Conserved gene expression patterns in the placenta underlie pregnancy complications associated with hypoxia in mice and humans.

During pregnancy hypoxia leads to complications like fetal growth restriction (FGR). Maternal inhalation hypoxia during late murine pregnancy impairs placental phenotype and fetal development in a severity-dependent manner. To identify the molecular mechanisms and sex-specificity of these effects, placentas from pregnant mice exposed to moderate (13% O₂) or severe (10% O₂) hypoxia were analysed using RNA-sequencing, qPCR, western blotting, histology, and nutrient transport assays. Transcriptomic profiling of male 13% O2 placentas revealed differential gene expression regulating calcium binding

Association of polymorphic loci of the matrix metalloproteinase genes with the development of fetal growth retardation: A case-control study.

BACKGROUND: Matrix metalloproteinases (MMPs) play a crucial role in successful pregnancy as molecular regulatory factors of the developing organism. Additional studies focusing on MMPs activity and function in fetal growth retardation (FGR) may help in the search for diagnostic markers of this pregnancy complication. OBJECTIVE: To study the association of MMPs gene polymorphisms with the development of FGR. MATERIALS AND METHODS: This case-control study included 122 pregnant women with FGR and 243 pregnant women with normal birth weight. We studied polymorphic loci of metalloproteinase genes

Microbial dysbiosis and immunological dysregulation in pathological pregnancy.

Adverse perinatal outcomes of preterm birth (PB), premature rupture of membranes (PROM), intrauterine growth restriction (IUGR), and preeclampsia contribute to a growing number of maternal and fetal deaths each year. Altered immunological regulation of the maternal innate and adaptive immune systems has been implicated in the pathological inflammation dictating obstetrical complications, but the pathogenesis of aberrant maternal immune activation has not been fully elucidated. Microbial dysbiosis within the female reproductive tract may alter the regulation of immunological tolerance at the ma

Perinatal outcomes of pregnant adolescents in rural Japan: a retrospective cohort study at a comprehensive perinatal center.

BACKGROUND: Globally, even within developed nations, specific regions exhibit disproportionately high rates of adolescent pregnancy that require comprehensive perinatal medical care, including social interventions. We aimed to compare perinatal outcomes between pregnant adolescents and pregnant adults in rural Japan. METHODS: We conducted a retrospective cohort study of pregnant women between 2014 and 2025, categorizing participants into two groups based on maternal age: pregnant adolescents (< 20 years) and adults (20–34 years). Adjusted odds ratios (aORs) were calculated using logistic regre

Steroidogenic compensation and lipid deficiency with enhanced NAD+ salvage in small-for-gestational-age placenta.

Fetal growth restriction (FGR) affects approximately 8% of pregnancies in Western countries and is characterised by complex placental adaptations at both metabolic and transcriptional levels. In this study, we integrated RNA sequencing and metabolomic analyses to investigate alterations in steroidogenesis, NAD+ metabolism and ω-3/ω-6 polyunsaturated fatty acid (PUFA) pathways in placental biopsies and trophoblast organoids. Placentas from small-for-gestational-age (SGA10 and SGA3) infants, compared with appropriate-for-gestational-age (AGA) controls, showed increased cholesterol uptake and enh

Improving the accuracy of fetal growth restriction diagnosis by integrating fetal genetic growth potential: a proof of concept study.

BACKGROUND: Fetal growth restriction (FGR) is a common obstetric complication where a fetus fails to reach its genetically determined growth potential. Current diagnostic methods rely on population-based fetal biometric percentiles, which struggle to distinguish pathological FGR from healthy small-for-gestational-age (SGA) infants and may miss cases of growth restriction in fetuses with a high genetic growth potential who fall above conventional SGA thresholds. This study aimed to develop a novel diagnostic framework for FGR by incorporating individualized genetic potential modeling to improve

Outcomes in pregnant women diagnosed with preeclampsia with and without fetal growth restriction at high altitude: A cross-sectional study.

BACKGROUND: Preeclampsia (PE) and fetal growth restriction (FGR) are major contributors to maternal and neonatal morbidity and mortality globally. The pathophysiology of these conditions is further complicated by high-altitude (HA) environments due to hypoxia-induced changes in maternal and fetal physiology. This study aimed to evaluate the maternal and neonatal outcomes of PE, with and without FGR, in a HA setting (Bogotá, Colombia, 2,640 m.a.s.l.). METHODS: This retrospective, cross-sectional study analyzed 437 patients diagnosed with PE between 2020 and 2023. Participants were stratified i

Insights from animal models: Dissecting the independent roles of oxygen and nutrients in the fetal origins of cardiovascular disease.

Depending on the pregnancy complication, substrate delivery to a developing fetus can be reduced, changing the course of fetal growth below the genetically determined in utero growth potential, resulting in fetal growth restriction (FGR). FGR is linked to an increased risk of developing cardiovascular disease (CVD) in later life. When caused by placental insufficiency, FGR is characterized by fetal hypoxaemia and hypoglycaemia due to reduced substrate supply to the fetus. However, other common pregnancy complications exist, where fetal hypoxaemia or hypoglycaemia may or may not occur. It is th

Proline Alleviates Low-Protein Diet-Induced Adverse Pregnancy Outcomes by Enriching Akkermansia and Activating Placental Wnt/Igf1 Signaling.

Alterations in gut microbiota and its metabolites are associated with placental and fetal development. Although proline has been shown to enhance placental function and improve pregnancy outcomes, its interaction with the gut microbiota remains poorly understood. In this study, a gestational low-protein diet (LPD) leads to decreased fetal birth weight and impaired placental vascular development, as well as increased embryonic resorption rates; these effects are mitigated by dietary proline supplementation. Mechanistically, proline enhances the abundance of Akkermansia and the production of but

COVID-19 lockdown and association with placental function - a quasi-experimental analysis.

OBJECTIVE: To evaluate the effect of COVID-19 lockdown measures on the occurrence of hypertensive disorders of pregnancy and fetal growth restriction in a national pregnancy population. STUDY DESIGN: This is a national registry-based quasi-experimental study, including singleton births ≥24 + 0 weeks from nulliparous women between 2016-2020. We evaluated the effect of gestational age upon entering the first nationwide lockdown period (March 9th-June 1st 2020) on outcome. This effect can be assumed to be unconfounded (a so-called quasi-experimental analysis), because this period was unexpected
