Epigenetic markers as predictors of neurotoxicity and abuse potential of psychotropic drugs: integration of epigenetic modifications in New Approach Methodologies.

Neurotoxicity and abuse potential are strongly associated with psychotropic drugs. Although animal models have been used for decades to investigate different forms of toxicity, including neurotoxicity and abuse potential, increasing efforts have been directed toward overcoming their inherent limitations. In this context, the development of New Approach Methodologies (NAMs), aimed at improving chemical safety assessment through more protective and physiologically relevant models, has emerged as a promising strategy to predict the neurotoxic potential of drugs while contributing to the replaceme

Do question type and topic affect the performance of large language models in answering periodontology questions? A comparative study.

BACKGROUND: There is increasing interest and ongoing debate about the integration of artificial intelligence and the large language models it supports in health science education. The performance of these models may vary depending on educational domains, clinical branches, complexity of questions, and question formats. This study aimed to compare the performance of ChatGPT-5, Gemini 2.5 Flash, and Copilot on periodontology questions and to determine whether their performance was affected by question type and topic. METHODS: One hundred thirty-four multiple-choice periodontology questions from

Population-specific transcriptomic bioactivity assessment of perfluorooctanoic acid using human liver spheroids and physiologically based toxicokinetic modeling.

Perfluorooctanoic acid (PFOA) has been widely used in industrial and consumer products and was recently classified as a Group 1 carcinogen by the International Agency for Research on Cancer. However, uncertainties remain regarding transcriptomic bioactivity thresholds associated with liver effects and their variability across clinically relevant populations. In this study, previously published transcriptomic data obtained following 24-h and 10-day exposure of primary human liver spheroids (LS) to PFOA were integrated into a physiologically based toxicokinetic (PBTK), population-specific framew

Antithetical roles of tau in hippocampal long-term synaptic depression in rats versus mice.

Synaptic dysfunction is a primary feature of tauopathies, a group of major neurodegenerative diseases that includes Alzheimer's disease. However, the normal synaptic functions of tau are poorly understood. Since rats provide advantages over mice for understanding human tau, such as expressing six tau isoforms and showing more robust signs of tau pathology, as well as offering a richer behavioural repertoire, we developed a tau knockout (Mapt KO) rat line. We primarily investigated the impact of tau elimination on long-term synaptic depression (LTD)-a form of synaptic weakening that tau mediate

Force-driven single-molecule transformations through tip-mediated stabilization of transition states.

Mechanochemistry uses mechanical forces to drive chemical transformations, offering an energy-efficient alternative to traditional thermal, optical, or electrical reaction pathways. At the single-molecule scale, controlling structural changes with mechanical stimuli is fundamental for developing nanoscale devices and understanding chemical reactivity. In conventional single-molecule reactions driven by light or electrical currents, energy is supplied through one-way excitation processes, requiring the single-event energy input to exceed the activation energy barrier. Here we show that mechanic

Ten rules for identifying horizontal gene transfers in deep plant phylogenies.

Horizontal gene transfer (HGT) is one of the most intriguing aspects of genome evolution, challenging classical views of vertical inheritance. Growing genome availability across the tree of life offers opportunities to discover HGT, yet recent reanalyses suggest many reported cases may not withstand scrutiny. Detecting ancient transfers is especially difficult, as phylogenetic signal erodes over time and analytical choices strongly shape conclusions. Here we propose 10 practical rules for evaluating HGT in deep plant phylogenies, organised around three themes: genomic foundation, rigorous anal

Expression Profiling of a Recombinant Inbred Line Panel Identifies the dod-19 Gene as a Regulator of Ethanol Response Behaviors in C. elegans.

BACKGROUND: Variation in gene expression is hypothesized to be a major source of phenotypic variation underlying the liability for alcohol misuse. Caenorhabditis elegans (C. elegans) has proven to be a valuable model in the identification of genes that modify behavioral responses to alcohol. Genetic variation in wild strains represents allelic differences that are tolerated by natural selection pressures and, therefore, are more likely to model the allelic differences that are present in humans compared with laboratory-generated mutations. METHODS: We examined the influence of natural alleles

Transgenerationally inherited incisor aberration mediated by stress in two isolated populations of Microtus hartingi vole.

Environmental and social stress (SS) can alter phenotypic expression at both individual and population levels. We examined the effects of ancestral SS, imposed through forced polygyny, on reproductive success and maxillary incisor overgrowth in 2 isolated populations of Microtus hartingi vole: from Central Anatolia (CAP) and Eastern Rhodope (RP). In addition to ancestral SS, CAP voles were exposed to environmental stress during prenatal, lactation, and early-life periods. Under control conditions, the prevalence of incisor overgrowth was at a stable rate of ∼16.7% in RP and occurred very rarel

Assessment of teachers' knowledge, awareness, and response to students with asthma in Greater Cairo schools.

Bronchial asthma remains a significant public health challenge, especially among school-aged children. Schoolteachers are often the initial responders to asthma attacks, making their role vital for early symptom recognition and first aid provision. Therefore, this study aimed to explore the knowledge, awareness, and response of schoolteachers to students with asthma. Between March 2024 and October 2024, a self-administered questionnaire was created on Google Forms and distributed to schoolteachers through WhatsApp groups of private and public schools across different districts in Greater Cairo

Targeting Artemis sensitizes acute lymphoblastic leukemia cells to topoisomerase 2 poisons by disrupting DNA-PKcs-dependent repair.

Topoisomerase 2 (Top2) poisons are widely used in cancer therapy but are associated with toxicity and secondary malignancies. Top2 adduct removal requires endonuclease activity prior to repair of the resulting DNA double-strand break (DSB). We show that the non-homologous end joining (NHEJ) enzyme Artemis is a key player in the process and a major target for treatment of human B-cell acute lymphoblastic leukemia (ALL). An Artemis knockout is sensitive to etoposide treatment at nanomolar levels and has significantly more unrepaired DNA DSBs. Inhibition of the Artemis activator, DNA-dependent Pr

Navigating anticoagulation in heart transplantation with thrombin-antithrombin complex: when conventional monitoring fails.

BACKGROUND: Heart transplant recipients often face the clinical dilemma of coexisting bleeding and thrombotic risks postoperatively. Conventional anticoagulation monitoring parameters (e.g., platelet count, fibrinogen, D‑dimer, and anti‑Xa activity) frequently exhibit limitations in evaluating complex postoperative patients, including delayed response, poor sensitivity, and lack of clear target ranges. This case report describes the use of thrombin-antithrombin complex (TAT) to guide anticoagulation management for complex thrombotic events after heart transplantation and explores its potential

Dactolisib outperforms Rapamycin in potentiating apoptosis induced by XPO1, Bcl-2 and Mcl-1 blockade in preclinical glioblastoma models.

Glioblastoma resists therapy through interconnected survival programs. Combined XPO1, Bcl-2, and Mcl-1 inhibition (Eltanexor, Venetoclax, A1210477; EVA) enhances the efficacy of chemotherapy, yet resistance persists. We hypothesized that PI3K/AKT/mTOR (PAM) signaling drives adaptive escape. TCGA/GTEx analyses and immunohistochemistry indicated PAM pathway activation in glioblastoma, with AKT significantly overexpressed, whereas PIK3CA showed a clear trend towards higher expression, although it did not reach statistical significance. qPCR analysis showed that PAM-component expression was highes

Cytarabine-based mobilisation yields higher CD34+ cell counts than cyclophosphamide regardless of prior daratumumab exposure in multiple myeloma.

INTRODUCTION: Anti-CD38-based quadruplets such as daratumumab-bortezomib-lenalidomide-dexamethasone (D-VRd) or daratumumab-bortezomib-thalidomide-dexamethasone (D-VTd), where lenalidomide is not reimbursed, are the internationally recommended first-line treatment for transplant-eligible myeloma. There is ongoing debate about whether anti-CD38 exposure impairs stem cell collection. Most studies compare daratumumab-containing with non-daratumumab induction whilst keeping the mobilisation strategy constant. None have compared two mobilisation agents against each other in both daratumumab-exposed
