Amniotic membrane promotes doxorubicin potency by suppressing SH-SY5Y neuroblastoma cell angiogenesis.

BACKGROUND: Doxorubicin (DOX) remains a mainstay for neuroblastoma (NB) treatment, but side effects hamper efficacy. We previously showed that DOX induces SH-SY5Y NB cell angiogenesis via the PHD-2/HIF-1α axis. Adjuvant therapies offer a promising avenue to improved outcomes. Human amniotic membrane (hAM) extract (hAME) consists of various proteins that exhibit anti-cancer and anti-angiogenic properties. This study investigates hAME as a potential adjuvant for targeting NB angiogenesis when combined with DOX. METHODS: We used cellular, molecular, and biochemical assays to evaluate the antitum

Human amniotic epithelial stem cells-conditioned media (hAECs-CM) reduces inflammation and promotes wound healing in a mouse burn model.

This study aimed to assess the impact of human amniotic epithelial stem cells-conditioned medium (hAECs-CM) on the healing of burn wounds and the reduction of inflammation in a mouse model of deep second degree burns. The findings indicated that hAECs-CM markedly enhanced the primary metrics of burn wound healing within a brief duration of 14 days. The rate of wound healing was substantially greater than that of the control group following 21 days of treatment with daily administration of hAECs-CM. The CD31 concentration was markedly elevated in burn tissues treated with hAECs-CM, whereas the

Application of Cryopreserved Decellularized Human Amniotic Membrane for Healing of Treatment-resistant Cesarean Section Wounds: A Case Series.

Considering the increase in cesarean surgeries in recent years and, therefore, the complications related to this type of surgical wound, attention has been drawn to treatment methods such as biological wound dressings. In this case series, the authors describe the treatment of infected cesarean wounds with impaired healing in 4 women. For this aim, human placentas were collected under sterile conditions and amniotic membranes were separated from the placenta. To decrease immunogenic response, the membrane was decellularized using 1% Triton-x-100, 1 mmol/L EDTA, and Trypsin 0.05. Then, using gl

Spontaneous partial chorioamniotic membrane separation followed by the formation of amniotic bands: a case report.

BACKGROUND: Chorioamniotic membrane separation (CMS) is associated with various adverse pregnancy outcomes, including miscarriage, preterm labor, stillbirth, amniotic band syndrome, etc. Spontaneous CMS is extremely rare, especially followed by the formation of amniotic bands. At present, there is no standardized monitoring and treatment protocols for CMS. CASE PRESENTATION: We present an unusual case of spontaneous partial CMS with amniotic bands at 27+6weeks of gestation in a 34-year-old woman. About one month later, an emergency cesarean section was performed because of unavoidable preterm

Effect of autologous amniotic membrane and fluid on wound healing and complications of cesarean section: Study protocol of a factorial randomized controlled trial.

BACKGROUND: Cesarean section rates have risen globally. While a cesarean section offers benefits, it is associated with complications like infection, fluid collection, and wound dehiscence. The psychological impact of cesarean section scars can also affect women's quality of life. Amniotic membrane and fluid have shown promise in enhancing wound healing due to their rich content of growth factors and nonimmunogenic properties. This study aims to assess the effects of autologous amniotic membrane and amniotic fluid on wound healing, surgical site infection, pain, and complications following ces

Application of Amniotic Membrane to Effectively Treat Chronic Cervicitis: A Case Series.

Chronic cervicitis is a clinical syndrome characterized by persistent inflammation of the cervical epithelium. Women with chronic cervicitis often experience symptoms such as vaginal discharge, dyspareunia, post-coital bleeding, and fatigue. The treatment of chronic cervicitis is controversial, particularly in cases where no specific pathogenic agent be identified. Natural wound dressings with human amniotic membranes have been successfully used in tissue healing procedures. Thus, this approach may be a potential therapeutic option for patients suffering from chronic cervicitis. This case seri

Biofabrication of small-diameter vascular graft with acellular human amniotic membrane: a proof-of-concept study in pig.

Expanded polytetrafluoroethylene (ePTFE) grafts are Food and Drug Administration approved and effective for large vessel surgeries but face challenges in smaller vessels (Inner Diameter, ID ⩽ 6 mm) due to reduced blood flow and higher risks of thrombosis, stenosis, and infection. This study developed a vascular graft with an ID of 6 mm from decellularized human amniotic membrane (DAM graft) and compared its performance to ePTFE grafts in a porcine carotid artery model for one month. DAM grafts retained key extracellular matrix structures and mechanical properties post-decellularization, with c

Surgical Approaches for the Management of Peripheral Ulcerative Keratitis With Corneal Perforation.

BACKGROUND: Corneal perforation is a serious complication of peripheral ulcerative keratitis (PUK), which requires prompt control of the inflammation while restoring the integrity of the globe. This can often be challenging and may require 1 or a more surgical approaches. METHODS: We report the management of 3 cases of PUK with repeated perforations and failed attempts at corneal gluing. The first case had a combination of an acute hydrops with perforation in a patient with longstanding eczema and undiagnosed keratoconus. After multiple failed attempts at gluing, a partial thickness sclerocor

Severe Corneal Ulceration After Cosmetic Keratopigmentation.

PURPOSE: Keratopigmentation is a well-described procedure often used to address corneal opacities. Purely cosmetic keratopigmentation, in the absence of underlying corneal pathology has increased in popularity recently, with the intention of changing eye color appearance. In the context of this increased popularity, public knowledge and data regarding the risks of this procedure are lacking. METHODS: This study is a case report conducted at a single academic institution. RESULTS: Here we present a case of a patient who underwent purely cosmetic keratopigmentation and who developed severe cor
