Comparative genomic analysis of clinically relevant human skin-associated fungi.

Fungal skin infections represent a significant global health burden, affecting approximately one billion people annually. Despite their prevalence and major global health impact, the molecular mechanisms underlying pathogenicity remain largely uncharacterized. Here we present the genomes of 66 fungal strains, representing highly prevalent and clinically relevant species associated with human skin infections. Using comparative genomics, we investigate whether the taxonomic groups in this collection of skin fungi differ in their genome architecture, metabolic capacity, secreted enzyme repertoire

Comparative genomic analysis of clinically relevant human skin-associated fungi.

Fungal skin infections represent a significant global health burden, affecting approximately one billion people annually. Despite their prevalence and major global health impact, the molecular mechanisms underlying pathogenicity remain largely uncharacterized. Here we present high quality genomic datasets for 51 fungal strains, representing highly prevalent and clinically relevant species associated with human skin infections. Comparative genomics reveal substantial variation in genome size and gene contents, indicating genome compaction occurred as the fungi transitioned from free-living to h

Trichosporon and Antifungal Resistance: Current Knowledge and Gaps.

Trichosporon spp. are ubiquitous environmental and emerging opportunistic fungi that can be part of the normal microbiota of the skin, vagina, and gastrointestinal tract in humans and other animals. In a state of microbiota imbalance, both immunocompromised and non-immunocompromised patients are susceptible to trichosporonosis, which can present as either superficial infections, such as White Piedra, or deep-seated infections, such as fungemia. Due to intrinsic resistance or limited efficacy reported with fluconazole, polyenes, echinocandins, and flucytosine, voriconazole is currently recommen

White piedra: fungal extracellular matrix formation and its importance in pathogenesis. An ultrastructural study.

BACKGROUND: White piedra is a disease caused by some species of the genus Trichosporon. A case of white piedra was investigated, whose molecular examination identified Cutaneotrichosporon (Trichosporon) debeurmannianum as the causative agent. METHODS: Scanning electron microscopy (SEM) was used to examine the affected hairs, as well as the fungal colony of C. debeurmannianum obtained from the hairs. For comparative purposes, a colony of Trichosporon mucoides obtained from a mycotheque was also examined. RESULTS: Examination of the affected hairs using SEM easily demonstrates nodules on the h

A rare case of white piedra caused by Candida orthopsilosis.

OBJECTIVES: White piedra (WP) is a fungal infection that affects hair shafts, resulting in the formation of soft nodules that can be white, gray, or brown. While it was initially believed to be caused by Trichosporon beigelii, genetic analysis has reclassified Trichosporon inkin, T. ovoides, and T. cutaneum, as the primary causative agents of WP. In this report, we present a rare documented case of WP caused by Candida orthopsilosis, as a member of Candida parapsilosis complex, occurring without presence of Trichosporon in a temperate region in northern Iran. CASE PRESENTATION: A 3-year-old g

The Answer Is Blowin' in the Wind: Styling Trauma of the Hair Shaft as a Piedra Mimic.

Hair shaft disorders/abnormalities are usually easily diagnosed with the help of in-office trichoscopy and, if necessary, electron microscopy. Here we present a distinctive case of damaged, stripped hair cuticles clinically mimicking white piedra in a patient with a history of weekly hair blowout treatments. We demonstrate that staining sampled hair shafts with Periodic Acid-Schiff stain (PAS) and Gomori Methamine Silver stain (GMS) stains can be helpful to highlight cuticle fraying in instances where other explanations for these pseudonits have been ruled out. These "balled up" cuticle fibers

Adhesion and biofilm formation by two clinical isolates of Trichosporon Cutaneum in various environmental conditions.

Trichosporon spp. is an emerging opportunistic pathogen and a common cause of both superficial and invasive infections. Although Trichosporon asahii is the most frequently isolated species, Trichosporon cutaneum is also widely observed, as it is the predominant agent in cases of white Piedra and onychomycosis. Trichosporon spp. is a known to produce biofilms, which serve as one of its virulence mechanisms, however, there is limited data available on biofilms formed by T. cutaneum. Thus, the aim of this study was to assess the adhesion and biofilm formation of two clinical isolates of T. cutane

White piedra on pediatric scalp: A case report.

This case report describes a rare fungal infection, piedra alba, in a 5-year-old female initially misdiagnosed. Treatment with 2 % ketoconazole shampoo led to significant regression within a week, without the need for hair cutting. We discuss the importance of early and accurate diagnosis, highlighting potential hair damage and complications in immunocompromised cases. Dermatoscopy aided diagnosis, and 2 % ketoconazole demonstrated efficacy, emphasizing the need for a multidisciplinary approach and dermatological follow-up.

Identification of Trichosporon yeast isolates from superficial infections in male patients from Central Brazil: an approach to the diversity of infections caused by this basidiomycete fungus.

The genus Trichosporon are currently recognized as opportunistic pathogens capable of causing superficial "white piedra" infections and potentially fatal invasive diseases (Trichosporonosis). In this work, determine the agent Trichosporon spp. isolated from the skin and appendages of a male population group in the Central-West region of Brazil. The isolates were analyzed by phenotypic, biochemical and molecular methods. Twenty-five strains of Trichosporon were isolated: T. asahii (18; 72%), followed by T. inkin (4; 16%) and T. faecale (3; 12%). Skin infections were the most affected (16; 64%)

First report of white piedra caused by Cutaneotrichosporon debeurmannianum.

Piedras are small nodules found on the hair shafts. White piedra was initially described as caused by Trichosporon beigelii, which was later reclassified in several species. We describe the first case of white piedra caused by Cutaneotrichosporon debeurmannianum. Affected hairs were examined in natura with scanning electron microscopy, after gold metallization. The typical whitish cerebriform creamy colony was obtained in Sabouraud medium. Fungal genomic DNA extracted from cultures and locus was amplified with the NL1/NL4 primer pair from the D1/D2 region of the large ribosomal subunit (LSU) o

Antifungal susceptibility profile of Trichosporon inkin: About three cases of White Piedra.

Trichosporon spp. usually cause systemic or superficial infections. Three cases of White Piedra produced by Trichosporon inkin are described. The in vitro antifungal activity to fluconazole, amphotericin B, ketoconazole and caspofungin against the three clinical isolates were evaluated. Sensitivity to fluconazole and ketoconazole was evidenced. However, the treatment of this mycosis is still a challenge.

Review Article White Piedra: Review of 131 cases.

This study analyzes the clinical characteristics of patients diagnosed with White Piedra through a systematic review of cases in the literature. A sample of 131 subjects was considered, of which 91.6% were female and most were 18 years of age or younger. Most studies were conducted in Brazil, followed by India, and Mexico. The most common etiologic agent found was Trichosporon spp (34.3%). Most affected patients were asymptomatic (94.6%) and predisposing factors included long hair, use of a hair band or hair accessories, and wet hair. The most common clinical feature was the presence of nodule

Molecular identification of yeasts from the order Trichosporonales causing superficial infections.

BACKGROUND: The molecular reclassification of the order Trichosporonales placed the medically relevant Trichosporon species into three genera of the family Trichosporonaceae: Cutaneotrichosporon, Trichosporon, and Apiotrichum. From the clinical and epidemiological standpoint, it is important to identify any species of the family Trichosporonaceae because they present different antifungal susceptibility profiles. In Mexico, little is known about trichosporonosis etiology because the fungi are identified through phenotypic methods. AIMS: To identify at a molecular level 12 yeast isolates morfol

Multidrug-resistant Trichosporon species: underestimated fungal pathogens posing imminent threats in clinical settings.

Species of Trichosporon and related genera are widely used in biotechnology and, hence, many species have their genome sequenced. Importantly, yeasts of the genus Trichosporon have been increasingly identified as a cause of life-threatening invasive trichosporonosis (IT) in humans and are associated with an exceptionally high mortality rate. Trichosporon spp. are intrinsically resistant to frontline antifungal agents, which accounts for numerous reports of therapeutic failure when echinocandins are used to treat IT. Moreover, these fungi have low sensitivity to polyenes and azoles and, therefo
