YTHDC1 Is Essential for Postnatal Liver Development and Homeostasis.

Hepatocytes play a crucial role in liver function, with their maturation occurring during postnatal development. However, the molecular mechanisms underlying this maturation are not yet fully understood. In this study, YTHDC1 is identified as a key positive regulator of hepatocyte maturation and an essential factor in maintaining liver homeostasis. YTHDC1 expression increases in the liver after birth, and hepatocyte-specific deletion of Ythdc1 impairs hepatocyte maturation, leading to reduced liver weight, liver injury, inflammation, and fibrosis. These defects contribute to the development of

Multi-organ involvement in preterm neonatal encephalopathy.

BACKGROUND: Hypoxic-ischemic encephalopathy (HIE) is a complex condition resulting from oxygen deprivation at birth. As the body redirects cardiac output to protect the brain, this can lead to multiorgan dysfunction (MOD). While most previous studies traditionally focused on term neonates with HIE, our goal was to focus on preterm neonates to study the effect on organs during critical periods of brain development. We aim to assess the incidence and severity of MOD in relation to brain magnetic resonance imaging (MRI) findings and electroencephalogram (EEG) abnormalities in this vulnerable popu

Bone mesenchymal stem cells attenuate hepatic stellate cell activation and liver fibrosis through REDD1/autophagy pathway.

BACKGROUND: Bone mesenchymal stem cells (BMSCs) have demonstrated therapeutic potential in attenuating liver fibrosis. However, the precise molecular targets through which BMSCs regulate hepatic stellate cells (HSCs) activation, as well as liver fibrosis remains unclear. METHODS: BMSCs were isolated from rat bone marrow, cultured, and characterized. BMSCs were administered via tail vein injection into bile duct ligation (BDL)-induced liver fibrosis mice. The downstream target of BMSCs was analyzed using RNA-sequencing (RNA-seq) and detected in liver tissues of Primary Biliary Cholangitis (PBC

CFH nonsense mutation-mediated pregnancy-associated atypical hemolytic uremic syndrome: Case report.

Pregnancy-associated hemolytic uremic syndrome (P-aHUS) is characterized by microvascular hemolytic anemia, thrombocytopenia, and acute organ damage, particularly acute kidney injury, occurring during pregnancy or in the postpartum period. This rare disease has been associated with mutations in genes that regulate the complement system in most reported cases. This article introduces a 38-year-old maternal, who gave birth again after 13 years. Approximately four days post-cesarean section, she developed severe anemia, thrombocytopenia, renal impairment, and abnormal liver function, prompting ur

PXR mediates maternal cholestatic liver injury and associated fetal dysplasia induced by antenatal dexamethasone exposure.

Previous studies have focused on advantages and disadvantages of antenatal dexamethasone therapy (ADT) on offspring development, but the effects on the mother have yet to be reported. In this study, the clinical cohort study found that ADT caused maternal susceptibility to cholestatic liver injury, which was manifested by elevated plasma total bile acid (TBA) levels, altered bile acid metabolic profiles and changed gut microbiota (mainly characterized by increased Bacteroides), accompanied by fetal dysplasia associated with high bile acids. Animal experiments confirmed that prenatal dexamethas

Role of Iron Metabolic Disturbances and Inflammatory Iron Biomarkers in Liver Transplant Prognosis.

Iron metabolism plays a pivotal role in liver transplantation, significantly impacting outcomes for both donors and recipients. The liver, central to iron homeostasis, is often impaired in chronic liver diseases leading to metabolic disorders that exacerbate liver damage. Liver transplantation (LT) is a critical treatment for end-stage liver diseases, with iron status in both donors and recipients influencing post-transplant outcomes. Studies indicate that pre-transplant iron overload in recipients is associated with poor liver function recovery, increased graft rejection risk, and reduced pat

Prenatal Interventions for Congenital Diaphragmatic Hernia: A Systematic Review.

Life-threatening congenital diaphragmatic hernia (CDH) is a defect with subsequent herniation of abdominal organs. Herniation of abdominal organs into the thoracic cavity causes pulmonary hypoplasia and hypertension. Despite improved baby critical care and surgical therapy, severe CDH has high mortality and morbidity rates. The prenatal intervention of fetoscopic endoluminal tracheal occlusion (FETO) has become popular for improving postnatal survival and lung development. However, its efficacy in reducing maternal risks, morbidity, and improving newborn survival is still debated. This systema

SOX9 Is a Key Factor for the Postnatal Maturation of the Intrahepatic Bile Duct Network.

AIM: The sex-determining region Y-box 9 (SOX9) plays a critical role in the development of intrahepatic bile ducts (IHBDs) during the embryonic stage. However, its role in postnatal IHBD maturation remains unclear. This study aimed to investigate the function of SOX9 in the postnatal development of the bile duct network, with particular emphasis on its role in structural organization. METHODS: Three-dimensional imaging analysis was conducted using a mouse model in which SOX9 deficiency is predominant after birth to assess the structural organization of IHBDs. Additionally, transcriptional pro

The composition, influencing factors, and physiological functions of bifidobacteria in the infant gut: a review.

Bifidobacteria are naturally found in the human gut and quickly establish dominance shortly after birth, playing a crucial role in the development and stability of the infant gut microbiota. A growing body of research suggests that host and environmental factors shape the colonization and the relative abundance of bifidobacteria in the infant gut during early life. Understanding the factors that influence bifidobacterial colonization and maintaining normal colonization levels are keys to ensuring gut health. Bifidobacteria exhibit antibacterial, anti-inflammatory, and pathogen-resistant proper

Temporal transcriptional and functional dynamics of jejunal mucosal response to early weaning in piglets.

Early-life adversity, including abrupt weaning, imposes significant psychosocial and environmental stress during a critical window of gastrointestinal (GI) development, leading to long-term consequences for gut function and disease susceptibility. In piglets, early weaning profoundly disrupts GI development, altering the intestinal epithelial barrier, reshaping immune function, and inducing lasting changes in the enteric nervous system. Despite these adverse outcomes, the early molecular mechanisms that initiate these alterations and set the gut on a divergent developmental trajectory remain p

Aluminum adjuvant promotes liver inflammation and fibrosis in mice: A novel approach to establish a liver fibrosis animal model.

BACKGROUND AND AIMS: Liver fibrosis is a prevalent pathological stage of various chronic liver diseases and has the potential to progress to liver cirrhosis and hepatocellular carcinoma. However, experimental models for in vivo research are limited. Unexpectedly, increased liver inflammation and fibrosis were previously observed in mice treated with aluminum adjuvant (commercial Imject Alum, a mixture of Al(OH)3 and Mg(OH)2). Our study aimed to reveal the pathogenesis and pathological features of Imject Alum-induced liver injury and evaluate its potential as an experimental model of fibrotic l

7,8-Dihydroxyflavone protects acetaminophen induced liver injury through activating PI3K/Akt/NRF2/GPX4 mediated ferroptosis suppression.

Acetaminophen (APAP)-induced liver injury remains a leading cause of acute liver failure, with limited therapeutic options beyond the narrow therapeutic window of N-acetylcysteine. Emerging evidence highlights ferroptosis as a critical mediator of APAP hepatotoxicity. Here, we investigated the hepatoprotective effects of 7,8-dihydroxyflavone (7,8-DHF), a natural flavonoid with antioxidative properties, against APAP-induced liver injury and explored its underlying mechanisms. In a murine model of APAP overdose, 7,8-DHF administration significantly attenuated liver damage, as evidenced by reduce
