The World Health Organization has classified five species of Candida as pathogens of critical concern on its recent 'Fungal Pathogen Priority List'. Candida albicans is the most common species aetiologically associated with human and animal mucosa; however, we know almost nothing about the environmental survival and the transfer pathways of C. albicans in natural environments. Here, we have isolated C. albicans from the faeces of newly arrived migratory geese in two agricultural fields in Scotland (the United Kingdom). All 14 confirmed isolates were characterised in terms of their pathogenicit

The emergence of fluconazole-resistant Candida albicans poses a significant therapeutic challenge requiring alternative antifungal agents for treatment options. This study repurposed the antifungal potential of amitriptyline hydrochloride (AMP), a tricyclic antidepressant, against fluconazole-resistant C. albicans strain SC5314. The antifungal activity was evaluated using agar well diffusion and agar cup assays at a concentration of 2 mg/mL. Minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) assays were performed to quantify efficacy, complemented by time kill ki

The rise of drug resistance and limitations of current antifungal treatments highlight the urgent need for innovative antifungal strategies. Here we present the development of cis-fumaramidmycin-derived analogs inhibiting the interactions of ribosome assembly factor Mrt4 with rRNA to combat fungal infections. Through antifungal screening, we identified a promising lead 20 with strong efficacy against various drug-resistant fungi, including notorious super-fungus Candida auris. A comprehensive approach combining active-and-inactive-based protein profiling (AIBPP), chemical-genetic profiling, an

Candida auris (Candidozyma auris) is an emerging multidrug-resistant yeast with distinct phylogenetic clades that differ in their epidemiological characteristics. Rapid and practical methods for clade differentiation are needed for effective surveillance and infection control. We performed clade differentiation of C. auris clinical isolates collected in 32 Korean hospitals between 1996 and 2024 using multilocus sequence typing (MLST). All isolates were identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, with species confirmation via internal transcrib

The clinical management of Nakaseomyces glabratus is becoming increasingly difficult because of its intrinsic and acquired resistance to multiple antifungal drugs, particularly azoles. This situation highlights the urgent need for new antifungal agents. In this study, the antifungal efficacy and toxicity profile of an organometallic compound, named FE1, was evaluated in the in vivo model Galleria mellonella infection model. Larvae were inoculated with a standardized suspension of N. glabratus CBS138 and treated with FE1 or fluconazole at different concentrations (0.4, 4.0, and 20 mg/kg). Survi

Candida auris has emerged over the past fifteen years as a multidrug-resistant human fungal pathogen responsible for hospital-acquired infections associated with high mortality rates. Clinical isolates frequently exhibit resistance to one or more first-line antifungal drugs, and management is further complicated by persistent colonization, biofilm formation, and historical misidentification in diagnostic laboratories. Rapid species-level identification and accurate antifungal susceptibility testing are critical for effective patient care and infection control. In this article, we summarize the

The synergistic combination of two antimicrobial drugs is a promising therapeutic modality for many infectious diseases. However, systemic fungal infections still have a high mortality rate because of distinct in vivo distributions of the two drugs. Here we address this challenge by designing an antifungal polymer that forms micelles suitable for delivering a second antifungal agent to achieve temporal and spatial consistency of delivery. We show that the polymer, which mimics host defense peptides, exerts a synergistic effect with the antifungal amphotericin B (AmB). The AmB-encapsulated mice

Ocimum gratissimum L. (African basil) is widely used in ethnomedicine to treat infectious and inflammatory conditions, yet its antifungal mechanisms remain unclear. In this study, we evaluated the essential oil of O. gratissimum L. (OGEO) against Candida albicans biofilms, a clinically relevant driver of antifungal resistance in device-associated infections. Gas chromatography-mass spectrometry identified eugenol as the predominant constituent (59.5%), supported by terpenoid and sesquiterpene derivatives. Functional assays demonstrated potent antibiofilm activity, as exposure to 1% and 2% OGEO

Candida glabrata is the second leading cause of mortality in immunocompromised patients hospitalized for invasive candidiasis (IC). Several drugs have been available to treat this disease for decades, such as polyenes, azoles, echinocandins, flucytosine, and, in critical cases, amphotericin B. However, these antifungals' constant and routine use have led to the development of resistance mechanisms, making the design and development of new drugs indispensable. The first step for the design and subsequent synthesis of a new chemical molecule as a potential antifungal is the identification of new

Candida glabrata is an opportunistic fungal pathogen and a leading cause of invasive candidiasis, particularly in immunocompromised patients, where treatment is increasingly compromised by intrinsic and acquired antifungal resistance. Despite lacking morphological plasticity, C. glabrata employs distinct virulence strategies, including adhesin-mediated host colonization, intracellular survival within phagocytes, stress tolerance, iron acquisition and biofilm formation. This review synthesizes current knowledge of C. glabrata virulence and antifungal resistance mechanisms, with a particular foc

Neutrophil extracellular traps (NETs) constitute a vital antimicrobial defense mechanism of neutrophils, contributing to various physio-pathological processes; however, the role of plasma membrane asymmetry in this process remains unknown. Here we identify Xk-related protein 8 (XKR8), a plasma membrane phospholipid scramblase, as a pivotal regulator of NETs formation. Upon NETs induction, XKR8 is cleaved by caspase-3, thereby disrupting plasma membrane lipid asymmetry via phospholipid scrambling. Mutation of the caspase-3 cleavage site in XKR8 impairs NET formation. Inhibition of calcium signa

Contemporary data on risk factors for invasive candidiasis (IC) in solid organ transplant recipients (SOTr) are rare. This follow-up study on a large multicenter cohort study, nested within the Swiss Transplant Cohort Study, identifies risk factors for IC during the first year posttransplant. Among 4755 and 4519 SOTr identified between 0-3 months (early) and 4-12 months (late) posttransplant, 90 (1.9%) and 48 (1.1%) had 109 and 50 episodes of IC, respectively. Posttransplant Candida colonization (HR:3.2; 95%CI:1.67,6.14; p<0.001), and posttransplant anastomosis (HR:3.14; 95%CI:1.74,5.69; p<0.0

Diagnosis of catheter-associated candiduria is limited by absent colony-forming unit (CFU) thresholds and heterogeneous laboratory workflows, complicating patient management. Within a quality-improvement (QI) initiative we aimed to (1) implement and evaluate a standardised quantitative culture (QM) workflow for catheter urine, (2) compare QM with routine semi-quantitative culture (SQM) for CFU detection and reproducibility, and (3) assess whether visible urine aggregates predict Candida spp. presence. From February 2024 to February 2025, we analysed 222 yeast-positive urine samples from 74 cat

Candida tropicalis is a leading cause of invasive candidiasis in the Asia-Pacific region with reported crude mortality rates exceeding 50%. The rising prevalence of azole-resistant strains presents a significant clinical challenge. We analyzed 1,016 C. tropicalis clinical isolates collected over nine years from 27 hospitals across North India. Fluconazole resistance was detected in 5.1% (n = 52) of isolates, with cross-resistance observed to voriconazole in 55.7% and itraconazole in 44.2% of isolates. Multilocus sequence typing (MLST) analysis of global 1,630 isolates including 208 Indian and

Verrucous carcinoma of the esophagus (VCE) is a rare variant of squamous cell carcinoma that poses diagnostic challenges due to nonspecific endoscopic and histologic findings. Its typical Candida overgrowth and hyperkeratotic, non-malignant superficial layer, combined with biopsy results showing fungal hyphae and minimal cytological atypia, are often misleading. Consequently, VCE is frequently diagnosed at advanced stages, limiting curative treatment options and contributing to poor outcomes. Clinicians should consider VCE in patients with persistent verrucous or hyperkeratotic esophageal lesi
