Laennec Attenuates Alcohol-Induced Hepatic Steatosis and Oxidative Stress in a Murine Model.

BACKGROUND: Alcoholic liver disease (ALD) represents a major global health burden, encompassing a spectrum of hepatic abnormalities ranging from steatosis to cirrhosis and hepatocellular carcinoma. Laennec, a human placenta-derived hydrolysate, was evaluated for its therapeutic effects on alcohol-induced fatty liver in an experimental animal model. METHODS: Animals were pretreated with alcohol for 2 weeks, followed by the co-administration of alcohol and Laennec for 4 weeks. The study included four groups: normal control, alcohol-only, and low- and high-dose Laennec-treated groups. RESULTS:

Structural and Mechanical Changes to the Extracellular Matrix in Human Umbilical Arteries in Pregnancies Complicated by Gestational Diabetes.

The umbilical cord serves as a critical lifeline, connecting the fetus to the pregnant individual. Stillbirth, fetal death at or beyond 20 weeks' gestation, can occur due to developmental anomalies or complications of the umbilical cord. Gestational diabetes (GDM), which affects 8% of pregnancies in the United States, is associated with vascular abnormalities of the placenta and umbilical cord. While cellular and microstructural changes in the umbilical vasculature have been observed in pregnancies complicated by gestational diabetes, it is unknown if these influence the macroscale mechanical

Deficiency of Pdap1 Results in Embryonic Lethality and Diminished Vascularization.

Platelet-derived growth factor associated protein 1 (PDAP1) is implicated in various biological processes, including tumorigenesis and the apoptosis of mature B lymphocytes. However, its functions in organismal development and homeostasis remain poorly understood. To address this gap, we generated Pdap1 knockout mice to elucidate the physiological functions of PDAP1 in vivo. Notably, our findings indicate that the absence of Pdap1 results in early embryonic lethality by embryonic day 11.5, accompanied by a spectrum of developmental abnormalities, including growth retardation, enlarged pericard

Association of human chorionic gonadotropin and post-pregnancy persistent enhanced myometrial vascularity.

OBJECTIVE: To explore the role of circulating human chorionic gonadotropin (hCG) in the pathophysiology of enhanced myometrial vascularity (EMV) postpregnancy and the usefulness of determining hCG level in the clinical evaluation and management of this condition. METHODS: This was a retrospective observational case series of patients managed clinically at our centers who had postpregnancy EMV diagnosed using ultrasound by the observation of a dilated vascular web in the myometrium, with or without retained products of conception (RPOC), that persisted beyond 4-6 weeks following a failed or te

Current approaches to the diagnosis and management of retained products of conception and enhanced myometrial vascularity of the first trimester.

UNLABELLED: Introduction:Aim of this narrative review is to provide an update on diagnosis, management and outcome of RPOC and EMV diagnosed after first trimester pregnancy. Retained products of conception (RPOC) and enhanced myometrial vascularity (EMV) are considered complications following pregnancy and there is scarce agreement upon their management. Methods:A comprehensive, narrative, research was conducted. A literature review of relevant studies published between 2000 and 2025 was carried out using MEDLINE and the Cochrane library using the following terms: 'Arteriovenous Malformations'

Tunneling nanotubes contribute to Zika virus pathogenesis at the maternal-fetal interface in vivo.

Zika virus (ZIKV) infection during pregnancy can result in severe fetal outcomes, yet the mechanisms of transplacental dissemination remain incompletely defined. We previously showed that ZIKV induces tunneling nanotubes (TNTs), actin-rich intercellular conduits that enable direct cell-to-cell transfer of viral components. Here, we investigated the in vivo role of TNTs in ZIKV maternal-fetal transmission using complementary pregnancy models. A TNT-deficient ZIKV mutant (ZIKVΔTNT), harboring a change between residues 40 and 52 of the nonstructural protein 1 (NS1), showed markedly reduced viral

Sex-Dependent Placental Neuroendocrine Disruption by Maternal Arsenic Exposure: From Neurotransmitter Dysregulation to Neurobehavioral Outcomes.

Epidemiological evidence suggests that developmental arsenic (As) exposure increases behavioral abnormalities in offspring. However, how placental neuroendocrine function and sex-specific responses contribute to these effects remains poorly understood. Here, we demonstrate that maternal As exposure induces trophoblastic damage, vascular disruption, and placental labyrinth thinning, leading to placental dysplasia, fetal growth restriction, and sex-specific behavioral outcomes in offspring. Integrated transcriptomic and neurobehavioral analyses revealed sex-dependent alterations primarily involv

Placenta accreta and uterine arteriovenous malformation following medically treated missed abortion in patient with undiagnosed cesarean scar pregnancy.

Cesarean scar pregnancy (CSP) is a rare condition, accounting for 6% of ectopic pregnancies, with a rising incidence. If not diagnosed early, it may lead to severe complications such as hemorrhage, need for surgery, placenta accreta, and arteriovenous malformation (AVM). Treatment consists of expectant management, medical, or surgical regiments. A 31-year-old woman (G3P2) with one previous cesarean section presented for early pregnancy evaluation. Ultrasound initially showed an intrauterine pregnancy, but follow-up confirmed a missed abortion, and mifepristone/misoprostol regimen was initiated

Clinical and multi-omics characterisation of early neurodevelopmental disorders associated with critical congenital heart disease: the prospective cohort CATAMARAN neonatal study protocol.

INTRODUCTION: Critical congenital heart disease (CHD) is associated with neurodevelopmental disorders, recognised as the most common long-term morbidity in affected children. In critical CHD, that is, CHD requiring cardiac surgery within the first 3 months of life, 30%-50% of children have lower developmental scores. Therefore, early identification of at-risk infants is crucial, yet there is no scientifically evaluated care programme in France. This study aims to evaluate early neurodevelopmental status in infants with prenatally diagnosed critical CHD and to determine how intrinsic susceptibi

Placental pathology in fetuses with and without congenital heart defects.

INTRODUCTION: Placental histopathologic evaluation of pregnancies affected by congenital heart defects (CHDs) can provide insight into the mechanism connecting placental and fetal vascular systems. The objective of this study was to evaluate the prevalence of maternal vascular malperfusion (MVM) and fetal vascular malperfusion (FVM) lesions of the placenta in pregnancies with and without fetal CHD on blinded pathologic examination. METHODS: This is a case-control study of pregnancies complicated by CHD at our institution from January 1, 2011 to June 25, 2021. Controls were matched to cases in

Mechanisms of Hexavalent Chromium-Induced Reproductive Toxicity: A Focus on the Ovary and Placenta.

Hexavalent Chromium (Cr(VI)) is a Group A carcinogen, mutagen, and teratogen. Cr(VI) has been used by more than 50 industries, and its contamination of drinking water is widespread across the United States (U.S.). Epidemiological data of women who lived in Willits, California, U.S., indicate that environmental exposure to Cr(VI) adversely affects pregnancy outcomes and the health of their immediate offspring, resulting in a low birth rate, pregnancy loss, and spontaneous abortion, and their children (F1 offspring) experienced birth defects. However, the molecular mechanisms behind Cr(VI)-induc

Feasibility and safety of cellular therapy for in-utero repair of myelomeningocele (CuRe Trial): a first-in-human, phase 1, single-arm study.

BACKGROUND: The Management of Myelomeningocele Study (MOMS) trial established the benefit of in-utero repair of myelomeningocele, with a decreased need for ventriculoperitoneal shunt placement. However, although there was some improvement of motor function, over half of the patients were unable to ambulate independently. Live placenta-derived mesenchymal stem cells (PMSCs) seeded on an extracellular matrix have shown promise in rescuing neurological function in the fetal ovine model of myelomeningocele. We aimed to evaluate the safety of this novel, living, stem cell product to augment the pre

Maternal-fetal immune conflict contributes to male-specific impairments in a mouse model of neurodevelopmental disorders.

Autism spectrum disorder (ASD) arises from genetic and environmental risk factors. One environmental factor, maternal immune activation (MIA)-in which pathogenic infection during pregnancy increases ASD risk in offspring-disproportionately affects males. However, the basis for this male-specific vulnerability and the mechanisms by which inflammatory signals cross the maternal-fetal interface to affect the male embryo remain largely unknown. Using the poly(I:C) mouse model of neurodevelopmental disorders, we characterize fetal, placental, and amniotic changes occurring within 24 hours of MIA. W

Advances in the mechanisms of histone modification-mediated developmental toxicity induced by organophosphorus pesticides.

Organophosphate pesticides (OPPs) are widely used and common environmental contaminants. Certain OPPs can cross the placenta and reach embryonic tissues, and prenatal exposure has been associated with developmental delays, structural abnormalities, and potential multigenerational effects. Beyond the classic mechanism of acetylcholinesterase inhibition, emerging evidence indicates that epigenetic dysregulation may contribute to OPP-induced developmental toxicity. However, the mechanistic evidence remains fragmented across different model systems and experimental designs, and a coherent framewor

Embryotoxic Effects of Monosodium Methyl Arsenate (MSMA) on Gallus gallus: Morphological and Morphometric Evidence of Developmental Disruptions.

Anthropogenic activities and the large volume of compounds released into the environment are sources of contamination for plants, animals, and humans. The herbicide monosodium methyl arsenate (MSMA) ranks among the best-selling agricultural pesticides in Brazil. It is classified as highly toxic, containing arsenic, which crosses the placenta, reaching the developing embryo. To assess the toxic effects of these chemicals, model organisms such as chicken embryos are used, as they share phylotypic features with human embryos in the early stages of development, allowing extrapolation between speci

Cuproptosis: a novel mechanism linking placental metabolic stress to trophoblast injury in preeclampsia.

Preeclampsia (PE) is characterized by placental hypoxia, metabolic stress, trophoblast dysfunction, and impaired spiral artery remodeling, but the mechanisms linking these abnormalities remain incompletely understood. Cuproptosis, a recently identified form of regulated cell death driven by copper-dependent aggregation of lipoylated tricarboxylic acid cycle proteins, may provide a mechanistic link between placental metabolic stress and trophoblast injury. Here, we propose a hypothesis in which hypoxia-induced glycolytic reprogramming and lactate accumulation promote histone H3K18 lactylation,

Maternal-fetal interface abnormalities in unexplained recurrent pregnancy loss: mechanisms, models, and a framework for subtype identification.

Recurrent pregnancy loss (RPL) is a heterogeneous reproductive condition, and unexplained recurrent pregnancy loss (URPL) denotes repeated pregnancy losses for which no established cause is identified after standard evaluation. Recurrent miscarriage (RM) and recurrent spontaneous abortion (RSA) are overlapping terms used in the literature, whereas recurrent implantation failure (RIF) is a related but distinct infertility context. In this Mini Review, URPL is treated as an exclusion-based clinical category rather than a single disease entity. This review examines the hypothesis that a subset of

Altered Glutamine/α-Ketoglutarate Metabolism Associates with Decidualization Defects in Recurrent Implantation Failure and is Ameliorated by α-Ketoglutarate Supplementation in Mice.

Endometrial decidualization is a critical determinant of embryo implantation, and disruption in this process can lead to implantation failure. While various metabolic pathways regulate decidualization, the detailed metabolic characteristics and the mechanisms behind this remain unclear. Here we identified that during the process of decidualisation, the concentrations of glutamine and α-ketoglutarate (α-KG) were significantly elevated. Impaired glutamine/α-KG metabolism disrupts decidualization and triggers metabolic reprogramming characterized by decreased oxidative phosphorylation and glycoly

Ischemic placental disease and hospitalization for neurological and psychiatric disorders.

BACKGROUND: Ischemic placental disease, a syndrome that comprises preeclampsia, placental abruption, and fetal growth restriction, is characterized by ischemic, endothelial, and microvascular dysfunction. These processes, in turn, may serve as sentinel markers of maternal brain vulnerability, resulting in structural abnormalities, altered neuronal connectivity, and impaired neurogenesis. OBJECTIVE: To estimate the burden of postpartum readmissions for neurological and psychiatric disorders (hereafter referred to as brain disorders) in women associated with ischemic placental disease in the sa
