Elevated glucose levels trigger trophoblast cell invasion dysfunction through enhanced senescence and IGFBP4 signaling pathway.

Gestational diabetes mellitus (GDM) is related to multiple negative pregnancy outcomes such as miscarriage, pre-eclampsia and so on. Cell senescence was considered potential mechanism of GDM. However, the role and mechanism of high glucose and cellular senescence in trophoblast cells are largely unknown. Here, we observed cell senescence and expression of glucose transporter 3 (GLUT3) were up-regulated in GDM placenta. In vitro study confirmed the migration and invasion abilities were impaired in H2O2-induced senescent choriocarcinoma JEG3 cells. Additionally, high glucose triggered senescent

A risk assessment model for gestational diabetes mellitus integrating maternal demographics and hematological indices in Thai women.

BACKGROUND: Universal screening for Gestational Diabetes Mellitus (GDM) is the gold standard, but it can be difficult and expensive in resource-limited settings. Many current prediction models rely on specialized tests that are not available in primary care. This study aimed to create a cost-effective risk assessment model using common maternal demographic and blood test data to help identify GDM risk in Thai pregnant women. METHODS: We conducted a cross-sectional study with 200 pregnant women (24-28 weeks of gestation) at a secondary care hospital in Southern Thailand. We analyzed routine an

Continuous Glucose Monitoring for Women with Gestational Diabetes, CORDELIA: A Multicentric Randomized Controlled Trial, Methodology and Design.

Background/Objectives: Gestational diabetes mellitus (GDM) is associated with increased short- and long-term risks for both mother and child. Screening and treatment between 24 and 30 weeks' gestation reduces adverse pregnancy outcomes, while emerging evidence suggests that earlier diagnosis and treatment (<20 weeks') in high-risk populations may further improve neonatal outcomes. Currently, self-monitoring of blood glucose (SMBG) remains the standard of care for management but provides only intermittent glycemic data and limited insight into overall glucose patterns. Real-time continuous gluc

Maternal and Antenatal Complications in Pregnancies Complicated by Fetal Intra-Abdominal Umbilical Vein Varix.

Background/Objectives: Prior research suggests that fetal intra-abdominal umbilical vein varix (FIUVV) is associated with an increased risk of structural anomalies, aneuploidy, and poor obstetrical outcomes such as growth restriction and demise. Data in prior studies, however, are limited. The aim of this study is to determine the natural progression of FIUVV and its association with maternal and antenatal complications. Methods: This is a retrospective cohort study of singleton pregnancies that received an ultrasound between July 2020 and June 2023. FIUVV was diagnosed based on the presence o

Impact of matched samples on domain harmonization in cross-cohort NMR metabolomics.

INTRODUCTION: Machine learning models trained on metabolomics data frequently show reduced performance when applied to independent cohorts because of domain shifts introduced by differences in analytical platforms, sample handling, storage conditions, and metabolite quantification procedures. Robust approaches for harmonizing metabolomics datasets across cohorts are therefore needed to improve model transferability and reproducibility. OBJECTIVES: To evaluate the impact of matched samples on domain harmonization and cross-cohort prediction performance in NMR metabolomics data, and to compare

Maternal Gestational Hypertension Is Associated with Reduced TSG-6 in Human Milk: Potential Implications for Neonatal Immunological Support.

Tumor necrosis factor alpha-stimulated protein 6 (TSG-6) is a potent anti-inflammatory mediator secreted by mesenchymal stem cells (MSCs). In human milk (HM), these cells (human milk mesenchymal stem cells, HM-MSCs) contribute to postnatal programming of the neonatal gastrointestinal immune axis. This study assessed how maternal gestational hypertension and maternal age affect the cytoprotective TSG-6 pool and its associated immunomodulatory microRNA signature in early milk. Early colostrum, 2-4 days postpartum, from healthy mothers and hypertensive patients was analyzed. TSG-6 gene expression

Sugar-Specific Modulation of Trophoblast Function and Bioenergetics Under Metabolic Stress Conditions.

How sugars other than glucose affect trophoblast function is poorly understood, particularly under the metabolic stress of diabetic pregnancy. We compared glucose, fructose, and the minimally metabolized sugar allulose in two trophoblast cell lines, SW71 (first-trimester, extravillous-like) and BeWo (choriocarcinoma-derived, villous). Each sugar was added at 25 mM for 24 h to the cells' standard glucose-containing media (11 mM, SW71; 17.5 mM, BeWo) to model metabolic stress. All three sugars increased the number of cells recovered (2.4- to 6.5-fold in SW71; ~10- to 15-fold in BeWo) and reduced

Maternal vitamin D status, deficiency prevention strategies, and perinatal health outcomes.

Adequate maternal nutrition is vital for fetal development and maternal health. Although vitamin D is a widely utilized prenatal supplement, its clinical efficacy in improving major pregnancy outcomes remains a topic of debate. This study reviewed the physiological roles of vitamin D, evaluated the clinical impact of maternal supplementation on perinatal indices, and provided practical clinical recommendations within the context of regional public health challenges. Although observational data have consistently associated low maternal 25-hydroxyvitamin D levels with adverse outcomes, intervent

Placental Insulin-like Growth Factor Signaling and Neurodevelopment: Emerging Evidence from the Placenta-Brain Axis.

The field of neuroplacentology is a growing area that focuses on the influence of placental function on brain development. This field has expanded our understanding of the placenta's crucial role in proper neurodevelopment as well as placental anomalies that increase the risk of adverse neurodevelopmental outcomes. Neuroplacentology studies have revealed distinct roles of placentally provided hormones and nutrients in neurodevelopment whose disruption by perinatal problems, including inflammation, can increase the risk of neurodevelopmental disorders such as autism spectrum disorder and ADHD.

Integrated single-cell multi-omics analysis delineates neonatal immune dysregulation associated with gestational diabetes mellitus.

INTRODUCTION: Gestational diabetes mellitus (GDM) is a common pregnancy complication which could induce immune dysfunction. Although intrauterine hyperglycemia impairs fetal immune development, its cell-type specific effects on neonatal immune cells, and cell‑cell communication remain unclear. METHODS: In this pilot study, we performed integrated 5' single‑cell RNA sequencing and TCR repertoire analysis of cord blood mononuclear cells from two GDM and one type 2 diabetes mellitus (T2DM) mother-neonate pairs, with public healthy cord blood data (GSE157007) as controls. After quality control, 5
