moreover, maternal PADI6 depletion leads to defective epigenetic reprogramming and zygotic genome

Oocyte gene mutations increase rates of total fertilization failure: a systematic review and meta-analysis.

INTRODUCTION: Total fertilization failure (TFF) remains a critical barrier in assisted reproductive technology (ART), occurring in 5–10% of treatment cycles. While TFF can result from both male and female gamete factors, emerging evidence suggests that oocyte gene mutations may play a significant role. This study systematically reviews and meta-analyzes the influence of specific oocyte gene mutations (OGMs) on fertilization outcomes, focusing on their association with TFF and fertilization rates (FRs). METHODS: This study has a preregistered protocol (CRD42024608566). A comprehensive search wa

Reproductive health - a blind spot in psychotherapeutic treatment? Evidence of insufficient consideration of reproductive factors in routine care.

INTRODUCTION: Reproductive health impacts mental health. Consequently, reproductive factors should be assessed in psychotherapy to ensure correct differential diagnosis, precise case conceptualisation and efficient treatment. It has not been analysed to which extent psychotherapists assess reproductive factors in routine care. METHODS: Analyses were based on data from Germany-wide surveys with n = 390 psychotherapists and n = 291 patients. From their respective perspective, we examined how frequently psychotherapists proactively assessed whether patients experienced a menstrual cycle, menopau

Male- and female-factor infertility and partners' quality of life during assisted reproductive technology: A dyadic actor-partner analysis.

BACKGROUND: Infertility constitutes a dyadic stressor, as it inherently involves both members of a couple; nevertheless, its psychological impact is frequently evaluated from an individual perspective. We tested whether etiologic diagnosis (male and female factors) is associated with both partners' quality of life (QoL) during assisted reproductive technology (ART). METHODS: We conducted a retrospective cross-sectional study at the assisted reproduction center of a tertiary university hospital in southern Taiwan, enrolling married heterosexual couples undergoing ART who completed the Fertilit

Haematopoietic loss of the X chromosome is associated with a lower likelihood of natural conception.

RESEARCH QUESTION: Is haematopoietic loss of the X chromosome (LOX), which increases with age in women, but whose reproductive relevance is unclear, associated with the likelihood of natural conception? DESIGN: Multicentre case-control study at three hospitals in Osaka, Japan. Cases were women who did not achieve natural conception after exclusion of male-factor infertility (n = 381). Controls were women who conceived naturally (n = 123). The LOX burden in peripheral blood (%LOX) was quantified by multiplex single-cell droplet digital polymerase chain reaction. Serum anti-Müllerian hormone (A

Evaluating Sperm DNA Damage: When and Why It Does Not Add to the Evaluation of Male Factor Infertility.

Male infertility accounts for nearly half of all infertility cases, yet standard semen analysis often fails to reflect sperm's functional competence or fertilization potential. Sperm DNA fragmentation (SDF) has emerged as a potential biomarker of sperm quality, with elevated levels frequently associated with reduced fertilization rates, lower IVF/ICSI success, and an increased risk of recurrent pregnancy loss (RPL). Meta-analyses suggest an inverse relationship between SDF and both clinical pregnancy and live birth rates, particularly in IVF/ICSI cycles; however, assay variability limits the i

The paradoxical association between endometrioma size and serum anti-Müllerian hormone levels in infertile women.

The impact of endometrioma size on ovarian reserve remains controversial. Although endometriomas are known to reduce anti-Müllerian hormone (AMH) levels, some studies suggest a paradoxical association between larger cysts and higher AMH. This cross-sectional study enrolled 210 infertile women between January 2023 and March 2025: 150 with laparoscopy-confirmed untreated unilateral endometriomas and 60 controls with male-factor infertility and normal ovaries. Serum AMH, follicle-stimulating hormone, luteinizing hormone, and prolactin were measured on cycle day 3. Endometrioma diameter was measur

Clinical use of recombinant human luteinizing hormone beyond hypogonadotropic hypogonadism: a multicenter descriptive analysis from five Italian fertility centers.

BACKGROUND: Recombinant human luteinizing hormone (rLH) was originally introduced to supplement recombinant human follicle-stimulating hormone (rFSH) for controlled ovarian stimulation (COS) in patients with hypogonadotropic hypogonadism. Promising evidence is emerging regarding the efficacy of rLH in conditions which mimic hypogonadotropic hypogonadism; however, current data remain insufficient to draw definitive conclusions. The aim of the present multicentre observational study was to characterize the in vitro fertilization (IVF) population in which a fixed combination of rFSH/rLH is prescr

The role of satellite DNA-enriched heterochromatic variants in reproductive disorders: Insights from standardized cytogenetic analysis.

Non-coding DNA, long considered "junk", is now recognized as a central regulator of genome architecture. Highly repetitive satellite DNA sequences shape heterochromatin and are essential for chromosome stability, segregation, and gene regulation. Pericentromeric heterochromatic variants, or chromosomal heteromorphisms (CHs), have emerged as modulators of human fertility, potentially affecting gametogenesis and early embryonic development. Despite their ubiquity, the functional and clinical significance of CHs remains largely enigmatic. Molecular reference genomes fail to fully capture these re

Sperm DNA Fragmentation Testing in the Era of Assisted Reproduction: Clinical Utility, Thresholds, and Management Strategies.

Sperm DNA fragmentation (SDF) has emerged as a clinically important biomarker in male infertility evaluation, offering diagnostic information beyond conventional semen analysis. The inclusion of SDF testing into the World Health Organization 6th edition laboratory manual underscores its growing relevance in reproductive medicine. Elevated SDF levels have been associated with reduced fertilization rates, impaired embryo quality, decreased pregnancy rates, and increased miscarriage risk, particularly in couples with otherwise normal semen parameters. This narrative review examines current SDF te

Testicular sperm aspiration enhances fertilization rates in MMAF patients undergoing ICSI: a retrospective cohort study.

Multiple morphological abnormalities of the sperm flagella (MMAF) compromise intracytoplasmic sperm injection (ICSI) outcomes. Testicular sperm aspiration (TESA) may be beneficial, but its impact on early embryonic development and pregnancy in MMAF remains uncertain. This study evaluated the effect of TESA on fertilization, cleavage, on-time Day-3 embryo development, and clinical pregnancy and explored modification by female age and MMAF genotype. In this retrospective cohort, 196 MMAF patients undergoing ICSI (January 2019-December 2024) without female-factor infertility were analyzed: TESA g

Attitudes of healthcare providers towards fertility preservation in Portugal: perceived support across indications and their ethical implications.

BACKGROUND: Healthcare providers (HCPs) play a pivotal role in shaping access to and ethical deliberation around fertility preservation. While fertility preservation due to oncological and non-oncological diseases is widely accepted, its use for other reasons, such as career development, gender transition, or military deployment, raises ethical and professional questions. This study aimed to explore the attitudes of HCPs in Portugal towards fertility preservation across different indications and to identify professional factors associated with variations in perceived support. METHODS: We cond

Seasonal and geographic patterns of gastroschisis in Canada: protective effect of periconceptional sunlight exposure.

BACKGROUND: Gastroschisis is a right-sided, full-thickness defect with extra-amnionic herniation of abdominal contents directly into the amniotic cavity. The causes of gastroschisis remain obscure. We hypothesized that limited periconceptional sunlight exposure might lead to insufficient vitamin D during fetal development and play a role in gastroschisis etiology. METHODS: We conducted a population-based cohort study of infants conceived in Canada between April 2006 and March 2020. We used winter months (i.e., November and December) and northern geographic latitudes as indicators of insuffici
