Convergent Synthesis of a Desmethyl Analogue of Amycomycin.

The convergent synthesis of a desmethyl analogue of amycomycin, the aglycon of vancoresmycin, is presented. The central challenge of the convergent strategy was an initial aldol condensation approach, which failed on advanced substrates due to steric hindrance, prompting a revised coupling strategy. The successful route employed nucleophilic vinyl addition, complemented by a Boron-Wittig olefination to construct sterically congested vinyl boronates. A late-stage O- to C-acyl transfer on a complex 3-acyl tetramic acid was essential for the efficient assembly of the complete skeleton of desmethy

Generalized Form of Multilayer Adsorption Isotherm and Its Application for Determining Surface Areas of Adsorbents.

We propose a new approach to determining surface areas of macro-porous adsorbents by considering multilayer adsorption isotherm in generalized form. Model assumptions behind this form are significantly less restrictive than those used in conventional approaches. The BET method "continues to be the most widely used procedure for evaluating the surface area of porous and finely-divided materials, in spite of the weakness of its theoretical foundations" (quote fromThommes, M.; et al.Pure Appl. Chem.2015, 87, 1051-1069.). BET and corrected BET isotherms are based on a number of well known restrict

Separating Detection Performance from Thermodynamic Interpretability in Quantum-Capacitance Affinity Assays.

Quantum capacitance, Cq, is proportional to the electronic density of states of a redox monolayer and is used as a label-free transducer in affinity sensing. Here, we develop a theoretical framework and apply it to a reanalysis of previously published impedance data from a DNA-receptive interface; no new experimental data are reported. Under the isoscopic condition, the charging energy per accessible redox state is fixed at 2kBT, so that the analyte dependence of the ensemble Cq resides in the number of accessible states. Binding renders a fraction χ of these states electrochemically inaccessi

Dynamic multi-scale topology learning and cross-modal graph fusion for autism spectrum disorder diagnosis.

Multi-modal fusion methods have demonstrated potential in integrating complementary information from different data modalities. However, existing methods often rely on static fusion strategies, failing to capture the heterogeneous interactions across modalities. To address this challenge, we propose the dynamic multi-scale topology learning and cross-modal interaction graph fusion network (DMTL-Net) for autism spectrum disorder (ASD) diagnosis. The model is designed to extract distinctive and complementary features from brain functional and structural networks while capturing their dynamic top

Nasal high-frequency oscillatory ventilation as primary respiratory support in preterm infants with respiratory distress syndrome: current evidence and clinical implications.

Continuous positive airway pressure (CPAP) is the standard initial therapy for respiratory distress syndrome (RDS) and associated with failure rates of up to 50% in extremely preterm infants. Nasal intermittent positive pressure ventilation (NIPPV) has demonstrated consistent benefits over CPAP in reducing respiratory failure and intubation. Nasal high-frequency oscillatory ventilation (nHFOV) has also shown reduced treatment failure compared with CPAP, particularly in extremely preterm infants. However, direct head-to-head comparisons between nHFOV and NIPPV as primary support are lacking, an

Accounting for Competing Events in Longitudinal Studies of Geriatric Outcomes.

BACKGROUND: In time-to-event analyses of geriatric outcomes, competing events such as mortality are common but are often handled inappropriately, leading to biased risk estimates and potentially incorrect inferences. We present a case study illustrating the consequences of failing to account for competing events and review how such analyses are conducted in recent JAGS publications. METHODS: For the case study, we analyzed data from the Precipitating Events Project to estimate the risk and rate of two geriatric outcomes-injurious falls and restricted activity-using methods that appropriately

Common peroneal nerve entrapment in an adolescent during a growth spurt: a case report and hypothesis on growth-related vulnerability.

BACKGROUND: Common peroneal nerve entrapment at the fibular neck is a well-recognized cause of foot drop, but atraumatic cases in adolescents may be diagnostically challenging. Rapid growth during adolescence may increase local nerve vulnerability through growth-related neuro-musculoskeletal factors, although direct evidence remains limited. CASE PRESENTATION: We report a 16-year-old male who presented with a two-week history of weakness in the left ankle and hallux dorsiflexion without a clear history of trauma, habitual leg crossing, prolonged squatting, tight footwear, systemic disease, or

Structural analysis of de novo-designed binders targeting the closed state of HSP90.

Heat shock protein 90 (HSP90) assists the folding and maturation of numerous signaling proteins and is an important therapeutic target. Most HSP90 inhibitors target the ATP-binding pocket and disrupt ATP-induced closure, whereas inhibitors targeting the closed state have not been developed. Here, we report de novo-designed protein binders that selectively recognize closed HSP90. Two binders, H2 and H4, have similar folds and affinities for HSP90 but distinct binding kinetics. Cryo-EM structures of HSP90 complexes with H2 and H4 revealed high accuracy of the BindCraft predictions. H2 and H4 com

Homoacetogenic bioaugmentation with Clostridium magnum WY-4 enhances upflow anaerobic sludge blanket reactor resilience under progressive organic loading and short-term complex shock.

Upflow anaerobic sludge blanket (UASB) reactors are widely used for treating high-strength organic wastewater but are vulnerable to short-term complex shocks caused by increased organic loading, hydraulic retention time regulation, and partial sludge loss. At the same time, the role of homoacetogenic bioaugmentation in improving UASB recovery after such disturbances remains poorly understood. This study evaluated the potential of a novel homoacetogenic strain, Clostridium magnum WY-4 (WY-4), to enhance UASB resilience. Two laboratory-scale UASB reactors were operated for 184 days, and reactor

Factorization of solutions to generalized Feynman-Kac equations for jump-diffusion models.

Standard affine jump-diffusion models treat shocks as increments to the system's state, whereas conventional stochastic resetting models reduce their impact to a full reset of the system's state. Neither model is adequate when diffusion along one of the state components persists, while shocks affect another component. Our study examines a linear system perturbed by two independent additive Markovian components. We demonstrate that in this model the solutions to the generalized Feynman-Kac (GFK) equations describing the expected evolution operator admit an exact factorization to solutions corre

Staggering dominolike blast front motion in a one-dimensional cold gas.

One-dimensional alternating particle systems are widely used to study interconnections between the hydrodynamics of blast waves in a gas-like medium and the Newtonian dynamics of its corpuscular constituents. In this article, we study the model in which point particles with masses m,μ,m,μ,⋯, (m≥μ) are distributed on the positive half-line R_{+}. Their dynamics are initiated by giving a positive velocity to the leftmost particle; in its course, the particles undergo elastic collisions. For this model with m/μ=2, it has been established that the dynamics that start from random initial positions

A PebZIP44-PeHSF12-PeHSP17.8 transcriptional module controls heat shock protein expression and thermotolerance in passion fruit(Passiflora edulis).

Heat stress is a major constraint for passion fruit (Passiflora edulis) production, yet the transcriptional mechanisms underlying thermotolerance in this perennial vine remain poorly understood. This study identified 15 HSF genes (PeHSFs) in Passiflora edulis (P. edulis) genome. Phylogenetic analysis classified these PeHSFs into three distinct clusters containing 9, 4, and 2 genes, respectively, with conserved structural features supporting their functional divergence.Tissue-specific expression profiling demonstrated significant organ-dependent expression patterns of PeHSFs across roots, stems,

Impact of oxidative degradation on the bioavailability of subcutaneously administered adalimumab.

Subcutaneous (SC) administration of therapeutic antibodies enhances patient convenience and healthcare resource efficiency compared to intravenous (IV) delivery, but therapy can be limited by incomplete bioavailability (BA) of SC biotherapeutics. This limitation may be attributed to processes inherent to the unique composition of SC tissue, including the potential presence of reactive oxygen species (ROS) within the SC microenvironment, which can induce chemical and structural instability and functional impairment. This study investigated the impact of oxidation on the SC BA of adalimumab (Hum

Continuous versus intermittent, non-invasive monitoring of vital signs in hospitalised, ward based adult patients: A systematic review and narrative synthesis.

BACKGROUND: Intermittent monitoring (IM) of vital signs remains the standard of care for hospitalised ward patients, yet this approach creates gaps during which patient deterioration may go unrecognised. Wearable wireless devices capable of continuously measuring physiological parameters offer the potential for earlier detection of clinical decline and prevention of intensive care unit admissions. This systematic review aimed to compare continuous, non-invasive vital signs monitoring with standard IM for the identification of deterioration and prevention of complications in adult, ward-based h

IVIS assessment of mRNA-LNP protein expression: effects of dosing parameters and prior LNP exposure.

Bioluminescence imaging using in vivo imaging systems (IVIS) is widely used to assess reporter protein expression after administration of messenger RNA (mRNA) lipid nanoparticles (LNPs). However, methodological variation and prior LNP exposure can complicate interpretation. Here, we evaluated selected parameters affecting IVIS readouts after intramuscular administration of firefly luciferase mRNA formulated in SM-102 or ALC-0315 LNPs in BALB/c mice. We also investigated the effects of repeated administration. D-luciferin delivery, imaging interval and mouse position influenced signal acquisiti

Bioinspired Active-Defense Coating with a Thermal Stability Relay for Mitigating Matrix Thermal Runaway Propagation.

Inspired by the iron birch (Betula schmidtii), a biomimetic active-defense coating (IBMC) is engineered. Within a polymeric-aerogel matrix, rigid mica platelets mimic protective bark, while temperature-responsive sustained-release microspheres (SRMs) function as artificial cells. Upon thermal shock, SRM actively release repair agents to trigger a distinct thermal stability relay mechanism. Initially (<400 °C), the released amino-silicone oil induces rapid interfacial cross-linking to counteract polymer decomposition. Subsequently, at extreme temperatures (>800 °C), an amorphous inorganic skele

Upcycling of cardoon root biomass into polyhydroxyalkanoate nanocarriers for docetaxel delivery.

Lignocellulosic residues represent a promising low-cost feedstock for the sustainable production of polyhydroxyalkanoates (PHAs), although their integration into high-value biomedical applications remains limited. This work reports an integrated approach for the valorization of cardoon root biomass into poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx) and its subsequent application as a nanocarrier for docetaxel delivery. Cardoon roots were subjected to cascade fractionation followed by deep eutectic solvent (DES)-based pretreatment to enhance lignin removal while preserving the carbohyd

Comparative generation and characterization of probiotic-derived bacterial ghosts from gram-negative and gram-positive bacteria using chemical induction.

OBJECTIVES: Bacterial ghosts (BG) are empty bacterial cell envelopes that retain native surface architecture and antigenic structures, making them promising platforms for vaccine delivery and immunotherapy. This study aimed to develop a simple, scalable chemical method for generating probiotic BG using representative Gram-negative and Gram-positive bacterial strains. METHODS: BG were produced by Tween-80-mediated lysis followed by acidic treatment. Structural and morphological integrity was assessed using scanning and transmission electron microscopy. Particle size and zeta potential were det

Prefrontal ensembles represent freezing of conspecifics.

Social buffering is a process in which the presence of other members of the same species (conspecifics) decreases stress reactions during threat exposure. In parallel, prior reports showed that groups of individuals display synchronous actions in non-threatening situations. However, it is unknown if individuals also synchronize actions when facing threats. Prior studies showed social buffering of freezing in pairs of subjects. We now expand these findings and show social buffering of freezing during exposure to a predator and a shock grid in mice of both sexes in a larger group of four conspec
