Sporotrichosis, a subcutaneous mycosis, can manifest as severe disseminated disease in immunocompromised hosts. This report describes a challenging case of extracutaneous sporotrichosis in a 28-year-old female with systemic lupus erythematosus (SLE) on immunosuppressive therapy, who also faced significant socioeconomic constraints. The patient initially presented to the emergency department with a painful right inguinal mass. Physical examination revealed a hemorrhagic lesion, and aspiration of the material was performed for culture, which returned negative. She was discharged but returned 2 m

AIM: To investigate the antifungal activity of Anacardium occidentale leaf extract can fight Sporothrix brasiliensis, a fungus that cause sporotrichosis.
MATERIAL AND METHODS: We tested the extract against 20 fungal samples obtained from humans. We also evaluated whether the extract could reduce the growth of fungal communities called biofilms and examined changes in fungal cells after treatment.
RESULTS AND CONCLUSION: The extract, rich in gallic acid, rutin, and ellagic acid, was able to stop fungal growth at low concentrations and reduced mature biofilms by up to 68%. Microscopy showed da

Sporotrichosis caused by Sporothrix brasiliensis represents an emerging public health challenge in Brazil, with increasing therapeutic limitations and antifungal resistance. This study investigated Brazilian geopropolis produced by Melipona quadrifasciata anthidioides as a potential source of antifungal bioactive compounds. Bioassay-guided fractionation of the ethanolic extract led to the isolation of three constituents, identified by NMR, Fourier transform infrared spectroscopy (FTIR)-ATR, and LC-HRMS analyses as kaurenoic acid, cinnamoylgrandifloric acid, and 3S(+)-vestitol, the latter two b

UNLABELLED: Esophageal cancer remains a significant global health challenge with increasing rates of incidence and mortality, particularly esophageal squamous cell carcinoma (ESCC), with higher prevalence in Asia. This study aims to investigate the cytotoxicity and potential cell death mechanisms (i.e., apoptosis, necroptosis, ferroptosis, and pyroptosis) induced by three newly synthesized benzamidine derivatives (NBDs) against the ESCC KYSE-150 cell line. MTT cell viability assay was used to assess the cytotoxic effect of the NBDs on the ESCC cell line, KYSE-150, and the normal esophageal epi

BACKGROUND: Sporotrichosis is a chronic, deep fungal infection of skin caused by Sporothrix schenckii. Macrophages are predominant in S. schenckii-infected skin and are able to phagocytize and kill the fungus. Local hyperthermia is effective for treating sporotrichosis, but its mechanism of action is still not fully understood.
METHODS: Using single-cell RNA sequencing of sporotrichosis lesions, coupled with in vitro and in vivo sporotrichosis models, we investigated the role of tumor necrosis factor receptor-associated factor 1 (TRAF1) and nitric oxide synthase, inducible (NOS2). Mechanistic

Sporothrix brasiliensis is the primary cause of sporotrichosis in Brazil. Reports of therapeutic failures with itraconazole treatment exist. We evaluated the antifungal activity of the hydroacetone extract of Psidium guajava in planktonic cells and biofilms of S. brasiliensis. The extract was obtained from the leaves, and high-performance liquid chromatography was performed. Broth microdilution tests were performed according to the CLSI M38. The strains were subjected to biofilm formation, and the antifungal activity of the extract against a strong producer strain was determined. The extract i

Sporothrix species are thermally dimorphic fungi responsible for sporotrichosis, a globally prevalent subcutaneous mycosis and an emerging zoonotic threat, particularly in South America. The high virulence of Sporothrix brasiliensis and its efficient transmission from cats to humans have intensified recent outbreaks, underscoring the importance of understanding the pathogenic mechanisms. While several putative virulence factors have been identified, such as melanin production, cell wall remodeling, extracellular vesicles, and thermotolerance, functional studies remain hampered by limited molec

Cat-transmitted sporotrichosis is one of the fastest spreading zoonosis in Rio de Janeiro State, Brazil. A retrospective study was conducted with analysis of incidence of human sporotrichosis complemented with spatial methodologies. Data from case reported to the Notifiable Diseases Information System (SINAN) from 2007 to 2023 were studied. Incidence, demographic variables, temporal, and spatial dynamics of this endemic disease were investigated. During 2007-2023, 15,401 cases of sporotrichosis were reported. Most cases (64.4%) occurred with women. The annual incidence from 2007 to 2023 was 5.

Osteoarticular sporotrichosis is the most common extracutaneous type of the disease and may occur either concomitantly with cutaneous lesions or as isolated musculoskeletal disease, the latter frequently resulting in delayed diagnosis. We describe five confirmed cases of osteoarticular sporotrichosis diagnosed between 2002 and 2023 at a university hospital in Brazil. Diagnosis was confirmed by fungal culture, with serology and histopathology used as complementary methods. Clinical and epidemiological data, radiologic findings, treatment, and outcomes were analyzed. Patients were 39 to 67 years

BACKGROUND: Sporotrichosis is a neglected tropical disease caused by several species of Sporothrix, of which S. brasiliensis is the most virulent. Transmission primarily occurs through scratches, bites, or direct contact with exudates from infected cats and potentially through fomites.
METHODS: We evaluated the efficacy of different disinfectant solutions in eliminating S. brasiliensis from contaminated surfaces, including medium-density fiberboard (MDF), tiles, stainless steel, and polyester fabric, by simulating both domestic and clinical environments. Sterile 9-cm² sections were uniformly

Sporotrichosis is a subcutaneous mycosis caused by species belonging to the genus Sporothrix. While zoonotic transmission has been primarily associated with cats, reports have pointed to sporotrichosis transmission by various non-feline animals, albeit infrequently compared with the aforementioned. These reports suggest the potential for zoonotic or environmentally mediated transmission routes with clinical and epidemiological relevance that have yet to be extensively explored. This systematic review organizes the current knowledge on sporotrichosis acquired by non-feline zoonotic transmission

OBJECTIVE: To describe the outcomes of the Trap, Neuter, Diagnosis, Return, Treatment, and Monitoring (TND-RTM) protocol applied to a free-roaming cat colony in a sporotrichosis-endemic area in southern Brazil.
METHODS: Cats were captured and neutered, clinically assessed for sporotrichosis, and managed according to predefined colony-level therapeutic criteria, whereby the colony was classified as sporotrichosis-positive when at least one individual was affected. All cats within a positive colony were included in antifungal management, including isolation of clinically affected cats and preve

BACKGROUND: Sporotrichosis is a subcutaneous mycosis caused by the traumatic inoculation of dimorphic fungi from the genus Sporothrix. Traditional diagnostic methods, including direct microscopy and fungal culture, are time-consuming, require specialized expertise, and may yield false-negative results.
AIMS: This systematic review and meta-analysis aimed to evaluate the diagnostic accuracy of the enzyme-linked immunosorbent assay (ELISA) technique in detecting sporotrichosis in humans and animals.
METHODS: A comprehensive literature search was conducted across PubMed, LILACS, EMBASE, Scopus,

Sporotrichosis is a globally distributed subcutaneous mycosis caused by fungi belonging to the Sporothrix genus. The limitations of current treatment options for sporotrichosis have driven an urgent search for new antifungal drugs. Diphenyl diselenide (PhSe)₂, a stable organoselenium, has promising activity against Sporothrix spp. Since chemical modifications in molecules can enhance their antifungal properties, this study aimed to evaluate the anti-Sporothrix activity of two analogs of (PhSe)₂—p-methoxyl-diphenyl diselenide (BMD) and bis-(3-trifluoromethylphenyl) diselenide (CF3)—both alone a

The rapid expansion of zoonotic sporotrichosis in South America necessitates innovative surveillance strategies to identify natural ecological niches. Roadkill provides a unique, underutilized opportunity to monitor Sporothrix circulation within human-impacted landscapes. We conducted a molecular survey via a triplex probe quantitative real-time PCR (qPCR) assay targeting pathogenic Sporothrix species in 81 roadkilled vertebrates (mammals, birds, and reptiles) collected along highways BR-376 and PR-445 traversing the Atlantic Forest in Paraná, Brazil (2017-2023). Genomic DNA from visceral orga

Cutaneous Rosai-Dorfman disease (CRDD) is a rare form of non-Langerhans cell histiocytosis. Because the etiology and pathogenesis remain unclear and the cutaneous manifestations are highly variable, no definitive diagnostic criteria have been established. As a result, the risk of diagnostic errors and missed diagnoses is common in clinical practice. This report describes the case of a 64-year-old woman with facial CRDD that initially misdiagnosed as sporotrichosis. This case broadens the known clinical spectrum of CRDD and underscores the importance of careful differential diagnosis to avoid m

Background/Objectives: Species of the Sporothrix genus were used as a biological model to identify potential antifungal compounds within the NIH Clinical Collection library. Methods: A total of 707 compounds were screened for antifungal activity using in vitro susceptibility assays. Compounds exhibiting significant inhibitory effects (growth inhibitions greater than 80%) were further evaluated by determining their minimum inhibitory concentrations (MICs). As cerivastatin demonstrated the highest activity after itraconazole, it was selected for further evaluation, either alone or in combination
