Adaptive drug resistance in melanoma is characterised by lipidome remodelling while maintaining cell membrane biophysical properties.

Aim: Cancer drug resistance is driven by dynamic, systems-level adaptations, including cellular state transitions, epigenetic reprogramming, metabolic rewiring, and proteome remodelling. Lipid metabolic reprogramming is increasingly recognised as a contributor to resistance through alterations in the membrane composition and has been hypothesised to promote resistance by increasing membrane rigidity and limiting drug permeability. Here we tested this hypothesis in melanoma using a cell state-resolved multi-omics approach. Methods: We combined lipidomics, surface-resolved phospholipid profiling

Centering Patient Health Over Legal Status: A Student-Driven Cannabis Risk Stratification Framework for Undergraduate Medical Education.

BACKGROUND: Cannabis use is increasing worldwide, both for evidence-supported indications and conditions with limited evidence such as anxiety, depression, and chronic pain. Although incongruence between state and federal cannabis laws has delayed expanding cannabis education within medical schools, patients will continue to use cannabis and physicians must be prepared to counsel them. Therefore, evidence-informed cannabis education is needed to prepare medical students to discuss cannabis use using a patient-centered and harm reduction-oriented framework rather than relying on legal status al

Sex, BMI, skeletal maturation, and height among children and adolescents in high-altitude Xizang: a cross-sectional study.

BACKGROUND: Skeletal maturation influences the interpretation of childhood height, but its relationship with sex, body mass index (BMI), and attained height in children living at high altitude remains incompletely characterized. We examined concurrent associations among sex, BMI, bone-age deviation, and height relative to national reference values in children and adolescents living in Lhasa, Xizang. METHODS: This cross-sectional study included 199 participants aged 3-16 years (101 girls and 98 boys). Bone-age deviation was defined as bone age minus chronological age. Height difference and BMI

One- and two-hour storage delays improve nutritional components in Sarcocornia fruticosa ecotypes, but not in the Arthrocaulon macrostachyum ecotype.

The effects of postharvest storage delays of 1 and 2 hours on the nutritional quality of halophytic plants irrigated with 100 and 150 mM NaCl solutions were assessed. The study compared the new Sarcocornia fruticosa ecotypes Shikmona, Megadim, and Ruhama with the commonly cultivated VM and Arthrocaulon macrostachyum (AM). All new ecotypes outperformed the VM ecotype in terms of nutritional composition and yield. Except for Shikmona, the biomass of all ecotypes increased with elevated salinity; however, storage delays reduced water content, with a greater reduction under lower salinity. Storage

Online Academic Cheating in Young Children and its Prevention.

The global social situation changed dramatically after the COVID-19 pandemic, with various activities being shifted online. Some academic activities, including those for young children, are still conducted virtually. The transition introduced various challenges regarding academic honestly. However, whether young children engage in more academic cheating in online (vs. offline) settings remains unclear. Accordingly, this study examines this aspect, as well as how such behavior can be mitigated. We conducted two experimental studies with five-year-olds (N = 148), using a between-subjects design

First-principles description of pumped inelastic x-ray scattering: Example of K-edge RIXS in graphite.

We present an ab initio framework for predicting resonant inelastic x-ray scattering (RIXS) in optically pumped materials. Our methodology is based on the Kramers-Heisenberg formula for the double-differential cross section formulated using the results of the Bethe-Salpeter equation from many-body perturbation theory. To extend this approach to the time domain, we incorporate non-equilibrium charge-carrier distributions obtained from real-time, time-dependent density-functional theory. Generalizing the RIXS implementation with respect to arbitrary polarizations allows us to consider different

A MADS51 allele is associated with flowering-time divergence and contributes to ecological adaptation during wild rice speciation.

Flowering-time divergence can contribute to ecological speciation, but causal genes and mutations remain largely unknown in wild plants. Here, we investigate flowering-time divergence between the wild rice species Oryza rufipogon and O. nivara. By establishing a transformation system for wild rice and combining genome scans, population genomics, and functional assays, we identify MADS51 as a contributor to flowering-time divergence. Knockout of MADS51 in O. rufipogon accelerates flowering, whereas overexpression delays flowering. A functional haplotype (functional haplotype 2 [FH2]), defined b

Optogenetic reduction of theta oscillations induces remapping of time cells without memory impairment.

In a delayed alternation spatial working memory task, hippocampal time cells fire during specific moments of the delay period to form a sequential representation of the entire delay interval. The causal relationship between these sequences and working memory remains unclear. Similarly, hippocampal theta oscillations are thought to support working memory, primarily through the generation of time cell sequences. To causally examine these relationships, we optogenetically silence the medial septal GABAergic theta-generating circuit during the delay portion of a delayed spatial alternation task. W

Effect of randomness and delay in extracellular glucose transportation into cardiac contractility under diabetic conditions.

The present in-silico study incorporates a stochastic framework for GLUT4 recruitment to examine the coupled dynamics of glucose transport and myocardial contractile function. In their previous study, Das et al. (2020) developed a glucose-insulin interaction model incorporating a transportation delay associated with GLUT4-mediated glucose uptake. However, prior evidence suggests that GLUT4 recruitment exhibits stochastic behavior. Accordingly, the present study extends the model by introducing stochastic perturbations into GLUT4-mediated glucose transport. The results indicate that stochastici

Safety and Performance Evaluation of a Clinical Decision Support System for Imatinib Therapeutic Drug Monitoring: In Silico Study.

BACKGROUND: Therapeutic drug monitoring (TDM) aims to optimize clinical outcomes but is hindered by logistical delays and susceptibility to data errors. Integrating model-informed precision dosing with point-of-care testing, fortified by rule-based validation and alerts, forms a transformative clinical decision support system (CDSS) essential for securing this complex workflow. OBJECTIVE: This study evaluates, in silico, the effectiveness of a rule-based CDSS for imatinib in detecting data-quality issues and optimizing therapeutic dosing, compared with a human TDM expert. METHODS: We simulat

Delayed school start-times and adolescent sleep duration: A systematic umbrella review.

Expert panels have long recommended delayed school start-times (SSTs) as necessary for addressing widespread adolescent sleep loss; nevertheless, adoption of recommendations has been slow. The major goals of this umbrella review were to explore the consistency of review teams' recommendations while describing and assessing review quality. We used systematic procedures to identify five meta-analyses and four narrative reviews. Authors of all nine reviews concluded that adolescent sleep duration (ASD) likely increased when SSTs were later but usually recommended limited adoption. Reviewers' hesi

Demand for de novo serine biosynthesis in C. elegans embryos is buffered by the glycine cleavage system and maternal diet.

Metabolic network plasticity helps organisms survive change, including dietary fluctuations, transitions between developmental stages and genetic perturbations. To maintain a robust supply of resources under these conditions, metabolic networks must rebalance flux across individual pathways. However, how pathways coordinate their activity to support homeostasis remains poorly understood. Here, we use metabolomics, dietary supplementation, and genetic screening to investigate how the network compensates for the loss of phosphoglycerate dehydrogenase PHDH-1, the rate-limiting enzyme in serine bi

Associations between comorbidities and survival outcomes in older adults with cancer: a scoping review of international and Japanese evidence.

BACKGROUND: Advances in cancer treatment have improved survival among older adults with cancer, many of whom experience multiple comorbidities. This scoping review synthesized international and Japanese evidence regarding associations between comorbidities and survival outcomes in older adults with cancer. METHODS: We conducted a two-stage scoping review using PubMed and Ichushi. In the primary analysis, we searched for large-scale population- or registry-based studies globally to examine whether the pathways through which comorbidities influence survival outcomes in older adults with cancer,

Healthcare Inequities and Structural Racism as Determinants of Treatment and Survival among Individuals Recently Diagnosed with Cervical Cancer from Two Diverse Population-Based Cancer Registries: The ACHIEVE Study.

Persistent inequities in cervical cancer burden disproportionately impact marginalized communities and reflect social and structural barriers to high-quality care. Guided by socioecological models, the Assessing Cervical Cancer Healthcare Inequities in Diverse Populations (ACHIEVE) Study examines social and structural influences on cancer treatment among survivors (aged 21-79 years, diagnosed with any stage of cervical cancer between 2021 and 2025, who can complete study surveys in English, Spanish, Chinese, or Haitian Creole) identified through two population-based cancer registries: the New

Peripheral Nerve Stimulation Reduces Opioid Use and Facilitates Early Ambulation Following Fibular Flap Harvest: A Retrospective Study.

BACKGROUND: Fibular free flap (FFF) reconstruction is the gold standard for mandibular and maxillary defects, but donor-site pain remains challenging. Multimodal analgesia (MMA) relies on opioids, which risk dependence, systemic side effects, and delayed recovery. Regional nerve blocks reduce pain but have inconsistent efficacy owing to variable drug metabolism, potential effects on flap perfusion, and motor blockade. Temporary percutaneous peripheral nerve stimulation (TPNS) is a promising alternative that selectively modulates sensory pathways while preserving motor function. METHODS: This

Advance care planning in people with intellectual disabilities, more than 'to resuscitate or not to resuscitate': A qualitative interview study with general practitioners and family caregivers.

BACKGROUND: Advance care planning for people (ACP) with intellectual disabilities requires timely and carefully communication between family caregivers and general practitioners. OBJECTIVES: Despite its recognised importance, little is known about how these stakeholders experience advance care planning and decision-making processes, particularly regarding goals of care and do not resuscitate (DNR) orders, highlighting a critical gap in ensuring person-centred palliative care for this population. METHODS: A qualitative design was used to investigate how Belgian general practitioners and famil

A Bridge to Comfort: Nurses Driving Early Palliative Consults in the Intensive Care Unit.

This project established a protocol that enabled all intensive care unit (ICU) patients to receive a palliative care (PC) screening. Those who met criteria received timely access to PC in the ICU setting. Delayed access to PC can result in untreated symptoms, lack of understanding of care preferences, unwanted invasive procedures, and underutilization of hospice care. A Plan-Do-Study-Act framework was utilized to establish this protocol. The multidisciplinary team sought to increase PC consults in the ICU by 30% using a nurse-driven standardized screening tool. The PC Referral Criteria establi

ALYREF binds to poly-ADP-ribose at double strand breaks impairing repair in tumoral cells.

DNA-RNA hybrids cause genome instability by promoting transcription-replication conflicts. However, as they are stimulated at double-stranded breaks (DSBs), they can also impact DSB repair. We investigated whether the RNA chaperon ALYREF and its RNA helicase partner UAP56/DDX39B, both of which regulate RNA biogenesis and R-loops, also act at DSBs. We found that ALYREF, but not UAP56/DDX39B, is recruited to DSBs. Recruitment is partially dependent on transcription and hybrids, and fully dependent on polyADP-ribose (PAR). Consistently, we show that ALYREF, but not UAP56/DDX39B, binds PAR in vitr
