Utilization of human amniotic membrane (HAM) in tympanoplasty: a systematic review and meta-analysis.

Tympanic membrane perforation is a common otologic disorder. In cases where spontaneous closure does not occur, or when the perforation is associated with clinically significant symptoms that adversely affect quality of life, surgical repair is often indicated. This review and meta-analysis aimed to synthesize and critically appraise the current evidence on the use of human amniotic membrane (HAM) as a graft material in tympanoplasty. A systematic literature search was conducted in multiple databases for studies published up to November 2025. The following search terms were used in titles and

Progesterone-KISS1 axis impairs amniotic epithelial cell function and promotes premature rupture of membranes.

BACKGROUND: Premature rupture of membranes (PROM) is a major cause of preterm birth and perinatal morbidity. Disruption of fetal membrane integrity is central to PROM pathogenesis; however, the molecular mechanisms underlying membrane weakening remain incompletely understood. METHODS: Proteomic analysis was performed on fetal membrane tissues from patients with early PPROM, late PPROM, full-term PROM, and full-term normal pregnancies. An LPS-induced infection-associated preterm birth mouse model was established to evaluate fetal membrane pathology, hormone levels, inflammatory responses, and

[Interaction between oxidative stress and inflammatory processes in fetal membranes: One Health].

The balance between oxidative mechanisms and inflammatory responses is essential for the proper progression of pregnancy, as it preserves the functionality and integrity of the fetal membranes. Among them, the amnion plays a key protective role by regulating maternal-fetal exchange and interpreting chemical and mechanical signals from the uterine microenvironment. However, various adverse stimuli, particularly bacterial infections mediated by lipopolysaccharide (LPS), environmental contaminants, sustained physiological stress, or metabolic disturbances, can disrupt this homeostasis, increasing

Comparative Effectiveness of Amniotic and Chorionic Grafts in the Treatment of Lower-Extremity Diabetic Ulcers Using U.S. Medicare Real-World Evidence (2018-2022): A Retrospective Observational Cohort.

OBJECTIVE: Lower-extremity diabetic ulcers (LEDUs) affect more than 500,000 U.S. Medicare beneficiaries each year. Dehydrated human amnionic and chorionic allografts (DHACAs) are clinically effective complements to standard of care (SoC; e.g., surgical debridement, offloading, infection, and moisture control) when treating LEDUs. However, Medicare and commercial payer coverage have restricted access to DHACAs. Our objective was to compare the effectiveness of DHACAs versus SoC among Medicare beneficiaries with LEDUs for reduction of adverse outcomes such as mortality, recurrency, and major amp

Beyond transplants: current and future therapeutic potential of amniotic membrane extract (AME) in ophthalmology.

There is no established consensus or standardized method for the preparation of amniotic membrane extract (AME). Consequently, various preparation, preservation, and sterilization techniques have been employed. To obtain AME rich in bioactive components with high therapeutic efficacy, each step of the preparation process is of critical importance. The appropriate procurement of the amniotic membrane minimizes the risk of infection transmission and reduces inter- and intra-donor variability. For the subsequent extraction process, different approaches are utilized due to factors such as laborato

Clinical outcomes of lyophilised human amnion/chorion membrane in treatment of hard-to-heal diabetic foot ulcers in complex cases: a case series.

OBJECTIVE: Diabetic foot ulcers (DFUs) are a common and severe complication of diabetes, characterised by high morbidity, recurrence and risk of amputation. Hard-to-heal (chronic) DFUs often fail to respond to standard of care (SoC), necessitating advanced interventions. Lyophilised human amnion/chorion membrane (LHACM) is a trilayer placental allograft that provides extracellular matrix support, growth factors and anti-inflammatory properties to promote wound closure. This case series evaluates the effectiveness of LHACM as an adjunct to SoC in treating hard-to-heal DFUs unresponsive to conve

The Role of Beta-Defensin 2 in Preventing Preterm Birth with Chorioamnionitis: Insights into Inflammatory Responses and Epithelial Barrier Protection.

Antimicrobial peptides, such as beta-defensin 2 (BD2), are vital in controlling infections and immune responses. In this study, we investigated the expression and role of BD2 in the amniotic membrane and human amniotic epithelial cells (hAECs) from patients with preterm birth and chorioamnionitis, focusing on its regulation of inflammatory cytokines and its protective effect on the epithelial barrier. Our results show increased BD2 expression in chorioamnionitis, and Lipopolysaccharide (LPS)-induced inflammation increased BD2 release from hAECs in a dose- and time-dependent manner. BD2 treatme

Targeting Lung Damage: Amniotic Mesenchymal Stem Cells Mitigate Lipopolysaccharide-Induced Acute Lung Injury via Multiple Signaling Pathways.

Acute lung injury (ALI) is a life-threatening condition triggered by pneumonia, viral infections, or physical trauma. It manifests clinically as progressive respiratory failure and refractory hypoxemia. Using a lipopolysaccharide (LPS)-induced acute lung injury mouse model, we demonstrated that amniotic mesenchymal stem cells (AMSCs) exhibit robust reparative and anti-inflammatory properties. Our analysis encompassed inflammatory mediators; histological damage; tight junction integrity; epithelial-mesenchymal transition (EMT); and the TGF-β/Smad, TLR4/NF-κB/MAPK, pyroptosis, and apoptosis sign

3D Printed Human Amnion-Based Bioactive Hybrid Dressings for Effective Management of Complex Infected Wounds.

Chronic wounds are often afflicted with persistent infection, excessive exudate accumulation, and delayed healing, leading to prolonged hospitalization. Excess moisture overhydrates the wound, promotes infection, and causes edema. Peri-wound skin may develop rashes, immersion injuries, and epidermal detachment. Nutrient-rich exudates foster microbial growth, increasing the infection risk. High bacterial loads lead to crust formation, continuous leakage, and foul odor, further complicating healing. To address this challenge, we developed a 3D printed amnion-based hybrid dressing comprising a re
