Coarse grained molecular dynamics analysis of menthol interactions with ergosterol in a fungal mimetic membrane.

Menthol, a monoterpene from peppermint oil, has antifungal activity against Candida species. However, how menthol interacts directly with fungal membrane sterols is still not well understood. In this study, coarse-grained molecular dynamics simulations were used with the Martini force field to investigate menthol in a fungal-mimetic POPC/ergosterol (8:2) membrane. Radial distribution function analysis revealed preferential spatial association between menthol and ergosterol, including a local maxima at approximately 5.0 Å and a broader peak in the 9.5 Å range. Contact analysis further demonstra

Development of Pyrazole and Pyrazolo[4,5-b]pyridine Analogues with Potent Fungal Biofilm Inhibition Properties.

INTRODUCTION: Fungal infections continue to pose significant health risks because of two factors, which include drug resistance and the restricted availability of treatment options. Molecular hybridization has been an effective method for developing new drugs with improved therapeutic effects. Fused heterocyclic systems provide researchers with useful frameworks for synthesizing compounds that more effectively engage biological targets. The pyrazole and pyrazolo[ 4,5-b]pyridine derivatives have attracted significant scientific interest due to their multiple biological functions and their abili

Flavaglines as cross-kingdom translation inhibitors: Heavy halides at C6 potentiate antineoplastic, antiviral, and antifungal activities.

The growing prevalence of cancer, coupled with the rise of multidrug-resistant pathogens, presents a formidable challenge to modern medicine. The flavagline class of secondary metabolites has been shown to possess a unique capacity to target multiple conserved pathways across various human malignancies and microbial pathogens. While numerous structure-activity relationship (SAR) campaigns have established trends with respect to specific indications, cross-kingdom comparative analyses have not been reported to date. In this work, a structurally diverse library comprising 17 novel and 5 natural

Synergistic multifunctional biomedical activity of microwave-synthesized Au/Pt/Cu trimetallic nanofluids: experimental and DFT insights.

In the present study, a trimetallic Au/Pt/Cu nanofluid was synthesized via a rapid one-pot microwave-assisted strategy and systematically evaluated for multifunctional biomedical activity. Structural and spectroscopic characterization confirmed the formation of an integrated multimetallic nanostructure, likely to contain alloyed or compositionally mixed domains that generate synergistically active interfaces of Au, Pt and Cu. The nanofluid exhibited broad-spectrum antimicrobial activity, inhibiting Gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis) and Gram-negative bacteria (Es

Performance evaluation of Nile tilapia (Oreochromis niloticus) fed diets supplemented with Lactobacillus plantarum, Bacillus subtilis and Candida infanticola.

Aquaculture is the fastest-growing agricultural activity globally, providing healthy food with comparatively low environmental impact. However, high stocking densities can generate excessive organic matter, causing stress and increasing pathogen susceptibility in fish. This study evaluated the effects of dietary supplementation with Candida infanticola UFSJ7A, Bacillus subtilis ATCC 19,659, and Lactobacillus plantarum UFSJP2 on Nile tilapia (Oreochromis niloticus) performance, intestinal morphology, in vitro antagonistic activity against fish pathogens, and pond water quality. Drug repurposing-inspired design of miconazole-based 1,2,3-triazoles as antichagasic agents.

Drug-repurposing strategies have been widely explored to accelerate the development of new treatments for Chagas disease, leveraging compounds with previously established safety profiles. Among these, azole antifungals have attracted attention due to their inhibitory activity against 14-α-lanosterol demethylase (CYP51), an enzyme also present in Trypanosoma cruzi and essential for its ergosterol biosynthesis. Inhibition of this pathway compromises parasite membrane integrity and function and several azoles, including posaconazole, ravuconazole, and miconazole, have demonstrated significant in

Antifungal activity of propolis against Candida spp.

Fungal infections caused by Candida species remain a major clinical and pharmacological challenge, ranging from superficial mucocutaneous disease to invasive infections associated with considerable morbidity. Vulvovaginal candidiasis is among the most common manifestations and is frequently recurrent. Increasing antifungal resistance has reduced the effectiveness of azoles and other standard therapies, highlighting the need for alternative or complementary treatment strategies. This systematic review aimed to synthesize recent evidence on the antifungal activity of propolis and its mechanisms

Profile of Invasive Fungal Infections in a Paediatric Haemato-Oncology Unit: A Five-Year Retrospective Cohort Study From a Tertiary Care Centre in South India.

BACKGROUND: Invasive fungal infections (IFIs) are a leading cause of morbidity and mortality in paediatric haemato-oncology (PHO) patients. Empirical antifungal therapy in high-risk children must be informed by local epidemiological, species-level and antifungal-susceptibility data, yet contemporary Indian paediatric cohort data remain limited. OBJECTIVE: To describe the clinical, microbiological, therapeutic and outcome profile of probable and proven IFI in a tertiary care South Indian PHO unit over five years, and to benchmark the findings against contemporary international literature. Antifungal Activity Assessment of Postbiotic Mediators Derived From Lactobacillus rhamnosus GG and Lactobacillus reuteri Against Candida albicans.

The rise of antimicrobial resistance has increased interest in safe alternative therapeutic agents, including postbiotic mediators (PMs), which are sterile, cell-free, filter-sterilized preparations derived from probiotics. This study investigated the antifungal and antibiofilm activity of PMs derived from Lactobacillus rhamnosus GG (LGG) and Lactobacillus reuteri (LR) against Candida albicans, an opportunistic pathogen implicated in oral infections. PMs were prepared from LGG and LR cultures. The minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) against C. albi

Association of quantitative sputum Candida load with respiratory dysbiosis and airway inflammation in suspected invasive pulmonary candidiasis.

UNLABELLED: The clinical significance of Candida species isolation from sputum remains ambiguous due to the colonization-infection dilemma. Integrating quantitative culture with microbiome profiling may clarify its role, yet systematic evidence is scarce. In this exploratory study (June 2023-June 2026), we enrolled 140 hospitalized patients suspected of invasive pulmonary candidiasis (IPC) and stratified them into possible IPC (n = 36), probable IPC (n = 71), and negative control (n = 33) groups based on clinical criteria. All underwent quantitative fungal culture of sputum and metagenomic nex
