Clinical Progress in Virotherapy: Application and Future Prospects in Head and Neck Cancer.

Virus-based cancer therapy (virotherapy) is currently being researched as a novel form of immunotherapy and has already entered the clinical application phase. Among various types of virotherapy, oncolytic virotherapy involves infecting tumors with tumor-selective viruses, such as herpes simplex virus, adenoviruses, and vaccinia virus, and utilizing their replicative capacity to induce cell destruction within tumors. In addition, this therapy aims to enhance tumor immunity by changing the tumor microenvironment through viral infection. Genetic deletion in viruses is used to reduce their virule

Risk of postpartum infection after cesarean section in women with multiple sclerosis: A population-based cohort study.

OBJECTIVE: Cesarean section (CS) rates are increased in women with multiple sclerosis (MS). It is unknown whether CS in women with MS constitutes a risk of maternal postpartum infection compared to vaginal delivery. METHODS: We used the Danish national health registers to establish a cohort of all live births by women with MS in Denmark from 1995 to 2023. We estimated the risk of maternal postpartum infections according to the mode of delivery in women with MS using logistic regression models adjusted for confounders such as age, body mass index, and comorbidity. RESULTS: Overall, 872 women

Circulating PEG-indoleamine 2,3-dioxygenase ameliorates diverse inflammatory diseases without toxicity or compromising immunocompetence.

Indoleamine 2,3-dioxygenase (IDO), the enzyme responsible for tryptophan catabolism, is protective in many autoimmune and inflammatory diseases. We previously developed a localized immunomodulation approach through the fusion of IDO to a carbohydrate binding protein [E. Bracho-Sanchez et al., Nat. Biomed. Eng. 7, 1156-1169 (2023)]. Here we further develop IDO as a protein therapeutic for inflammation, investigating systemic delivery via conjugation of poly(ethylene glycol) (PEG) to IDO. PEGylation extended circulation time and treatment PEG-IDO demonstrated therapeutic efficacy in five autoimm

Neural Stem Cells in Multiple Sclerosis: A Comprehensive Review of Preclinical and Clinical Evidence for CNS Repair and Translational Hurdles.

Multiple sclerosis (MS) is a chronic, immune-mediated, neurodegenerative disease of the central nervous system (CNS) characterized by inflammation, demyelination, and axonal destruction. MS impacts about 2.6 million individuals worldwide, typically presenting a wide array of clinical manifestations, such as motor, sensory, visual, and dementia, often leading to progressive disability. This progressive condition and the accumulation of handicaps draw attention to a crucial unmet need for therapies able to preserve neurons, encourage myelin regeneration, and restore neurological function. Althou

Free Epstein-Barr virus-induced gene 3 functions as an autonomous immunosuppressive cytokine to maintain immune tolerance.

Epstein-Barr virus-induced gene 3(EBI3) is conventionally viewed as a subunit of IL-27 and IL-35, yet the function of uncomplexed "free" EBI3 is unknown. We show that free EBI3 is an autonomous immunosuppressive cytokine that acts via a gp130/WSX-1/STAT3 axis and epigenetic reprogramming. A free-EBI3-specific sandwich ELISA revealed 23.3 ± 2.2 ng/mL in healthy human sera and equivalent levels in mice; no cross-reactivity with IL-27/IL-35 was observed. Serum free EBI3 was reduced in rheumatoid arthritis(RA; 12.4 ± 1.4 ng/mL) and multiple sclerosis (MS; 14.6 ± 1.4 ng/mL; both p< 0.001) and inver

Structural basis for Mg²⁺-glycine synergy in the tandem glycine riboswitch revealed by cryo-EM.

Part I of this article discussed the potential functional importance of genetic mutations and alleles of the human cytochrome P450 2D6 (CYP2D6) gene. The impact of CYP2D6 polymorphisms on the clearance of and response to a series of cardiovascular drugs was addressed. Since CYP2D6 plays a major role in the metabolism of a large number of other drugs, Part II of the article highlights the impact of CYP2D6 polymorphisms on the response to other groups of clinically used drugs. Although clinical studies have observed a gene-dose effect for some tricyclic antidepressants, it is difficult to establ

Blood-Triggered In Vivo Reassembly of Oral Cystine Spherulites with Fasting Synergy Induces Disulfidptosis for Cancer Metabolic Intervention.

Integrating localized amino acid perturbation with systemic metabolic regulation represents a compelling strategy to exploit tumor metabolic vulnerabilities. However, current metabolic modulators remain challenged by poor tumor selectivity, limited oral bioavailability, and reliance on complex delivery systems that hinder clinical translation. Here, we report a fasting-augmented amino acid intervention strategy using iron-cystine spherulites (FeCST). Following oral administration, FeCST is converted in the stomach into ultrasmall cystine nanoparticles (FeCST-NPs) that traverse intestinal mucus

A generalizable covalent car-t platform for solid tumors enabled by oncolytic adenovirus-delivered artificial antigens.

BACKGROUND: Chimeric antigen receptor (CAR) T-cell therapy has shown limited efficacy against solid tumors due to antigen heterogeneity and scarcity of tumor-specific targets. METHODS: To address those challenges, we report a covalent CAR-T strategy that achieves programmable tumor recognition via oncolytic adenovirus-mediated (OAD) delivery of artificial antigens. An engineered OAD was designed to induce tumor-selective expression of a membrane-anchored SpyTag-containing artificial antigen on infected tumor cells. In parallel, we generated SpyCatcher CAR-T cells by replacing the conventional

Agonistic anti-CD40 antibodies in patients with solid tumors: a systematic review and evidence map of clinical safety, with exploratory proportional synthesis.

BACKGROUND: Agonistic anti-CD40 antibodies and related CD40 agonist biologics are being developed across multiple solid tumors using diverse biologic formats, delivery routes, and combination regimens. Safety is central to their clinical development but remains difficult to interpret because high-grade adverse events in combination settings may reflect partner therapies, and safety outcomes are reported inconsistently. METHODS: We conducted a systematic review and evidence map of the clinical safety of CD40 agonists in patients with solid tumors. Major bibliographic databases, trial registrie

Toward process-based inventory data for Portland clinker substitutes.

BACKGROUND: The World Trade Center (WTC) attacks on September 11, 2001 (9/11) resulted in over 2700 fatalities and thousands injured. Injury on 9/11 has been identified as a risk factor for physical and mental health conditions, but the reasons for this are not well understood. In a population exposed to 9/11 and since followed, an in-depth study on the impacts of injury on 9/11 was conducted to identify factors that contribute to long-term functional issues. This report sought to examine factors influencing participation, participant recall of injury status over time, and determinants of inju

Cesarean sections prior to 39 weeks among pregnant patients with multiple sclerosis.

BACKGROUND: Pregnant women with multiple sclerosis have higher Cesarean section (C-section) rates than controls. Whether these are occurring prior to 39 weeks' gestation is unclear and important, as C-sections <39 weeks are associated with increased neonatal morbidity and are not recommended unless medically indicated. OBJECTIVES: Determine whether pregnant women with multiple sclerosis experience higher rates of non-medically indicated C-sections <39 weeks and whether these were attributed to multiple sclerosis. METHODS: We conducted a retrospective, population-based case-control study of p
