Investigation of a GPC1-targeted and LIFU-responsive nanoplatform with ADV effect for visualized chemo-sonodynamic therapy against pancreatic ductal adenocarcinoma.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies. Regardless of surgical resection, chemotherapy acts as the first-line treatment for PDAC. However, the clinical outcomes are severely compromised by insufficient tumour-specific targeting, chemoresistance, and systemic toxicity. Thus, new synergistic therapeutic strategies are urgently needed to improve therapeutic efficacy of PDAC. PURPOSE: This study developed a glypican-1 (GPC1)-targeted and low-intensity focused ultrasound (LIFU)-responsive nanoplatform for visualized treatment against PDAC.

Extravertebral temperature exposure during spinal radiofrequency ablation: an experimental surrogate assessment of neural injury risk.

BACKGROUND: Radiofrequency ablation (RFA) has become an important minimally invasive option for the treatment of painful spinal tumors and benign bone lesions. While its effectiveness within the vertebral body is well established, there is ongoing concern about unintended heat exposure of nearby neural structures. Experimental data describing how heat propagates beyond the vertebra during spinal RFA are still limited. This experimental in vitro study aims to assess the extravertebral temperature exposure during spinal radiofrequency ablation as a surrogate parameter for potential neural injury

Disaster-related healthcare system disruption and maternal-neonatal outcomes.

INTRODUCTION: Natural disasters disrupt healthcare systems and create crises that severely impact maternal and perinatal health outcomes. The 2023 Kahramanmaraş earthquakes caused widespread destruction in southeastern Turkey, leading to marked inequalities in healthcare access and obstetric service delivery. This study compared maternal and neonatal outcomes under disaster-related healthcare system disruption between an earthquake-affected region and an unaffected reference region. METHODS: This retrospective comparative study included 403 women who delivered between February 6 and 21, 2023,

Penetrating the cell wall: a review on delivering exogenous materials into plant and fungal cells via micro/nanoinjection.

Intracellular delivery into walled cells is essential for advancing research in plant science, microbiology, and synthetic biology. Conventional enzymatic wall degradation methods are inherently destructive, leading to poor cell survival. Micro/nanoinjection technology, which enables the physical penetration of the cell wall for direct substance introduction, has emerged as a critical solution. This review article presents a systematic overview of this technology. First, we summarize the state-of-the-art in probe design and fabrication. Next, we analyze automation and force-feedback strategies

Bacteriophage therapy against American foulbrood in honey bees: a PRISMA-guided systematic review.

American foulbrood (AFB), caused by the spore-forming bacterium Paenibacillus larvae, is the most destructive bacterial brood disease of Apis mellifera. Control is challenging because endospores persist for decades and antibiotic use is restricted by regulations, resistance, and residue concerns. Bacteriophages have re-emerged as a species-specific, residue-free strategy for AFB prevention and treatment. A PRISMA-guided systematic review was conducted searching PubMed, Embase, and Web of Science using the terms (bacteriophage OR phage) AND (honeybee OR Apis mellifera). A total of 90 records we

Integrative network pharmacology and in vitro/in vivo validation reveal the protective effects of sotetsuflavone against osteoarthritis associated with PI3K/Akt/NF-κB signaling.

Osteoarthritis (OA) is a prevalent whole-joint disease characterized by progressive cartilage degeneration, extracellular matrix (ECM) breakdown, synovial inflammation, and pathological changes in multiple joint tissues. Inflammatory signaling plays a central role in chondrocyte catabolic activation and cartilage matrix degradation. Sotetsuflavone (SF), a naturally occurring biflavonoid isolated from Cycas species and other medicinal plants, has been reported to possess anti-inflammatory and antioxidant properties; however, its protective effects and related molecular mechanisms in OA remain u

Targeted Intra-Articular PRGF Delivery Using Enzyme-Powered Nanobots for Chondral Lesions: A Prospective Experimental Study with Biomarker Evaluation in Rabbits (Oryctolagus cuniculus).

Osteoarthritis (OA) is the most prevalent degenerative joint disease in both humans and companion animals, causing progressive cartilage destruction for which no curative treatment currently exists. Plasma rich in growth factors (PRGF) has shown chondroprotective effects in veterinary and preclinical models, but its efficacy is limited by the viscosity of synovial fluid and rapid degradation of growth factors within the joint. This study aimed to evaluate the effect of intra-articular co-administration of PRGF and urease-powered enzymatic nanobots on serum biomarkers of inflammation, oxidative

Epigenetic regulation in osteoarthritis: recent updates and emerging mechanisms.

Osteoarthritis (OA) is a common chronic degenerative joint disease characterized by progressive cartilage destruction, synovial inflammation, subchondral bone remodeling, and functional decline. Current treatments remain largely symptomatic and are unable to effectively halt or reverse disease progression. Increasing evidence indicates that epigenetic regulation provides a critical link between genetic susceptibility, mechanical loading, inflammation, aging, metabolic abnormalities, and pathological gene expression in OA. This review summarizes recent advances in OA epigenetics, with a particu

A T2 Translational Science Modified Delphi Study: Management of Patients With Embedded Unexploded Ordnance in Low-Resource Settings.

INTRODUCTION: Actors in conflict, those in asymmetric warfare, conducting criminal activity or participating in paramilitary training activities may possess explosive ordnance. For various reasons this ordnance can misfire resulting in failure to detonate and become impaled or embedded in those near an intended target. The management of the patient with an impaled or embedded unexploded ordnance (eUXO) is a high impact event with the potential to cause death of those in proximity and destruction of the health care facility. The probability of this event is low with less than 50 cases reported

Electroporation-primed cascade nanocatalysis boosts STING-driven chemoimmunotherapy in hepatocellular carcinoma.

Nanosecond pulsed electric field (nsPEF) ablation enables non-thermal tumor destruction with favorable tissue selectivity, but its long-term efficacy is often limited by postoperative recurrence. Here, we developed an electroporation-primed, tumor microenvironment-responsive core-shell cascade nanoparticle, ZnO2-NiFc@MSCU (ZNFM), to potentiate nsPEF-mediated hepatocellular carcinoma (HCC) therapy and induce durable antitumor immunity. ZNFM contains a degradable ZnO2 core that releases Zn2+ and generates H2O2, together with an acid-responsive NiFc shell that catalyzes H2O2 into cytotoxic reacti

Spatiotemporal ultrasound-driven piezoelectric electro-NO therapy restores neuro-vascular-immune homeostasis for radiation ulcer repair.

Radiation ulcers (RUs) represent a devastating and intractable complication arising from radiotherapy, driven by a self-perpetuating pathological cascade featuring persistent oxidative stress, unresolved chronic inflammation, progressive neurovascular destruction, and recurrent bacterial infection, which cannot be sufficiently mitigated by conventional single-target therapeutic interventions. Both electrical stimulation (ES) and nitric oxide (NO) therapy exhibit promising regenerative potential for tissue repair; nevertheless, traditional ES relies on invasive implanted electrodes with unsatis

A γ-polyglutamic acid-based artificial biofilm for field delivery of Bacillus amyloliquefaciens KNU-28: an uncontrolled multi-orchard demonstration on peach gummosis with exploratory bark fungal community profiling.

Peach gummosis, caused primarily by Botryosphaeria dothidea sensu lato, is a destructive canker disease with few environmentally friendly management options. Here we report an uncontrolled pre/post field demonstration of a γ-polyglutamic acid-based artificial biofilm - a biodegradable delivery platform whose agent-agnostic design remains untested - encapsulating Bacillus amyloliquefaciens KNU-28 across 143 trees in 18 commercial peach orchards in South Korea. Severity declined from a pre-treatment mean of 4.12 (April) to 2.32 by August (76.2% of 143 trees improved by ≥1 point) and to 1.78 by O

Consensus in prenatal management of Hemolytic disease of the fetus and newborn.

Hemolytic Disease of the Fetus and Newborn (HDFN) is complication caused by maternal alloantibodies crossing the placenta and leading to the destruction of fetal red blood cells. While RhD is the most common antigen causing HDFN, Kell alloimmunization is the most severe. The assessment of HDFN risk begins with identifying the fetal antigen status. The HDFN Delphi consensus supports the use of cell-free fetal DNA (cffDNA) to determine fetal antigen status for RhD, Kell, and Rhc as a noninvasive alternative to amniocentesis. Ultrasound monitoring, including middle cerebral artery (MCA) Doppler a

VEGF-D-induced intraosseous lymphangiogenesis drives site-specific heterotopic bone resorption.

Heterotopic ossification (HO) is a debilitating condition that commonly occurs after musculoskeletal injury and is characterized by the formation of bone in soft tissues. Despite advances in understanding its pathogenesis, effective therapies to reverse established heterotopic bone remain lacking. While lymphatic vessels are associated with the destruction of bone in rare diseases such as Gorham-Stout disease, their effect on HO has not been widely explored. Here, we use transgenic mice to determine whether targeted expression of the lymphatic growth factor VEGF-D can promote the therapeutic r

Thread- reinforced encapsulation devices (THRED) for therapeutic stem cell delivery.

INTRODUCTION: Mesenchymal stem cells (MSCs) have been studied as potential therapeutic agents for various diseases. However, their widespread application has been limited, in part, due to the concern for tumorigenic transformation and lack of longevity in host environments secondary to immune-mediated destruction. Therefore, we set out to study an MSC encapsulation and delivery method with potential application in the treatment of intestinal ischemic diseases. METHOD: Human vertebra bone adherent mesenchymal stem cells (vBA-MSCs) were encapsulated in an alginate-derived Thread Reinforced Enca

Localized Delivery of Mesenchymal Stem Cell Spheroids via an Injectable Hydrogel for Rheumatoid Arthritis Therapy.

Rheumatoid arthritis (RA) is an autoimmune disorder marked by chronic joint inflammation and tissue damage. While current systemic therapies mainly reduce inflammation, they often lack regenerative capacity and cause off-target effects. To address this, we developed an injectable collagen-hyaluronic acid (Col-HA-NHS) hydrogel for intra-articular delivery of umbilical mesenchymal stem cell (UMSC) spheroids. The hydrogel demonstrated excellent shear-thinning and regelable properties, biocompatibility, and controlled degradation. Encapsulated UMSC spheroids (S-UMSCs) exhibited enhanced paracrine
