Recurrent pregnancy loss, psychological distress and wellbeing support for women: a mixed-methods analysis.

BACKGROUND: Research suggests multiple experiences of miscarriage, also known as recurrent pregnancy loss (RPL), may be associated with compounded risk for adverse mental health impacts. However, there is a need for research evidence that offers clarity in relation to potential predictors of psychological distress and wellbeing among this neglected population of women. This paper aims to explore physical and experiential factors associated with miscarriage-related psychological distress and wellbeing among women who had experienced RPL. METHODS: Adopting a mixed methods approach, the paper pr

Insulin resistance and SIK1 hyperactivation: implications for vascular remodeling in recurrent pregnancy loss.

Recurrent pregnancy loss (RPL) is a multifactorial condition, with nearly half of cases remaining unexplained, and maternal insulin resistance is identified as a significant contributor. This study examined the role of salt-inducible kinase 1 (SIK1) in RPL patients associated with insulin resistance using villi samples, in vitro trophoblast models, and an insulin-resistant mouse model. Our results revealed that a marked increase in miscarriage risk was observed in women with a higher value of homeostatic model assessment for insulin resistance (>2.41). SIK1 expression was elevated in the villo

Identification and validation of paraptosis-related biomarkers in recurrent miscarriage.

BACKGROUND: Recurrent miscarriage (RM) is a pregnancy complication with growing evidence suggesting a role for paraptosis in its pathogenesis, though the underlying mechanisms remain unclear. This study investigated paraptosis-related genes (PRGs) as potential therapeutic targets. METHODS: Transcriptome data for RM were obtained from public databases, while PRGs were sourced from existing literature. Biomarkers were identified through the intersection of differential expression analysis, weighted gene co-expression network analysis, machine learning algorithms and expression validation, follo

Case Report: A novel approach to prevent chronic histiocytic intervillositis and recurrent pregnancy loss by targeting maternal alloimmunity.

Recurrent miscarriage is a distressing condition with limited therapeutic options. Chronic histiocytic intervillositis of unknown etiology (CIUE) is a rare inflammatory placental disorder characterized by maternal immune cell infiltration of the intervillous space, fibrin deposition, and ischemic tissue damage, leading to pregnancy loss. The condition likely reflects an immune response against paternal alloantigens, with histopathological features resembling antibody-mediated rejection in solid organ transplantation. We investigated two women with recurrent CIUE-related pregnancy losses. Detai

Identification of oxidative stress-responsive genes in recurrent miscarriage and their role in disease pathogenesis.

Recurrent miscarriage (RM) is a distressing reproductive condition affecting approximately 1-3% of couples. The underlying causes related to oxidative stress remain largely unclear and necessitate additional research. This study aimed to employ bioinformatics approaches to uncover the differential expression of oxidative stress-responsive genes in RM to elucidate their potential involvement in the disorder etiology. Upon examination of the data retrieved from the Gene Expression Omnibus (GEO), 18 oxidative stress-responsive differentially expressed genes (OSRDEGs) were identified. Bioinformati

Loss of Parkinson Disease Protein 7 (PARK7) upregulates ROS and cell migration and is associated with recurrent pregnancy loss.

BACKGROUND: Successful implantation is dependent on a synchronous dialogue between several proteins that act to control cellular dynamics including actin and microtubule reorganisation and cell motility. An impaired crosstalk can lead to complications including recurrent pregnancy loss (RPL) though the precise mechanisms remain unclear. Parkinson disease protein 7 (PARK7; encoding DJ-1), characterised for its participation in neurodegeneration, has emerged as a novel cytoskeletal and antioxidant regulator however, the role of DJ-1 in early pregnancy is unknown. METHODS: We employed systems bio

Endometrial Cell Senescence and Recurrent Spontaneous Abortion: Biomarker Potential of UCP2 and GSR.

This study explores the molecular mechanisms driving recurrent spontaneous abortion, a condition affecting 1-5% of women of reproductive age, with 40-50% of cases unexplained. Focusing on endometrial cell senescence, a process linked to irreversible cell cycle arrest, the research identifies uncoupling protein 2 (UCP2) and glutathione reductase (GSR) as key contributors to this pathology. By analyzing transcriptome data from the Gene Expression Omnibus database, 21 genes associated with cellular senescence were found to be differentially expressed, with UCP2 and GSR significantly upregulated.

Comprehensive DNA methylation profiling of sperm in male partners of couples with unexplained recurrent pregnancy loss.

BACKGROUND: Recurrent pregnancy loss (RPL) affects fertility problems in approximately 5% of couples, while the cause of RPL remains unknown in about half RPL cases, which is also called unexplained RPL. The male factors were associated with RPL in male partners, including chromosome abnormality and sperm DNA fragmentation. DNA methylation is one of the most extensively studied epigenetic factors that could help elucidate the mechanism underlying RPL in male partners. RESULTS: We revealed DNA methylation alternations occurring in sperm of RPL partners compared with the controls by genome-wide

Burden, causes, and treatment approaches of recurrent pregnancy loss: a scoping review.

Recurrent pregnancy loss (RPL), defined as the loss of two or more clinically recognized pregnancies before 20 weeks of gestation, poses a significant global medical challenge. Affecting up to 5% of couples trying to conceive, its multifactorial causes include genetic, anatomical, immunological, hormonal, and environmental factors. However, 65% of cases remain idiopathic, exacerbating the complexity of diagnosis and treatment. Low- and middle-income countries (LMICs) face additional challenges due to limited diagnostic capacities, socioeconomic disparities, and the burden of infectious disease
