Polyamine depletion inhibits murine and human norovirus infection.

UNLABELLED: Noroviruses are non-enveloped, positive-sense RNA viruses in the family Caliciviridae. Human noroviruses (HNoVs) are the leading cause of viral gastrointestinal disease. Despite their clinical relevance, therapeutic strategies for norovirus infections remain limited, partly due to challenges in cultivating HNoV and limited understanding of virus-host interactions. Polyamines (PAs) are small polycationic metabolites derived from amino acid metabolism that are essential for diverse cellular functions and serve as important host factors for many RNA viruses. However, their role in nor

Etiological surveillance of viral gastroenteritis in children aged less than 5 years in Tehran Province, Iran, 2023-2024.

BACKGROUND: Acute gastroenteritis (AGE) is a primary cause of death in children less than five years old in low- and middle-income countries (LMIC), mostly caused by viral intestinal infections. While the disease poses a significant global burden, further investigation is needed to clarify the role of enteric viruses in disease pathogenesis. This work aimed to study the frequency of five major enteropathogenic viruses-Rotavirus (RV), Norovirus (NoV), Adenovirus (AdV), Sapovirus (SaV), and Astrovirus (AstV)- in stool samples of young children in Tehran, Iran, during 2023-2024. METHODS: A cross

Gait variability and local dynamic stability are exacerbated by age and visual cues.

BACKGROUND: Gait variability increases and dynamic gait stability decreases with age, which may lead to an increased risk of falls. Additionally, the need to process visual cues can exacerbate changes in gait variability and dynamic gait stability in older adults. The purpose of this study was to compare gait variability and dynamic gait stability in young and older adults walking overground and on a treadmill with and without visual cues. We hypothesized that gait variability would increase and dynamic gait stability would decrease during the visually cued gait. We also hypothesized that olde

Clinical Importance of Autoantibodies to SOX10 and Lamin A/C as Potential Biomarkers in Sera From Vitiligo Patients.

BACKGROUND: Diarrhea has been the second leading cause of death among children under the age of five, and the rapid and accurate pathogen diagnosis in patients with diarrhea is crucial for reducing morbidity and mortality. A newly developed one-step multiplex real-time PCR assay, the Allplex GI-Virus Assay, was evaluated for its ability to detect six diarrhea-causing viruses (rotavirus, norovirus genogroup I (GI) and genogroup II (GII), enteric adenovirus, astrovirus, and sapovirus) in stool samples. METHODS: The performance of the Allplex assay was compared with those of another multiplex PC

Apigenin derivatives as promising norovirus RdRp inhibitors: insights from in silico docking and molecular dynamics studies.

Norovirus is a leading cause of acute gastroenteritis worldwide, yet no approved antivirals currently exist. In this study, we employed in silico approaches to evaluate apigenin derivatives as potential inhibitors of norovirus RNA-dependent RNA polymerase (RdRp). Seventy-three compounds were initially retrieved from the ZINC database, of which 36 satisfied Lipinski's Rule of Five and were advanced for detailed analysis.Pharmacokinetic predictions revealed high intestinal absorption and favourable drug-likeness profiles for most derivatives, with limited central nervous system penetration, desir

Variable and conserved B cell epitopes of GII.4 human noroviruses.

Norovirus is a leading cause of acute gastroenteritis worldwide, imposing a major burden on public health and healthcare systems. Despite its significant medical impact, no licensed vaccines or specific antiviral therapies are currently available. Norovirus vaccine development is complicated by several factors, including the extreme genetic and antigenic diversity. In particular, the predominant genotype GII.4 exhibits a continuous emergence of novel variants that can evade immune responses acquired from previous infections. In this manuscript, we will summarize the characteristics and current

[Gastrointestinal PCR Panel Results and Evaluation of Inflammatory Biomarkers in Acute Gastroenteritis Cases].

Acute gastroenteritis (AGE) is an important public health problem that is very common all over the world. Knowing the causative agents is important for understanding the clinical course of the disease, effective treatment and necessary precautions to be taken. In this study, we aimed to determine the pathogen distribution in AGE cases admitted to our center using multiplex real-time polymerase chain reaction (mRt-PCR) and to demonstrate the clinical and laboratory differences between luminal (type I) and mucosal/invasive (type II) infections. We also aimed to define biomarker thresholds to fac

Rotavirus Outbreak Traced in Clinical, Environmental, and Food Matrices: Whole Genomic Characterization of an Equine-like G3P[8] Genotype.

The emergence of new viral strains has been frequently associated with the onset of outbreaks in different epidemiological settings. This study investigated an acute gastroenteritis (AGE) outbreak, which occurred at a university campus in southern Brazil, in 2024, affecting approximately 30-40% of undergraduate students, with over 400 reported cases. The objectives were to describe the epidemiological and molecular features of the outbreak, identify the probable transmission route, and assess the genetic relatedness of the detected strain to recently circulating rotavirus A (RVA) lineages. Norovirus Outbreak Due to Person-to-person Transmission in a College in Kerala, India, 2021.

We investigated a cluster of acute gastroenteritis reported from a college in Kerala on December 1, 2021. We defined a suspect case as an occurrence of vomiting or at least three episodes of loose stools within 24 h. We identified 113 suspected case patients; 63 (55.8%) were boys, and 110 (97.3%) were hostel inmates. All five stool samples and three out of eight water samples we collected tested positive for norovirus. We conducted a retrospective cohort and did univariate and multivariable binomial regression to calculate the adjusted risk ratio (aRR). Norovirus infection risk was higher amon

Detection of the globally emerging GII.17[P17] norovirus during an acute gastroenteritis outbreak in Romania, 2024.

Norovirus is a major cause of acute gastroenteritis (AGE) globally. GII.17[P17] has been reported in the last decade as a prevalent norovirus genotype worldwide. We documented the circulation of norovirus GII.17[P17] isolates during an AGE outbreak occurring in western Romania, in 2024. Norovirus whole-genome sequences were obtained by Illumina technology and used for inferring phylogeny and to identify amino acid substitutions harboured by the VP1 major capsid protein and by the RNA-dependent RNA polymerase (RdRp). Phylogenetic analyses indicated high degree of relatedness between the Romania

Ancestral FUT2 genetic adaptation confers resistance to modern norovirus and rotavirus infections.

Human genetic diversity plays a key role in modulating susceptibility to infectious diseases, reflecting a long history of host-pathogen co-evolution. While clinical outcomes were once attributed primarily to pathogen virulence, evidence today underscores the impact of host genes in infection susceptibility. Noroviruses and rotaviruses, leading causes of acute gastroenteritis, illustrate this dynamic interplay. Resistance to gastroenteritis infection in a subset of individuals is conferred by loss-of-function mutations in FUT2, which encodes α(1,2)-fucosyltransferase responsible for histo-bloo
