The teratogenic effect of caffeine has been clearly demonstrated in rodents. The sensitivity of different animals species is variable. Malformations have been demonstrated in mice at 50-75 mg/kg of caffeine, whereas the lowest dose usually needed to induce malformations is 80 mg/kg in rats. However, when caffeine is administered in fractioned amounts during the day, 330 mg/kg/day are necessary to reach teratogenicity in rats. In rodents, the most frequently observed malformations are those of the limbs and digits, ectrodactyly, craniofacial malformations (labial and palatal clefts) and delays

OBJECTIVE: To determine the effects of passive maternal smoking on birthweight.
DESIGN: Cohort of consecutive singleton live births.
SETTING: Teaching hospital in South India.
SUBJECTS: 994 infants and their mothers.
MAIN OUTCOME MEASURE: Birthweight.
RESULTS: All the women were themselves non-smokers; 520 (52%) were passive smokers while the remainder were not exposed to tobacco smoke. Infants born to passive smokers were on the average 55 g lighter than those born to non-smokers. Passive smoking was associated with a decrease in birthweight of 63 g (95% CI 12-114 g) even after adjusting

Tobacco products and alcohol are commonly used as nonmedicinal drugs by pregnant women, and both are known to cause various effects on the fetus and the newborn. The objective of this study was to examine the fetal effects of both drugs when administered individually and simultaneously to pregnant CD-1 mice at moderate dosages. Specifically, we wanted to determine whether or not the effect on the fetus of these two biologically active substances was additive, ameliorative, or synergistic. A total of 65 CD-1 dams were divided among four groups receiving either ST equivalent to 8 mg/kg nicotine,

There are increasing numbers of reports on the tobacco smoking and ingestion of abused drugs (e.g. morphine, cocaine) by pregnant women and the effects of the substances on the developing fetus and newborn infant. The passage of drugs and chemicals from the mother to the fetus is influenced by the placental transport and metabolism of the substances. Further, these drugs and chemicals affect the nutrient transport systems in the placenta. The three major drugs of abuse-nicotine, morphine and cocaine-depress both active amino-acid uptake by human placental villi and transplacental amino-acid tr

Maternal cigarette smoking has been associated with some complications of pregnancy, including low birth weight and increased morbidity. Recently, it has been reported that maternal passive smoking also affects the fetal environment and causes fetal growth disturbance. In this study, we aimed to investigate the effects of maternal passive smoking on pregnant women and their fetuses by measuring cotinine concentrations in maternal urine and umbilical cord blood. The results were as follows: 1) Among 259 pregnant women, 17 cases (6.6%) were active smokers. The women who were not aware of passive

The association between maternal smoking and both morphometric birth parameters and the perinatal cadmium burden were studied. The cadmium concentrations were measured by atomic absorption spectrometry in 100 samples of maternal whole blood (MB) and in 93 samples of umbilical cord blood (CB). In the group of nonsmokers, significantly higher birth weight and decreased relative placental weight were noted as compared to the group of smokers who smoked more than 6 cigarettes a day (p less than 0.05). In both maternal and cord blood samples, the measured Cd levels were found to be significantly hi

IgG subclass levels in cord serum of unselected European newborns were studied in relation to parental smoking. Cord IgG3 was significantly (P less than 0.01) higher in newborns of smoking mothers (n = 18; geom.mean = 87.0%) compared to those of non-smoking mothers (n = 66; geom.mean = 61.0%). Maternal smoking did not influence (P greater than 0.10) cord IgG2 or cord IgG4. Paternal smoking did not affect any of the three immunoglobulins in cord serum (P greater than 0.10). These data suggest that tobacco smoking affects IgG3 synthesis in some direct or indirect way.

The effects of parental smoking on IgE and IgD levels in cord serum and subsequent infant allergy were investigated in an unselected series of 186 European newborn infants. Maternal smoking caused a significant rise in both IgE (p less than 0.05) and IgD (p less than 0.05), a finding that was most apparent in newborn infants with a negative biparental history of allergy (p less than 0.025 and p = 0.005, respectively). Furthermore, newborn infants of nonallergic parents had a more than threefold (p less than 0.01) higher incidence of elevated cord IgE (greater than or equal to 1.20 IU/ml) and a

The ability of the lungs to release fatty acids from circulating triglycerides or lipoproteins for its own phospholipid synthesis may be one of the factors which limit the rate of surfactant formation. Therefore, the development of the lipolytic activity of lungs obtained from late fetal and neonatal rabbits has been studied and the results correlated to the phospholipid content of lungs of similar ages. Isolated lungs were perfused with a medium which contained cold and radioactive triglyceride, and the release of fatty acids into the perfusion medium was analyzed by both colorimetric and rad

Numerous environmental contaminants can affect the developing embryo, fetus, or infant. This essay explores such questions as these: What is the importance in mutagenesis and teratogenesis of macroenvironmental pollutants such as the heavy metals, dioxin derivatives, polychlorinated diphenyl compounds, and pesticides? What is the significance of microenvironmental pollutants (or social environmental factors) such as tobacco smoke, alcohol, and pharmacologic agents over which exposed individuals have considerable control? What are some of the ethical and legal implications of these toxins of wh
