BACKGROUND: Down syndrome or trisomy 21, the most common chromosomal disorder, results from the presence of a third copy of chromosome 21 and manifests as mild to moderate intellectual disability, growth retardation, congenital heart defects, gastrointestinal abnormalities, and characteristic facial features. Several methods have been used to screen for Down syndrome in the prenatal period, such as ultrasound, biomarkers, cell-free DNA testing, and combinations of these tests. A positive result from one or more of these screening tests signals the need for confirmatory karyotyping to clinch th

INTRODUCTION: Meckel-Gruber syndrome (MKS, OMIM 24,900), also known as Meckel syndrome, is a rare and severe autosomal recessive disorder. The syndrome is typically characterized by a triad of occipital encephalocele, bilateral renal cystic dysplasia, and postaxial polydactyly. MKS shows significant clinical heterogeneity, which poses challenges for accurate prenatal diagnosis. Prenatal ultrasound is an important tool for detecting potential cases, but the complexity of MKS often requires additional advanced techniques such as prenatal whole-exome sequencing (WES) to provide more accurate mole

Caffey's disease, or infantile cortical hyperostosis, is a rare disorder characterized by subperiosteal new bone formation and diffuse soft tissue inflammation in infants younger than six months. A much rarer prenatal form presents before birth with diffuse, symmetric cortical thickening and often results in perinatal death, especially when it develops early in gestation. The severe form may be associated with pulmonary hypoplasia, hepatic dysfunction, and high neonatal mortality. We report the case of a premature infant born at 27 weeks' gestation to a 36-year-old mother with chronic hyperten

Fetal brain development is a complex and dynamic process, and its disruption can lead to significant neurological disorders. Early detection of brain aberrations during pregnancy is critical for optimizing postnatal medical intervention. We propose a deep generative anomaly detection framework, conditional cyclic variational autoencoding generative adversarial network (CCVAEGAN), that can identify structural brain anomalies using fetal brain magnetic resonance imaging. CCVAEGAN leverages covariate conditioning on gestational age and cyclic consistency training to generate high-fidelity normati

Placental mesenchymal dysplasia (PMD) is a rare disease with unclear etiology and peculiar placental findings (placentomegaly, abnormal chorionic vessels with thromboses, and grossly enlarged and often cystic-appearing stem villi). Songraphic findings are cystic changes, hypoechoic areas, placental enlargement, and dilated chorionic vessels.PMD can be associated with severe perinatal complications, e.g., intrauterine fetal demise, fetal growth restriction, and preterm delivery. Neonatal complications include Beckwith-Wiedemann Syndrome (BWS), hepatic tumors, and hematologic diseases, i.e., ane

BACKGROUND: Congenital dysfibrinogenemia is a rare autosomal dominant disorder involving abnormal fibrinogen function, leading to variable risks of bleeding and thrombosis. Its management during pregnancy is particularly challenging due to the potential for complications such as miscarriage, stillbirth, placental abruption, and fetal growth restriction. The absence of reliable predictive laboratory markers further complicate individualized risk assessment and clinical decision-making. CASE PRESENTATION: A 23-year-old primigravida was diagnosed with congenital dysfibrinogenemia after low fibrin

INTRODUCTION: Congenital capillary-lymphatic-venous malformations (CLVMs) often result in life-threatening complications, which may begin in utero. Sirolimus, a mammalian target of rapamycin (mTOR) inhibitor, has been successfully used in children and adults with CLVMs due to its antiproliferative and antiangiogenic properties. However, only two cases of prenatal sirolimus treatment of fetal CLVMs have been published to date, with very limited data on optimal therapeutic scheme and drug dosing.
CASE REPORTS: Here we report two cases of effective prenatal and postnatal sirolimus treatment of e

Dominant β-thalassemia is a rare form of thalassemia that is caused by a heterogenous group of molecular defects, including missense, nonsense, and frameshift mutations. Among the missense mutations involving the third exon of β-globin gene is the rare Hb Dieppe (HBB:c. 383A > G) which leads to a very unstable β-variant. In the current study we report this variant in an 8-year-old girl and her 35-year-old mother in an Iraqi Kurdish family, both presenting as β-thalassemia intermedia with moderate hypochromic anemia, increased hemoglobin F and borderline hemoglobin A2. This constitutes the firs

Xp21 deletion is a contiguous gene deletion syndrome characterized by adrenal hypoplasia congenita (AHC). If prenatal genetic diagnosis does not determine whether a male fetus at risk is affected, it is necessary to monitor him to diagnose adrenal insufficiency in the first few days of life to avoid a salt-losing adrenal crisis. Whether AHC can be diagnosed by prenatal ultrasound is not known. Ultrasound findings of fetal adrenal glands were compared between healthy and affected siblings whose mother was a carrier of the Xp21 deletion. The couple did not want an invasive prenatal diagnosis dur
