Deficiency of placental GDF15 causes fetal growth restriction by inhibiting fetal hepatic STAT3-IGF2 axis.

Fetal growth restriction (FGR) is a major cause of perinatal morbidity and mortality, yet its underlying mechanism remains elusive. Here, we show that growth differentiation factor 15 (GDF15) levels are significantly reduced in the placenta and fetal circulation in mouse and pig FGR models and correlate positively with fetal weight. Placenta-specific GDF15 deficiency causes FGR, while placenta-specific Gdf15 mRNA delivery through targeted lipid nanoparticle (LNP) effectively alleviates FGR. Mechanistically, placental GDF15 is a major contributor to the fetal circulating GDF15 pool and enters t

FUBP1 lactylation suppresses trophoblast function in fetal growth restriction.

BACKGROUND: Fetal growth restriction (FGR) is a common complication of pregnancy; however, its exact molecular mechanisms remain unclear. Excessive lactate accumulation in the placenta is often observed during FGR. High concentrations of lactate can regulate the proliferation of pathological cells through lactylation of key proteins. Therefore, we aimed to screen key lactylation modification-related genes (LMRGs) and provide new directions for the diagnosis and treatment of FGR. METHODS: The Differentially expressed LMRGs (DE-LMRGs) in FGR were evaluated using the Gene Expression Omnibus data

Gestational Exposure to the Food Processing Contaminant 3-MCPD Impairs Fetal Growth by Disrupting Placental Taurine Metabolism to Drive Vascular Senescence.

3-Monochloro-1,2-propanediol (3-MCPD), a common food contaminant, induces fetal growth restriction in mice by triggering senescence in the placental vasculature. Transcriptomic analysis reveals suppression of the taurine-hypotaurine pathway through downregulation of Fmo1. Mechanistically, 3-MCPD upregulates Ezh2, which deposits repressive H3K27me3 marks at the Fmo1 promoter, silencing its transcription and depleting placental taurine. Taurine supplementation or Ezh2 inhibition rescues 3-MCPD-induced vascular senescence in human endothelial cells. These findings identify an Ezh2-taurine axis es

Fetal growth restriction and lifelong risk of acute kidney injury: advancing the developmental origins of kidney disease.

Fetal growth restriction has long been associated with reduced nephron number and chronic kidney disease, but its relationship with acute kidney injury has remained unclear. Fog-Høyer et al. demonstrated that fetal growth restriction increases acute kidney injury risk from infancy through early adulthood, particularly in children with congenital anomalies. We examine the study's strengths, limitations, biological basis, and clinical relevance, highlighting opportunities for targeted risk stratification, preventive care, and future research.

Oropouche virus disrupts neurodevelopment and exhibits congenital infection potential.

Oropouche virus (OROV) historically caused a self-limiting disease, yet recent strains have been clinically linked to congenital infection and neurodevelopmental disease. These observations highlight the need to study OROV as a congenital pathogen. Here, using both a historical and currently circulating strain, we show OROV infects neurons across differentiation states in human forebrain organoids. Compared with the neurotropic congenital pathogen Zika virus (ZIKV), OROV exhibits a heightened capacity for neuroinfection and pathology in both forebrain organoids and neonatal mice. The increased

Effects of nutritional low phosphorus status caused by extremely low phosphorus diet during lactation and early childhood on endochondral ossification in offspring rats.

BACKGROUND: Fetal phosphorus is mainly deposited in bones in late pregnancy. Premature birth interrupts the supply of maternal phosphorus, combined with insufficient reserves, immature intestinal absorption and catch-up growth, the demand for mineralization surges, breast milk is difficult to meet, and it is easy to cause low-phosphorus bone disease; as the survival rate of premature infants increases, the incidence also increases. OBJECTIVE: The core of the DOHaD theory is that the early living environment permanently reshapes the body through epigenetic mechanisms, thereby affecting long-te

Maternal Plasma Afamin Levels in Pregnancies Complicated by Fetal Growth Restriction and Their Association With Doppler Markers of Placental Dysfunction.

AIM: To compare maternal plasma afamin between pregnancies with fetal growth restriction (FGR) defined by fetal biometry and healthy controls, to test whether the cross-sectional association between afamin and gestational age differs in FGR, and to examine associations with Doppler indices. METHODS: In this prospective study, 84 women assessed at 28-40 weeks (40 FGR, 44 controls) underwent same-day maternal blood sampling and umbilical-, middle-cerebral-, and uterine-artery Doppler. FGR was defined by an estimated fetal weight or abdominal circumference below the 5th percentile. Plasma afamin

An ESIPT-based ratiometric fluorescent probe for cysteine and its application in complex matrices: milk and fetal bovine serum.

Cysteine (Cys) was an essential biothiol whose dysregulation is linked to neurotoxicity, liver injury, childhood growth retardation, and cardiovascular diseases, making its accurate detection critical for clinical diagnosis and biochemical research. Although excited-state intramolecular proton transfer (ESIPT)-based fluorescent probes offer advantages such as a large Stokes shift and low background interference, conventional ESIPT probes struggled to distinguish Cys from structurally similar homocysteine (Hcy) and glutathione (GSH). Herein, we rationally designed a novel ESIPT probe named LYB-

Placental TRPV2 supports trophoblast function and normal fetal development.

While placental functioning arbitrates fetal outcome, the molecular players that underpin normal placental development remain poorly understood. Here, we provide evidence that the Transient Receptor Potential Vanilloid 2 (TRPV2) channel plays an important role in normal placental development, determining fetal size and survival. Its absence severely affects placental morphology, causing fetal growth restriction and embryonic lethality, which was found to be allocated to a non-redundant role in the trophoblast, but not in the embryo. Spatio-temporal expression of E-cadherin mediates trophoblast lineage transitions during human placentation.

Human placentation relies on the precise differentiation of embryo-derived trophoblasts into specialized lineages, a process strictly regulated by the spatio-temporal expression of key genes. Herein, we report that the expression of the transmembrane glycoprotein, known as E-cadherin, fluctuates dynamically across distinct trophoblast subpopulations rather than acting as a static structural tether. Along the villous pathway, E-cadherin expression initially increases to facilitate the aggregation of cytotrophoblast cells (CTs) and is subsequently downregulated via transcriptional repression, en

Predictors for intrapartum cesarean delivery after labor induction for suspected fetal growth restriction at term.

OBJECTIVE: In term pregnancies with suspected fetal growth restriction (FGR) undergoing induction of labor (IOL), predictors for intrapartum cesarean delivery (CD) remain unclear. We aimed to determine predictors for intrapartum CD in this population. METHODS: This study included a retrospective cohort of singleton pregnancies undergoing IOL at ≥37 weeks for suspected FGR, defined as estimated fetal weight (EFW) or abdominal circumference (AC) <10th percentile, at a tertiary medical center (2011-2019). Maternal characteristics, sonographic measurements, and Doppler parameters were compared be

Evolving Fetal Biometry in Fetal Intrahepatic Portosystemic Shunt With Late-Onset Preeclampsia and Focal Umbilical Cord Hypercoiling: A Case Report.

Fetal intrahepatic portosystemic shunt may complicate fetal growth assessment, particularly when maternal and umbilical cord abnormalities coexist. We report a pregnancy first evaluated at our center at 26 + 3 weeks for suspected severe early-onset fetal growth restriction, with estimated fetal weight below the 1st percentile and abdominal circumference below the 3rd percentile. Prenatal ultrasound and fetal magnetic resonance imaging findings were consistent with fetal intrahepatic portosystemic shunt. Subsequent serial assessments showed improved fetal biometry. Routine umbilical artery and

Accuracy of Ultrasonographic Transcerebellar Diameter for Gestational Age Estimation: A Systematic Review and Meta-Analysis.

BACKGROUND: Accurate estimation of gestational age (GA) is essential in prenatal care and assessment of perinatal outcomes. Standard dating methods such as last menstrual period (LMP) or conventional ultrasound biometry may be limited by fetal position, late antenatal booking, or intrauterine growth restriction (IUGR). The transcerebellar diameter (TCD) has been proposed as a robust alternative for GA estimation, particularly in late gestation or when other methods are unreliable. This study systematically evaluates the accuracy of TCD-based GA estimation in the second and third trimesters in

Acute Bilateral Visual Loss as an Atypical Manifestation of Wernicke Encephalopathy Secondary to Hyperemesis Gravidarum: A Case Report and Narrative Literature Review.

Wernicke encephalopathy (WE) is a severe neurological condition caused by thiamine deficiency that remains underreported in pregnancy. We report a clinical diagnosis of WE in a 41-year-old woman, gravida 2, para 1, at 16 weeks of gestation. She presented with acute, painless visual impairment, severe dietary restriction, and marked weight loss secondary to severe hyperemesis gravidarum (HG). Examination revealed reduced visual acuity, horizontal nystagmus, psychomotor retardation, and an ataxic gait. The initial diagnosis was delayed due to her atypical presentation and the absence of MRI or b

CD38-activated macrophages drive age-related placental senescence by depleting NAD+ in decidual stromal cells.

As delayed childbearing becomes increasingly common, elucidating the mechanisms of age-related placental senescence is critical for the development of effective therapeutic strategies. Here, we integrate metabolomics, single-cell RNA sequencing, and spatial transcriptomics to uncover conserved features of aged placentas in humans, mice, and pigs, namely, increased macrophage CD38 expression and NAD⁺ deficiency. Through pharmacological and genetic approaches, we demonstrate that macrophage CD38 depletes NAD⁺ in decidual stromal cells, thereby promoting placental senescence. Mechanistically, ear

Trio-Exome-Based Mendelian Inheritance Error Analysis Helps Identify Uniparental Disomies and Resolve Discordant NIPT Findings.

OBJECTIVE: Uniparental disomy (UPD), particularly heterodisomy (hUPD), is an underdiagnosed etiology in cases of fetal growth restriction (FGR) and discordant noninvasive prenatal testing (NIPT). Because conventional absence-of-heterozygosity (AOH) analysis frequently misses hUPDs, we aimed to validate a systematic Mendelian inheritance error (MIE) analysis pipeline for trio exome sequencing (trio-ES) to resolve ambiguous prenatal findings. METHOD: A two-stage study analyzed 1948 parent-offspring trios (postnatal and prenatal samples). A retrospective cohort (n = 992) established and validate
