Unraveling the Nano World in Paracoccidioidomycosis: Promising Applications of Nanotechnology in Diagnosis, Treatment, and Vaccines: A Mini Review.

Paracoccidioidomycosis (PCM) is a systemic mycosis caused by fungi of the genus Paracoccidioides. This disease is prevalent in Latin America, with Brazil being an endemic region. This mycosis can be classified as PCM infection, PCM disease and residual PCM. Although diagnosis and treatment exist, they have some limitations, and there is no vaccine available for this disease. Thus, the application of nanotechnology in the biomedical and health areas has become an innovative alternative. In this review, we highlight the main advances in the use of nanotechnology to improve and/or develop methods

In vitro biofilm formation and virulence of Paracoccidioides lutzii (LDR2) in a Galleria mellonella larval model.

Paracoccidioidomycosis (PCM) is a systemic mycosis caused by the thermodimorphic fungi Paracoccidioides brasiliensis and the newly identified species P. lutzii. This study investigated in vitro biofilm formation by P. lutzii (LDR2) and assessed its impact on Galleria mellonella larvae. Scanning electron microscopy (SEM) revealed that P. lutzii (LDR2) forms dense biofilms composed of a complex cellular network embedded in an extracellular matrix. Biofilm-associated infection significantly increased fungal virulence, resulting in higher larval mortality, reduced hemocyte density, and enhanced me

Antigenic Similarity Between Different Paracoccidioides Species in a Murine Model of Paracoccidioidomycosis Reveals High IgG Avidity for the 70 kDa Antigen.

Since the new taxonomic proposal for differentiating Paracoccidioides species, many questions have emerged regarding its relevance beyond molecular analysis. Geographical distribution, pathogenesis, morphology, and virulence are still under debate to confirm the diverse genotypic profile of these species. In this study, we evaluated the serological differences of mice experimentally infected with Paracoccidioides brasiliensis sensu stricto, Paracoccidioides restrepiensis, Paracoccidioides americana, Paracoccidioides venezuelensis, and Paracoccidioides lutzii. The serological differences betwee

Partial Alpha-Tubulin Gene Amplification by Nested PCR in Fresh Biological Samples and Formalin-Fixed, Paraffin-Embedded Tissues for Paracoccidioides spp. Identification.

Paracoccidioidomycosis is an important deep mycosis in Latin American countries. Its causative agents have been reclassified into five distinct species based on genetic differences: Paracoccidioides brasiliensis sensu stricto, Paracoccidioides americana, Paracoccidioides restrepiensis, Paracoccidioides venezuelensis and Paracoccidioides lutzii. In this study, we propose a new method, based on a nested PCR for partial alpha-tubulin gene amplification, as a tool for diagnosis and species differentiation directly from biological samples. The method could amplify the DNA of the five cultivable Par

Pulmonary Hypertension Secondary to Fungal Infections: Underexplored Pathological Links.

BACKGROUND/OBJECTIVE: Pulmonary fungal infections are a significant diagnostic challenge, primarily affecting immunocompromised individuals, such as those with HIV, cancer, or organ transplants, and they often lead to substantial morbidity and mortality if untreated. These infections trigger acute inflammatory and immune responses, which may progress to chronic inflammation. This process involves myofibroblast recruitment, the deposition of extracellular matrix, and vascular remodeling, ultimately contributing to pulmonary hypertension. Despite its clinical relevance, pulmonary hypertension se

Partial alpha-tubulin gene amplification by nested PCR as a tool for Paracoccidioides ceti identification.

Paracoccidioidomycosis ceti (PCMC), also called lobomycosis, is a cutaneous disease affecting cetaceans worldwide. It is caused by Paracoccidioides ceti, an uncultivable fungal species recently classified within the Paracoccidioides genus. Although several molecular markers have been used to investigate the PCMC pathogen, the alpha-tubulin gene (TUB1), commonly utilized in genetic studies of cultivable Paracoccidioides, has remained unexplored in this taxon. In this study, we applied a nested polymerase chain reaction targeting TUB1 to amplify fungal sequences from two new cases of PCMC in Bra

Systemic endemic mycoses: From a geographical risk to a concern expansion.

Blastomycosis, coccidioidomycosis, histoplasmosis, paracoccidioidomycosis, emergomycosis, and talaromycosis are sapronoses restricted to specific geographical areas associated with particular environmental niches that can lead to high morbidity and mortality if diagnosed or treated late. Challenges in clinical awareness arise from their nonspecific clinical presentations and the limited availability of sensitive diagnostic tools. Although advances in molecular diagnosis and antigen detection-based tests are promising, they are not available for all systemic endemic mycoses (SEM). Treatment gui

Acidic pH Modulates Cell Wall and Melanization in Paracoccidioides brasiliensis, Affecting Macrophage Interaction.

Paracoccidioides brasiliensis is a thermally dimorphic fungal pathogen and the main etiological agent of paracoccidioidomycosis (PCM), a neglected systemic mycosis endemic in Latin America. The virulence of P. brasiliensis is closely associated with its capacity to survive under hostile host conditions, including acidic environments. In this study, we demonstrate that acidic pH induces melanization in P. brasiliensis, modulates its cell wall composition, and alters the interaction with macrophages. Cultivation at acidic pH resulted in reduced fungal growth without compromising viability and tr

Jorge Lobo's Disease in Child with Tick Exposure, Brazil.

Jorge Lobo's disease (JLD), caused by Paracoccidioides lobogeorgii, primarily affects inhabitants of the Amazon Forest. We report a 9-year-old boy in Brazil who had JLD diagnosed after a tick bite. The rarity of pediatric cases likely reflects surveillance gaps. Increased clinical awareness is crucial for early JLD detection and intervention, especially in endemic regions.

IL-6 Degradation by Secreted Proteases From Paracoccidioides restrepiensis.

Paracoccidioidomycosis is a systemic fungal disease caused by Paracoccidioides spp., predominantly affecting populations in Latin America, with Brazil reporting the highest number of cases. The infection is associated with severe pulmonary and systemic manifestations. Previous studies have highlighted the role of fungal proteases in adhesion, invasion, and the modulation of host immune responses, implicating them as key virulence factors. Our group previously demonstrated that Paracoccidioides restrepiensis secretes proteases that activate protease-activated receptors (PAR-1 and PAR-2) in huma

Cutaneous clues to a rare osseous manifestation of paracoccidioidomycosis: The importance of early diagnosis.

Paracoccidioidomycosis (PCM), the most prevalent systemic mycosis in Brazil, is caused by dimorphic fungi of the genus Paracoccidioides and is acquired primarily through inhalation. PCM is associated with a high mortality, particularly among men living in rural areas. This case report describes a 47-year-old man with osteocutaneous involvement diagnosed by biopsy and direct fungal examination. The osseous form of PCM is rare and should be differentiated from other infections and neoplasms. Management involves prolonged antifungal therapy and follow-up for at least two years. This case highligh
