Gallbladder Cancer: Epidemiology and Prevention.

Gallbladder cancer (GBC) is an uncommon but highly lethal malignancy, characterized by striking geographic clustering, late diagnosis, and a prevention profile that differs from other gastrointestinal malignancies. This narrative review synthesizes epidemiologic, mechanistic, clinical, and public-health evidence on GBC determinants with a prevention-oriented lens. Rather than merely cataloguing risk factors, we propose a pragmatic framework that combines epidemiologic credibility, population-attributable burden, baseline risk, biologic plausibility, feasibility, and the benefit-risk balance of

Strengthened Anti-Inflammatory Potential of a Purified Antimicrobial Peptide Extract in Leukocyte-Biomaterial Interactions.

Excessive inflammation following implant insertion may impair early healing and predispose to postoperative complications. Since neutrophils constitute the predominant leukocyte population during the initial inflammatory phase, and monocytes subsequently orchestrate tissue resolution and repair, understanding their direct response to biomaterials and bioactive compounds is essential. In the present study, we evaluated the effect of purified bioactive extract combined with biomaterial on neutrophil and monocyte activity in vitro, comparing their modulatory potential with that of non-purified ex

Early blood pressure outcomes after SPYRAL™ renal denervation: an exploratory small cohort analysis.

BACKGROUND: Catheter-based renal denervation (RDN) is an interventional therapy for patients with resistant or uncontrolled arterial hypertension after the exclusion of secondary causes. This study aimed to evaluate the effects of the SPYRAL™ catheter system on systolic (SBP) and diastolic blood pressure (DBP) in a real-world clinical setting. Additionally, we explored whether the number of ablation points was associated with the magnitude of blood pressure (BP) reduction. METHODS: We conducted a retrospective, single-center study including patients aged ≥18 years with predominantly resistant

MSC-Derived Small Extracellular Vesicles Alleviate Renal Fibrosis in Hyperuricemic Nephropathy via Suppression of Epithelial Mesenchymal Transition.

BACKGROUND: Hyperuricemic nephropathy (HN) arises from excessive accumulation of uric acid, leading to subsequent renal injury. Mesenchymal stem cells-derived small extracellular vesicles (MSC-sEVs) have demonstrated beneficial effects in promoting tissue repair across various injury models. However, their potential therapeutic role in HN remains unknown. METHODS: We isolated and characterized MSC-sEVs by transmission electron microscopy, nanoparticle tracking analysis, and Western blotting. A rat model of HN was established by daily administration of adenine and potassium oxonate for three w

Dysregulated complement activation in COVID-19: The Role of Soluble Complement Inhibitors.

In viral infections, complement activation can contribute to viral clearance, but excessive activation, as observed in severe COVID-19, may drive immunopathology. While much is known about complement-activating factors in COVID-19, insight into the role of endogenous complement inhibitors remains limited. We performed a longitudinal analysis within the BEAT-COVID cohort, comprising hospitalized patients with SARS-CoV-2 infection admitted to the intensive care unit (ICU) or ward, and age-matched healthy controls. Serum levels of the complement activation marker soluble membrane attack complex (

HSP70-Mediated Macrophage-Stromal Crosstalk is an Immunopathogenic Driver of Recurrent Implantation Failure.

BACKGROUND: Recurrent implantation failure (RIF) remains a major challenge in assisted reproductive technology (ART), affecting a considerable proportion of patients despite multiple transfers of high-quality embryos. Although impaired endometrial receptivity is recognized as a critical contributor to RIF, the molecular mechanisms responsible for establishing a non-receptive endometrial microenvironment remain incompletely understood. The proliferative phase precedes decidualization and subsequent embryo implantation, representing a critical period for endometrial preparation. The endometrium

Targeted intestinal ROS scavenging alleviated rheumatoid arthritis by microbial metabolite kynurenic acid mediated Treg/Th17 balance through AhR pathway activation.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by immune dysregulation, which is often closely related to impaired gut microbiota homeostasis. Excessive reactive oxygen species (ROS) are recognized as critical drivers of microecological immunity disturbance, contributing directly to the inflammatory cascade. However, the underlying mechanism remains unclear. Here, we have used ROS scavengers to alleviate RA progression via inhibiting intestinal ROS and modulating gut microbial kynurenic acid metabolism. Black phosphorus nanosheets (BPNSs), as effective ROS scavengers,

Donor-source-specific competing risks of relapse and non-relapse mortality in adult B-ALL allogeneic hematopoietic stem cell transplantation on a rabbit antithymocyte globulin-based platform.

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a cornerstone of therapy for adult B-cell acute lymphoblastic leukemia (B-ALL), yet simultaneous comparison of all four major donor categories within a single transplant platform is scarce. We retrospectively analyzed 190 consecutive adults with B-ALL undergoing first allo-HSCT (2008-2025) at a single center on a predominantly rabbit antithymocyte globulin (rATG)-based myeloablative platform: matched sibling (MSD, n = 72), matched unrelated (MUD, n = 46), mismatched unrelated (MMUD, n = 40), and haploidentical (Haplo, n = 32) do

Mitochondrial dysfunction and PANoptosis-related signaling in acute central nervous system injuries: mechanistic convergence and therapeutic opportunities.

Acute central nervous system (CNS) injuries comprise a heterogeneous group of disorders caused by vascular or traumatic factors, including ischemic stroke, intracerebral hemorrhage (ICH), traumatic brain injury (TBI), and spinal cord injury (SCI). These injuries initiate secondary cascades that combine metabolic failure, oxidative stress, neuroinflammation, and regulated cell death. Mitochondrial dysfunction is positioned near the center of these processes because impaired bioenergetics, excessive mitochondrial reactive oxygen species (mtROS), abnormal fission and fusion, defective mitophagy,

Integrated Viscoelastic Microfluidics for Label-Free Separation and Concentration of Circulating Tumor Cells.

Developing advanced instrumentation systems for the automated, precise isolation of target circulating tumor cells (CTCs) from blood, followed by molecular marker analysis, stands as a pivotal trend in the field of liquid biopsy. However, current isolation technologies typically rely on subsequent centrifugation steps to enrich CTC samples for downstream assays, such as single-cell western blotting (scWB), creating challenges for developing integrated systems. Here, we present an integrated viscoelastic microfluidic system (IVEM) that leverages viscoelastic fluids to automatically separate and

Lower circulating angiotensinogen levels across ACLD stages are associated with hepatic dysfunction and mortality.

INTRODUCTION: The liver produces angiotensinogen (AGT), the key upstream precursor of the renin-angiotensin system (RAS). We measured AGT levels in patients across distinct advanced chronic liver disease (ACLD) stages and investigated the impact of AGT levels on clinical outcomes. METHODS: We included clinically stable ACLD outpatients undergoing hepatic venous pressure gradient (HVPG) measurement between May 2018 and October 2023, as well as healthy controls (HCs) recruited in the prospective Vienna Cirrhosis Study (VICIS; NCT03267615). AGT was assessed by converting it to angiotensin I (Ang

The Role of Neutrophil Extracellular Traps (NETosis) in Systemic Lupus Erythematosus: Molecular Mechanisms, Pathogenic Roles, Diagnostic Biomarkers, and Emerging Therapeutic Strategies.

Neutrophil extracellular traps (NETs) are web-like structures comprising chromatin scaffolds decorated with cytotoxic granule proteins released during a specialized form of neutrophil cell death termed NETosis. Although NETs function as a critical antimicrobial defense mechanism, their dysregulation contributes significantly to the pathogenesis of systemic lupus erythematosus (SLE). In SLE patients, both excessive NET formation by proinflammatory low-density granulocytes (LDGs) and impaired NET clearance due to reduced or acquired DNase1/DNase1L3 activity create a pathogenic cycle that amplifi

Polystyrene nanoplastics induced toxicity in human endothelial cells via atherogenic and autophagy pathways.

BACKGROUND: Polystyrene nanoplastics (PS NPs) pollution poses an emerging threat to the ecosystem and human health owing to their physicochemical properties. Excessive exposure to PS NPs can disrupt the cardiovascular system, especially the function of endothelial cells (ECs). Given the potential pathological significance of particles, the present study aimed to investigate the cytotoxic effects of PS NPs on ECs in in vitro and ex ovo models, as well as their underlying mechanisms. METHODS: PS NPs were characterized using DLS, SEM, and FTIR. Human umbilical vein ECs (HUVECs) were first treate

Iron Supplementation and Copper Homeostasis: Evidence for Potential Copper Depletion, Biomarker Challenges, and Clinical Implications.

Iron deficiency remains a major nutritional problem, particularly among women of reproductive age, pregnant women, infants, and children. Oral iron supplementation is the principal strategy for prevention and treatment, but the biological interdependence of iron and copper metabolism raises concern that prolonged or excessive iron exposure may impair copper availability in susceptible individuals. This review summarizes human and animal evidence linking iron supplementation or high dietary iron intake to altered copper homeostasis, with emphasis on underlying mechanisms, biomarker limitations,

Punicalagin Alleviates Diabetic Kidney Injury: Potential Involvement of SESN2-Related Mitophagy and Changes in Pyroptosis-/Ferroptosis-Related Indicators.

BACKGROUND/OBJECTIVES: Diabetic kidney disease (DKD) is a progressive nephropathy associated with excessive pyroptosis and ferroptosis. Impaired renal mitophagy has been implicated in inflammation and lipid peroxidation in DKD; however, it remains unclear whether and how it modulates pyroptosis and ferroptosis. Punicalagin (PU), a natural polyphenol with potent anti-inflammatory and antioxidant activities, has been shown to improve DKD, yet its underlying mechanism requires further elucidation. This study aimed to investigate whether PU alleviates pyroptosis- and ferroptosis-related indicators

Lipid Droplets in Poultry Lipid Metabolism: Characteristics, Regulatory Factors, and Implications for Fatty Liver and Muscle Lipid Deposition.

Lipid droplets (LDs) are dynamic metabolic platforms that integrate neutral-lipid storage, fatty-acid trafficking, organelle communication, cellular energy metabolism, and stress adaptation in poultry cells. Lipid metabolism plays a central role in poultry growth, reproductive function, production performance, meat quality, and metabolic health. Unlike mammals, de novo lipogenesis in poultry occurs predominantly in the liver, making hepatic lipid homeostasis particularly important for maintaining physiological function and production efficiency. Dysregulation of lipid metabolism contributes to
