Awareness and knowledge of obstetricians about prenatal findings of inherited metabolic disorders.

This study aimed to assess obstetric healthcare providers’ awareness and knowledge of IMDs and their prenatal manifestations. Participants were surveyed using an online questionnaire about clinical and imaging features observed in fetuses and neonates with IMDs, their approach to clinical management when an IMD is suspected in the fetus, and the management of pregnant women with IMDs. 270 obstetricians participated, with a mean age of 32.5 ± 7.1 years. Most participants (81.5%) had less than 5 years of experience. The more experienced group was more likely to consider their knowledge of antena

Integrating LNA-qPCR and full-length SMN1 sequencing for precision SMA carrier screening: large-scale validation in 30,849 individuals.

BACKGROUND: Spinal muscular atrophy (SMA) is an autosomal recessive neuromuscular disorder caused primarily by biallelic SMN1 loss-of-function mutations, most commonly homozygous deletions of exon 7. While large-scale carrier screening is critical for prevention, comprehensive population-level data remain limited in mainland China. METHODS: We conducted a prospective, large-scale SMA carrier screening study in Hunan Province, China, from September 2020 to September 2025. The cohort included 30,733 pre-pregnancy and early-pregnancy individuals without an SMA family history across 14 cities. SM

Utility of long-read sequencing to delineate a rare large deletion of beta-globin gene which escaped Sanger sequencing at prenatal diagnosis in a family clustered with hereditary persistence of fetal hemoglobin.

OBJECTIVE: Thalassemias, including alpha and beta thalassemia, are the most common monogenic inherited Mendelian disorder in Taiwan. The mutation spectrums of alpha and beta thalassemia are well known in the Taiwanese population. Among them, mutations with the format of large deletions are more prevalent in alpha thalassemia whereas point mutations or small insertion/deletions (indels) are much more prevalent in beta thalassemia. Hereditary persistence of fetal hemoglobin (HPFH), a clinical subtype of beta thalassemia, is a relative uncommon condition during prenatal diagnosis in the Taiwanese

Foetal achondroplasia: Prenatal diagnosis, outcome and perspectives.

BACKGROUND: Achondroplasia, due to a specific pathogenic variant in FGFR3, is the most common viable skeletal dysplasia and the diagnosis is mostly done in the prenatal period. Since 2021, the use of Vosoritide, a specific treatment for achondroplasia, validated in phase 3 placebo-controlled trials, has been recommended to significantly increase the height of children and infants. In the light of these new therapeutic prospects, a complete understanding of the pathophysiology of skeletal damages occurring from foetal life is required. OBJECTIVES: To describe foetal imaging and the antenatal a

Prenatal diagnosis of skeletal dysplasias using whole exome sequencing in China.

BACKGROUND: Skeletal dysplasias account for nearly 10% of fetal structural malformations detected by ultrasonography. This clinically heterogeneous group of genetic anomaly includes at least 461 genetic skeletal disorders with extreme clinical, phenotypic, and genetic heterogeneities, thus, significantly complicates accurate diagnosis. Researches have used whole exome sequencing (WES) for prenatal molecular diagnoses of skeletal dysplasias, however, data are still limited. METHODS: DNA extracted from umbilical cord blood or amniocytes from fetuses suspected of skeletal dysplasias based on ult

Genetic diagnoses and associated anomalies in fetuses prenatally diagnosed with esophageal atresia.

Esophageal atresia (EA) is a congenital anomaly occurring in 2.3 per 10,000 live births. Due to advances in prenatal imaging, EA is more readily diagnosed, but data on the associated genetic diagnoses, other anomalies, and postnatal outcome for fetuses diagnosed prenatally with EA are scarce. We collected data from two academic medical centers (n = 61). Our data included fetuses with suspected EA on prenatal imaging that was confirmed postnatally and had at least one genetic test. In our cohort of 61 cases, 29 (49%) were born prematurely and 19% of those born alive died in the first 9 years of
