Acute Acalculous Cholecystitis Complicating Typhoid Fever: A Case Report.

Typhoid fever continues to represent a significant public health concern in the developing world. With the rise in global travel, increasing population diversity, and the emergence of multidrug-resistant strains, clinicians must remain vigilant to both the disease and its potential complications. Among these, acalculous cholecystitis is an uncommon but clinically important complication. While the majority of reported cases in the literature involve paediatric patients, reports in adults remain scarce. We present the case of a 25-year-old traveler who developed acute acalculous cholecystitis as

Detection of Acute Tropical Fever Pathogens via Multiplex Polymerase Chain Reaction: A Cross-Sectional Study to Guide Appropriate Antimicrobial Use in Communities.

BACKGROUND: Pakistan faces a substantial burden of tropical diseases, including malaria, dengue, and typhoid fever, particularly in Sindh Province, where recent outbreaks and floods have further increased disease incidence, highlighting the urgent need for improved diagnostics and disease control. Currently, diagnosis relies on multiple laboratory tests, contributing to frequent empirical antimicrobial use due to many undiagnosed or misdiagnosed cases. This study aimed to validate a rapid molecular test for the early diagnosis of malaria, dengue, and typhoid infections, thereby facilitating ta

Characterization of Ty21a immunostimulatory effects in the mouse bladder.

Intravesical treatment with Salmonella enterica Ty21a, an oral typhoid-fever vaccine, has shown therapeutic potential against bladder tumors mainly through local immune-cell recruitment, particularly CD8+ T-cells. However, the mechanisms underlying its efficacy and the impact of bacterial formulation remain unclear. Here, we show that increased immune-cell infiltration was neither associated with modification in blood vessel density nor the generation of high endothelial venules, but rather with a transient increase in local vessel permeability, requiring live bacteria. Giving prior evidence t

Whole genome sequencing as a reliable alternative for Salmonella serotyping: a comparative study with the gold-standard method.

INTRODUCTION: Salmonella is a major foodborne pathogen of significant global health concern, leading to conditions like gastroenteritis, with symptoms such as diarrhea, abdominal cramps, and fever. Although most cases are self-limiting, Salmonella enterica can cause bacteremia or typhoid fever in some cases. There are over 2600 known serotypes of S. enterica. Each serotype is distinguished by differences in its surface antigens (somatic O and flagellar H), which is the basis for their classification. Considering that traditional phenotypic typing is laborious and time-consuming, the goal of th

Overcoming multidrug- and extensively drug-resistant Salmonella Typhi with natural product-metal synergy: Evidence of DNA polymerase inhibition and cell membrane disruption.

The declining efficacy of azithromycin-the primary oral treatment for typhoid fever in XDR-endemic regions-highlights the urgency against drug-resistant Salmonella Typhi. In response, we developed two ƞ6-arene ruthenium(II) series (9-20 and 37-45) by integrating natural product-based antimicrobial motifs with the DNA-intercalating/ROS-generating features of Ru-scaffolds. Among these, complex 11 and 16 exhibited potent activity against MDR and XDR S. Typhi, with lead complex 16 showing MIC90 as low as 0.06 μg/mL and MBC values significantly superior to azithromycin. Time-kill kinetics showed ra
