Candida albicans is an opportunistic human pathogen, that commonly causes localised infection of mucosal surfaces. Candida can also cause systemic infections, which remain difficult to treat and have a high risk of mortality. The immune response to C. albicans infection is complex and can be influenced by the surrounding microenvironment, for example, metabolic stresses or co-infection. Macrophages are a key immune cell in defence against C. albicans infection through phagocytosis of C. albicans and cytokine production to co-ordinate immune responses. Here, we utilise Data Independent Acquisit

CARD9 deficiency is characterized by predisposing to increased susceptibility to fungal infections, particularly invasive fungi and dermatophytes. In recent years, the incidence of fungal infections in patients with CARD9 deficiency is on the rise, and the types of fungal pathogens have become increasingly diverse. In addition, most of CARD9 deficiency patients with fungal disease were prone to recurrence and resistant to antifungal agents, which pose significant challenges to treatment. To investigate the association between CARD9 mutations and fungal infections, we conducted a systematic ana

BACKGROUND: Current international guidelines lack clear recommendations on the management of non-colonised patients undergoing intra-hospital transfer from the ward in which horizontal transmission of Candida auris is known to occur (defined as endemic for Candida auris) to wards with no horizontal transmission detected (defined as non-endemic wards), particularly regarding the timing and number of screening swabs needed to exclude colonisation.
METHODS: Single-center prospective observational study at a tertiary-care hospital in Genoa, Italy, including adults transferred from the C. auris en

The increase in invasive candidiasis caused by emerging Candida species presents a significant medical challenge due to their resistance to conventional antifungal treatments. Exploring novel therapeutic strategies, such as drug repurposing, is therefore essential. Tacrolimus, an immunosuppressive calcineurin inhibitor, shows potential antifungal effects. This study investigated the in vitro effect of combining tacrolimus and echinocandin anidulafungin using the checkerboard method against 60 clinical Candida isolates, including Candida albicans, Candida auris, Candida glabrata, and Candida pa

The collared peccary (Pecari tajacu) is a member of the Tayassuidae family widely distributed throughout the Americas, particularly in Brazil. These animals have important ecologic functions and, despite their good conservation status, are frequently affected by anthropogenic pressures. Candida spp. are commensal yeasts present in the microbiota of various hosts; however, under conditions that disrupt microbial balance, they can become opportunistic pathogens. Some Candida spp. are also of great relevance to the One Health framework, because they cause significant impacts on the health of huma

Invasive candidiasis is associated with high morbidity and mortality, and the rising of antifungal resistance underscores the need for new therapies. Rezafungin, a second-generation echinocandin, enables once-weekly dosing, achieves high plasma concentrations, and shows potent in vitro activity. We report two Brazilian cases showing its clinical utility: (i) fluconazole-resistant Candida tropicalis bloodstream infection in a patient with colorectal cancer and chronic kidney disease and (ii) azole-refractory C. albicans esophagitis in a patient with autoimmune polyglandular syndrome type 1. During 2022-2023, the Antimicrobial Resistance Laboratory Network tested 8,033 Candida auris clinical isolates in the United States. Overall, 95% of isolates were fluconazole resistant, 15% amphotericin B resistant, and 1% echinocandin resistant. Laboratory capacity for C. auris identification and antifungal susceptibility testing is essential to address this emerging public health threat.

Candida auris (Candidozyma auris, C. auris) is a fungal pathogen with multiple drug resistance, belonging to a species of yeast, known for its ability to sustain growth in the host. This organism can colonize human skin and spread through physical contact or contaminated surfaces and equipment. Given the limited reviews on skin load reduction methods of C. auris, this review aims to summarize and evaluate the current load reduction methods involving reducing skin load, and the risk of colonization. The findings will provide healthcare facilities and nursing homes with evidence-based guidance t

OBJECTIVES: Aureobasidin A (AbA) is a natural antifungal lipopeptide known to inhibit inositol phosphorylceramide (IPC) synthase. While its antifungal effect, mechanism via the inositol pathway involved in sphingolipid synthesis, and influence on ABC efflux pumps have been reported previously, its potential role in inducing programmed cell death and efficacy against oral candidiasis remain unexplored. This study aimed to elucidate a novel, complementary mechanism of AbA against Candida albicans (C. albicans), focusing on ROS-implicated apoptosis, and to evaluate its therapeutic potential for o

Cutaneous candidiasis remains a therapeutic challenge owing to limited skin penetration of anti-fungal agents, high recurrence, and increasing resistance to azoles. In this work, a chitosan-based film-forming gel (FFG) was developed incorporating luliconazole (LUL) and carvacrol (CAR) to improve dermal delivery and therapeutic performance. A 3² factorial design was employed to optimize polymer and plasticizer concentrations, with drying time, spreadability, and viscosity as key formulation attributes. The optimized gel exhibited rapid in situ film formation, suitable viscosity, and mechanical

Candidozyma auris (formerly Candida auris) has emerged within just over a decade as one of the most relevant multidrug-resistant fungal pathogens affecting human health worldwide. Its pathogenicity, capacity for skin colonization, environmental persistence, and resistance to antifungal drugs and disinfectants have all contributed to its consolidation as a leading cause of healthcare-associated outbreaks. Nevertheless, increasing evidence indicates that C. auris should not be viewed solely as a nosocomial yeast, but rather as part of a broader environmental continuum encompassing natural habita

Candida auris, an emerging multidrug-resistant fungal pathogen, poses a significant diagnostic and infection control challenge with limited identification methodologies. To overcome this, we developed and validated a rapid, specific, and direct colony real-time PCR assay for C. auris identification without DNA extraction. Species-specific primers were designed using comparative genomics and subjected to rigorous experimental validations. The optimized assay using a selected primer pair (Candida-auris-5F/5R) successfully identified C. auris directly from the cultured colonies within two hours.
