Bioaccumulation, biotransformation and hormesis effects of 6PPD-Q in Eisenia fetida: Impacts on metabolism and gut microbiota.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPD-Q) is an oxidative byproduct of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine, a widely used tire antioxidant, and has emerged as an environmental pollutant of increasing concern. However, its environmental behavior and effects in soil ecosystems remain poorly characterized. This study focused on earthworm Eisenia fetida to reveal the bioaccumulation, biotransformation, and hormesis effects of 6PPD-Q. Findings indicated that 6PPD-Q mainly accumulated in intestinal tissues of E. fetida with concentration of 3465.2 ± 470.8 ng/

Pharmacokinetic and pharmacodynamic concepts in antibiotic therapy for clinicians.

Antibiotic efficacy depends on attaining adequate drug exposure at the site of infection while minimizing toxicity and resistance selection. This clinician-oriented review integrates pharmacokinetic determinants of exposure with pharmacodynamic drivers of bacterial killing. Emphasis is placed on the relevance of unbound concentrations, tissue distribution, and the limitations of MIC-based targets when interpreted without appropriate biological and microbiological context. Determinants of antibiotic performance are examined from an integrated perspective, considering drug-related properties, mi

The post-acute phase of new-onset refractory status epilepticus (NORSE) / febrile infection-related epilepsy syndrome (FIRES).

In this narrative review, we will summarize the existing literature describing long-term outcomes after new-onset refractory status epilepticus and febrile infection-related epilepsy syndrome. Febrile infection-related epilepsy syndrome is more common in children and has been described as a multiphasic presentation with a prodrome, an acute epileptic phase and a chronic epileptic phase. Important long-term sequelae after resolution of the acute phase of new-onset refractory status epilepticus and febrile infection-related epilepsy syndrome include survival and mortality, seizures, cognition an

CTDNEP1 Expression Is Associated With Subtype-dependent Prognostic Patterns in Renal Cell Carcinoma.

BACKGROUND/AIM: Renal cell carcinoma (RCC) comprises multiple histological subtypes, such as kidney renal clear cell carcinoma (KIRC) and kidney renal papillary cell carcinoma (KIRP), each with distinct molecular characteristics. Identifying novel genes involved in the onset and progression of these subtypes is crucial for the development of more effective treatments. This study investigates the role of CTD nuclear envelope phosphatase 1 (CTDNEP1), a phosphatase-encoding gene, in KIRC and KIRP using multi-omics data from The Cancer Genome Atlas (TCGA). MATERIALS AND METHODS: We analyzed CTDNE

Systemic treatment paradigms for unresectable and advanced hepatocellular carcinoma in Europe.

The management of unresectable and advanced hepatocellular carcinoma (HCC) has been transformed by the introduction of immune checkpoint inhibitor-based combinations, which have improved response and survival and expanded first-line treatment options. The availability of multiple effective regimens has introduced new challenges in therapeutic decision making. In the absence of validated predictive biomarkers or head-to-head comparisons, first-line treatment selection remains largely guided by clinical characteristics, contraindications and anticipated toxicity, while the optimal sequencing of

Clerodane Diterpenes and Structural Modification of 12-Oxo-Hardwickiic Acid from the Roots of Croton krabas: α‑Glucosidase Inhibitory Activity, Molecular Docking, and ADMET.

The roots of Croton krabas have traditionally been used as a water decoction for treatment of bloating and flatulence. The preliminary bioactivity screening revealed that the crude n-hexane and EtOAc extracts exhibited significant α-glucosidase inhibitory activity. Accordingly, the first phytochemical investigation of the roots of C. krabas led to the isolation of two previously undescribed clerodane diterpenes (1 and 2), together with eight known compounds (3-10). The structures and stereochemistry were elucidated by extensive analysis of spectroscopic data (UV, IR, 1H NMR, 13C NMR, and 2D NM

The role of pharmacogenomics in epilepsy treatment: advancements in personalized medicine.

Epilepsy is a diverse and often severe neurological disorder characterized by frequent and unpredictable seizures caused by the simultaneous discharge of brain cells in one or both hemispheres. Recently, there has been a significant shift in the approach to healthcare, moving from a standardized model to a more personalized and targeted model. This transformation has been made possible by advances in pharmacogenomics. It has great potential in customizing treatments based on the specific needs of patients, and this is particularly evident in the management of epilepsy. With ongoing research, t

Recounting Workforce: The Impact of Multi-licensure on Behavioral Health Provider Estimates.

Accurate behavioral health workforce estimates are essential for workforce planning, shortage designation, and policy development. However, workforce reports often rely on license counts rather than unique providers, which can inflate workforce estimates when clinicians hold multiple licenses. This study examined the prevalence and geographic distribution of multi-licensure among Nevada behavioral health providers and assessed its impact on workforce estimates. A cross-sectional analysis was conducted using administrative licensure data from Nevada licensing boards for Marriage and Family Ther

Antipsychotic Polypharmacy in Hospitalised Adolescents and Young Adults: Predictors, Clinical Correlates and Post-Discharge Outcomes.

BACKGROUND: Antipsychotic polypharmacy (APP), defined as the concurrent use of two or more antipsychotics, is common in real-world psychiatric practice despite guidelines favouring monotherapy. While APP is linked to greater illness severity and behavioural dysregulation in adults, its correlates and outcomes in adolescents and young adults are less understood. METHODS: This retrospective chart review examined psychiatrically hospitalised patients under 25 prescribed at least one antipsychotic for psychosis between 2018 and 2022 at a single academic medical center (N = 229). APP was defined a

Biosynthesis mechanisms, antimicrobial effects, and characterization of silver nanoparticles produced by bacterial and actinomycete isolates derived from a gold mine.

BACKGROUND: Silver nanoparticles (AgNPs) are widely studied for their antimicrobial properties, and microbial synthesis offers an eco-friendly and scalable alternative to conventional methods. While Streptomyces spp. are well-known producers, little is known about AgNP biosynthesis by microorganisms adapted to metal-rich environments such as gold mines, which may represent a largely unexplored source of microorganisms with potential for AgNP biosynthesis. This study investigated the biosynthetic potential of bacterial and actinomycete isolates recovered from the Sukari Gold Mine, Egypt, with a
