Enteric fever caused by Salmonella enterica serovars Typhi and Paratyphi A and, to a lesser extent, S. Paratyphi B and C, remains a significant cause of mortality and morbidity in resource-constrained settings. Typhoid conjugate vaccines (TCVs) protect against S. Typhi but no vaccine to date protects against paratyphoid fever. There are several bivalent S. Typhi/Paratyphi A products in development; however, the low incidence of paratyphoid fever in many settings limits the feasibility of phase 3 efficacy studies. Two bivalent vaccines adding the S. Paratyphi A-specific O:2 lipopolysaccharide c

RATIONALE: Typhoid fever is a major cause of enteric disease-related morbidity and mortality. Vaccination reduces disease burden and prevents outbreaks, but policies and programmes should be informed by the most recent evidence as newer vaccines become available.
OBJECTIVES: To assess the benefits and harms of typhoid conjugate vaccines (TCVs) compared to no vaccine, placebo, typhoid-inactive agents (vaccines for another disease) or other typhoid vaccines for preventing morbidity and mortality associated with typhoid fever in adults and children.

Typhoid fever, primarily caused by Salmonella Typhi, can result in severe life-threatening complications such as encephalopathy. Here we elucidate the mechanisms by which typhoid toxin, a unique virulence factor of S. Typhi, mediates the neuropathology associated with typhoid fever. Utilizing mice engineered to have specific tissues protected from toxin action and an in vitro model of the blood-brain barrier (BBB), we demonstrate that, rather than direct action on neuronal or glial cells, typhoid toxin causes neuropathology by disrupting the BBB. Intravenous tracer studies confirmed significan

Typhoid has been a major infectious health threat for the masses in Pakistan with increasing antimicrobial resistance, socio-economic decline, and environmental challenges. Since 2019, the percentage of cases has reduced with the implementation of Typhoid Conjugate Vaccine (TCV) program but reports of widened geographical spread particularly as aftermath of flooding in 2022 and resistance to Azithromycin or Meropenem in adult population are a matter of serious concern. Now is the right time to reconsider whether strategies like mass immunization or health infrastructure development solely can

BACKGROUND: Salmonella enterica subspecies enterica serovar Typhi (abbreviated as 'Typhi') is the bacterial agent of typhoid fever. Effective antimicrobial therapy reduces complications and mortality; however, antimicrobial resistance (AMR) is a major problem in many endemic countries. Prevention through vaccination is possible through recently-licensed typhoid conjugate vaccines (TCVs). National immunisation programs are currently being considered or deployed in several countries where AMR prevalence is known to be high, and the Gavi vaccine alliance has provided financial support for their i

BACKGROUND: Carbapenem resistance in Enterobacterales is a growing public health concern, primarily driven by carbapenemase enzymes such as OXA-48, VIM, NDM, and IMP. Among these, New Delhi Metallo-β-lactamase (NDM) has disseminated widely across various Enterobacterales species, including Salmonella Typhi, though reports remain rare.

Extensively drug-resistant (XDR) typhoid, caused by Salmonella enterica serotype Typhi, has emerged as a critical global health security threat, with Pakistan, particularly Sindh province, at its epicenter. The misuse of antibiotics, inadequate diagnostic tools, and poor water and sanitation infrastructure have created ideal conditions for the rise of antimicrobial resistance (AMR). XDR typhoid strains resistant to multiple first-line antibiotics have been linked to environmental contamination, with urban areas like Karachi demonstrating high rates of waterborne transmission. Enteric fever is caused by Salmonella enterica serovar Typhi (S. Typhi) and Salmonella enterica serovar Paratyphi (S. Paratyphi) A, B, and C. Globally, an estimated 11 to 21 million cases of typhoid and paratyphoid fever occur annually, with approximately 130,000-160,000 deaths, most of which are reported in South/Southeast Asia and sub-Saharan Africa. The antibiotic susceptibility of S. Typhi strains varies between countries within broad limits, from 3% to 97% for ampicillin, 9% to 95% for ciprofloxacin, 4% to 94% for chloramphenicol (India vs. Pakistan), and 0% to 99% for ceftriaxone (India

Salmonella enterica serovar Typhi (S. Typhi) produces outer membrane vesicles (OMVs) that remain comparatively underexplored as potential biotechnological tools. Here, we investigated how hypervesiculating S. Typhi mutants (ΔtolR and ΔdegS) can be engineered to load and deliver the fluorescent reporter protein mCherry, targeting human epithelial cells and the murine immune system. Deletions in tolR and degS led to distinct OMV phenotypes characterized by higher vesicle production and altered cargo composition, underscoring the impact of disrupted membrane integrity and envelope stress on OMV b

Typhoid fever is one of the common enteric fevers in developing countries, especially in emerging metropolitan areas in Indonesia. Yet, studies on spatial and temporal distribution of tyhoid fever are lacking. This study was conducted to analyze retrospective hospital-based data at the village level over the period 2017-2023 to understand the spatial and temporal variation of typhoid fever in Jakarta. Spatial analyses were performed by Moran's I and Local Indicators of Spatial Association (LISA) to examine spatial clustering of typhoid incidence and to identify high-risk villages for typhoid f

INTRODUCTION: Salmonella Typhi (S. Typhi), a Gram-negative, serves as the etiological agent of typhoid fever. In contrast to other Salmonella serovars, S. Typhi exclusively infects humans. However, the molecular interactions it engages in with the host immune system remain inadequately characterized. This study adopts a multi-omics strategy to elucidate the immune and metabolic dynamics within the murine spleen during S. Typhi infection.
OBJECTIVES: To identify and analyze transcriptomic, proteomic, and metabolomic alterations in the spleens of mice infected with S. Typhi. By comparing these

Malaria and typhoid fever remain major health issues in developing countries like Cameroon, with frequent co-infections. The potential liver damage caused by their respective causative pathogens is overlooked in health management, posing a significant risk of severe liver injury and worsening patient conditions. Accordingly, this study investigated the risk factors associated with liver damage in febrile patients with malaria, typhoid fever, or co-infection at the Obili District Medical Center and Mvog-Betsi Dominican Hospital Center, Yaoundé, Cameroon. Over 8 months, 350 febrile patients were

UNLABELLED: Salmonella enterica serovar Typhi is the causative agent of typhoid fever, a human-restricted systemic infection. The rapidly disseminating multi-drug resistant H58 subclade is endemic in Africa, Asia, and Oceania, yet little is known regarding its intracellular behavior and virulence properties. It was of interest to understand the H58 response to host cell infection in terms of its response to acid stress and subsequent changes in gene regulation. We compared the H58 response in vitro and during infection of THP-1 human macrophages with the well-characterized response of Salmonel

UNLABELLED: Antimicrobial resistance (AMR) in Salmonella Typhi (S. Typhi) is a serious health issue that can complicate typhoid fever treatment and increase fatality rates. The National Centre for Disease Control, Government of India, established a National Antimicrobial Surveillance Network to detect AMR in high-priority infections, including S. Typhi throughout geographical areas of India, and understand AMR dissemination and possible treatment options. The Gujarat State Biotechnology Mission, Government of Gujarat, India, has funded the "Network Program on Antimicrobial Resistance, Superbug

Typhoid fever cases and carriers can transmit Salmonella enterica serovar Typhi (S. Typhi) through fecal shedding. It remains unclear whether the S. Typhi shedding by carriers exhibits similar phenotypic and genotypic characteristics to those from acute cases. We investigated multidrug resistance in S. Typhi from individuals residing in urban informal settlements in Nairobi, Kenya. We recruited participants ≤ 65 years from six health facilities and tested for typhoid infection through blood and stool cultures. The S. Typhi culture-positive cases were treated and followed up after treatment, wh

BACKGROUND: Enteric fever remains a serious global health challenge. Effective control of enteric fever depends on clean water and proper sanitation, which remains unattainable in resource-limited settings. Vaccination is therefore one of critical preventive measures. Nevertheless, there exists a paucity of systematic reviews integrating pathogenesis, the protective mechanisms, and translational vaccinology developments associated with enteric fever.
AIM OF REVIEW: To enhance the mechanistic understanding of enteric fever, this review summarizes the recent advances of pathogenesis, protective

Typhoid fever is a significant health concern in low- and middle-income countries, with millions of cases and thousands of deaths each year. Typhoid Conjugate Vaccines, such as Typbar TCV®, have been developed to provide longer-lasting protection for young children. This publication includes the readouts from two different studies in Children and elderly: a Comparator Study and Age Extension Study. In the first study, children are included, whereas the latter study also includes elderly subjects. A total of 340 children were enrolled, and a subset received a booster dose of Typbar TCV®, 180 da

Typhoid fever remains a major public health threat to Low- and Middle-Income (LMIC) countries. The typhoid conjugate vaccine (TCV) is a promising vaccine platform to avert the disease as they elicit sufficient immune response in both adults and children. However, data on long-lasting immune response among children below 2 years are very limited. In this article we present the data collected from healthy Filipino children who were administered with Vi-DT vaccine. In this long-term follow-up study, we included children who had been vaccinated with Vi-DT vaccine and received a booster dose either

BACKGROUND: Many clinically relevant bacterial pathogens are encapsulated, as exemplified by Salmonella enterica serovar Typhi. S. Typhi, the causative agent of the life-threatening systemic disease typhoid fever, expresses Vi as the outermost surface glycan that protects the bacteria from host immune responses. Drug resistant S. Typhi strains and Vi variants are spread globally.
METHODS: Whole genome sequences (WGS) of 24,206 S. Typhi clinical isolates were analysed for genetic traits for antibiotic resistance. Rifampin treated S. Typhi was characterised at the molecular and cellular level.
