Fabrication, spectral characterisation and exploration of DNA/RNA binding affinity and biological potential of transition metal complexes incorporating isoniazid and aromatic nitrogen-donor ligands.

Four mixed-ligand transition metal(II) complexes, [ML(L1)Cl2] (where M = Co, Ni, Cu, and Zn; L = Schiff base derived from isoniazid and 3-nitrobenzaldehyde; L1 = 1,10-phenanthroline), were synthesised and characterised using elemental analysis, FT-IR, UV-Vis, 1H NMR, EPR (for Cu), and molar conductance measurements. Physicochemical and spectroscopic data suggested an octahedral geometry for all complexes. Antimicrobial assays revealed that the complexes exhibited greater activity than the free Schiff base ligand, with inhibition zones of 14-22 mm against Staphylococcus aureus and Escherichia c

Mechanism of action of quinoline-based ionic liquids against the pathogenesis of the fungal pathogen, Candida albicans.

Quinoline-based ionic liquids (ILs) are being evaluated for their potential antifungal activity, making them promising alternatives to traditional antifungal therapies. The peculiar properties, characteristic of the utilization of ILs, including their solvation effect, greatly influence their broad-spectrum activity against a wide range of fungal species. Thus, in this study, for the first time, to the best of our knowledge, we aim at exploring the mechanistic action of the previously synthesized quinoline-based IL, 1-hexadecylquinolin-1-ium bromide, [C16quin]Br, against the pathogenesis of th

Harnessing probiotics against candidiasis: a systematic review of experimental and mechanistic evidence.

BACKGROUND: Candida species are among the most important opportunistic fungal pathogens responsible for superficial mucosal infections and life-threatening invasive candidiasis, particularly in immunocompromised individuals. The increasing prevalence of antifungal resistance, biofilm-associated tolerance, and the emergence of multidrug-resistant species such as Candida auris have highlighted the need for alternative or adjunctive therapeutic strategies. Probiotics have emerged as promising microbiome-based interventions owing to their ability to inhibit Candida growth, suppress virulence trait

Exploiting Neosartorya (Aspergillus) fischeri antifungal protein 2-defined fungal targets for drug repurposing against Candida albicans.

Invasive fungal infections caused by Candida albicans remain major clinical challenges because of the limited therapeutic options, toxicity, and emerging resistance. Drug repurposing offers a promising strategy to accelerate antifungal discovery by leveraging existing compounds with known safety profiles. In this study, we applied a structure-based drug repurposing approach guided by the antifungal protein Neosartorya (currently Aspergillus) fischeri antifungal protein 2 (NFAP2), which targets fungus-specific intracellular proteins, including glutamate decarboxylase, mitochondrial ATP synthase

Diverse genetic pathways drive convergent fluconazole and caspofungin resistance phenotypes in Candida albicans.

The emergence of antifungal resistance in Candida albicans is a major worldwide public health threat, particularly for immunocompromised patients. Antifungal resistance may arise through uniform resistance mechanisms or through variable evolutionary routes. The aim of this study was to investigate whether antifungal resistance arises in C. albicans through shared or isolate-specific mechanisms, in other words, following convergent or divergent adaptive pathways and genetic mechanisms. Six laboratory-induced fluconazole-resistant and six caspofungin-resistant isolates were analyzed using whole-

Electrospun PVP Nanofibrous Co-Loaded with Miconazole and Lidocaine for the Treatment of Oral Candidiasis.

INTRODUCTION: Oral candidiasis caused by Candida albicans is common among immunocompromised individuals. Conventional topical dosage forms have several limitations, including imprecise dosing, rapid salivary clearance, short residence time at the site of application, and inconsistent therapeutic efficacy. RATIONALE: To overcome these challenges, this study developed an electrospun nanofibrous patch for localized co-delivery of miconazole (MZ) and lidocaine (LD) to oral mucosa. MZ and LD were co-encapsulated in electrospun polyvinylpyrrolidone (PVP) nanofibers and evaluated for morphology, dru

Changes in age composition and epidemiology of candidaemia, Iceland, 2012 to 2023.

BACKGROUNDCandidaemia places a substantial burden on healthcare systems. Long-term, population-based studies of candidaemia rates with appropriate adjustments for changes in age composition are lacking.AIMWe aimed to determine trends in candidaemia incidence rates, species distribution, antifungal susceptibility, and concurrent antifungal use in Iceland between 2012 and 2023, as well as long-term (2000-2023) trends in age-standardised incidence rates (ASRs).METHODSWe analysed patient data with blood cultures positive for Candida species, or yeasts previously assigned to the genus Candida. We c

Agricultural azole fungicides induce cross-resistance to medical antifungals in Candida tropicalis: a One Health perspective on antifungal resistance.

The human opportunistic fungal pathogen Candida tropicalis is widely distributed in natural environments. In recent decades, the prevalence of azole-resistant isolates has increased markedly in the Asia-Pacific and Latin America. This epidemiological change could be associated with the extensive application of azole fungicides in modern agriculture. In this study, we demonstrate that exposure to four widely used agricultural azoles (difenoconazole, diniconazole, flutriafol, and hexaconazole) induces cross-resistance to medical azoles in an initially susceptible C. tropicalis strain. The evolve

NAD⁺-driven modular multi-enzyme cascade system enables the in vitro synthesis of 3-hydroxypropionic acid from glucose.

3-Hydroxypropionic acid (3-HP) is an important bio-based platform chemical with broad industrial potential. Most reported in vitro biosynthetic systems rely on costly NADPH, which limits scalability. Here, a modular in vitro multi-enzyme cascade pathway was designed using D-glucose as the sole carbon source, driven exclusively by the low-cost NAD⁺/NADH redox pair. Three functional modules were constructed: a pyruvate accumulation module built on thermophilic enzymes, an alanine cycling module proposed as a conceptual strategy for β-alanine regeneration through a CO₂-mediated carboxylation-deca

Postmarketing reporting patterns of Candida-related events associated with bimekizumab and selective IL-17A inhibitors in psoriasis: a FAERS and JADER pharmacovigilance study.

BACKGROUND: Candida-related events are established labelled events during IL-17 pathway inhibition. The remaining pharmacovigilance question is how Candida phenotypes are represented in spontaneous reports for bimekizumab and selective IL-17A inhibitors within psoriasis, while accounting for the interpretive limits of reporting-system data. METHODS: We performed a phenotype-focused, dual-database disproportionality study using FAERS 2023Q4-2026Q1 as the primary source and the JADER May 2026 release as a complementary consistency source. Analyses were restricted to psoriasis-associated reports

Impact of Early Bloodstream Infections on Clinical Outcomes Following Allogeneic Hematopoietic Cell Transplantation with Post-Transplant Cyclophosphamide-Based GVHD Prophylaxis.

BACKGROUND: Bloodstream infections (BSIs) remain a significant cause of morbidity and mortality following allogeneic hematopoietic cell transplantation (allo-HCT), particularly during the post-transplant neutropenia and immune reconstitution phase. Notably, studies have identified high rates of BSI in the early post-transplant period with PTCy regimens, including single-center cohorts that have highlighted Candida, Enterococcus, and gram-negative rods (GNRs) as predominant pathogens. However, the broader clinical impact of pathogen-specific BSI on post-HCT outcomes in PTCy recipients remains i

Topical Insulin and Infectious Keratitis: An In Vitro Microbial Growth Study.

PURPOSE: Topical insulin eye drops are used clinically to promote corneal epithelial healing in persistent epithelial defects. Given these benefits, insulin may also aid epithelial recovery in infectious keratitis. However, it remains unclear whether insulin impacts the growth of common ocular pathogens. This study aimed to evaluate the effect of insulin on microbial growth dynamics in vitro. METHODS: Regular and rapid insulin were tested at 1 IU/mL and 10 IU/mL in tryptic soy broth (TSB) and lubricating eyedrops. A single clinical isolate of each organism (Staphylococcus aureus, Staphylococc
