Phosphatidylcholine biosynthesis pathways in Cryptococcus neoformans: functional interplay and impact on virulence.

As fungal diseases emerge, new studies aim to understand how different metabolic pathways, including the biosynthesis of phospholipids, influence the fungal pathogenicity. Therefore, to investigate the role of phosphatidylcholine (PC) in the biology of the human fungal pathogen Cryptococcus neoformans, a double mutant lacking OPI3 (phosphatidylethanolamine methyltransferase) from the de novo pathway, and PCT1 (choline phosphate cytidylyltransferase) from the salvage Kennedy pathway was generated using double-joint PCR coupled with biolistic technique for gene deletion. Phenotypic and virulence

Headache as a Predictor of Cryptococcal Meningitis in Ambulatory Patients With Symptomatic HIV-associated Cryptococcal Antigenemia.

BACKGROUND: People with HIV-associated cryptococcal antigenemia are at high risk for meningitis and death. In resource-limited settings, lumbar puncture to evaluate for meningitis is infrequently performed in asymptomatic individuals; therefore, symptomatic individuals are prioritized. We evaluated the predictive value of meningeal symptoms for cryptococcal meningitis in people with cryptococcal antigenemia. METHODS: We conducted a secondary analysis from 3 randomized clinical trials conducted in Uganda between 2017 and 2024. We included adults with meningeal symptoms who had cerebrospinal fl

The cyclin dependent kinase (CDK)7 inhibitor BS-181 inhibits pathogenic Cryptococcus species, causing G2/M arrest and a splicing defect.

The fungal priority pathogen and basidiomycete, Cryptococcus neoformans (Cn), causes lung and brain infection in predominantly immuno-compromised individuals and there is an urgent need for new treatment options. The pyrazolopyrimidine-based cyclin dependent kinase (CDK)7 inhibitor, BS-181, has anticancer properties, but its antifungal activity has not been investigated. We show that cryptococcal CDK7 more closely resembles the human enzyme than that of ascomycetes, and that BS-181 inhibits its activity. BS-181 inhibited growth of both Cn and Cryptococcus gattii (Cg), but not ascomycete fungi

Presentation and prognosis of cryptococcosis requiring intensive care unit admission in France: the CRYPTO-ICU study.

BACKGROUND: Cryptococcus causes life-threatening opportunistic infections in immunocompromised hosts. Data on the most severe cases requiring ICU admission remain limited. METHODS: We conducted a retrospective, multicenter study of patients admitted to 30 French ICUs for severe cryptococcosis between 2000 and 2022. RESULTS: Among 151 patients included, 56.9% were patients with HIV. Cases in patients without HIV became increasingly prevalent over time (51.3% in 2012-2022 vs. 32.4% before 2012); 82.5% were receiving immunosuppressive therapy. Central nervous system infection was predominant (9

Metabolic plasticity and virulence of Cryptococcus neoformans are regulated by mitochondrial homeostasis.

The mitochondrion is a versatile organelle involved in diverse processes, such as cell death, metal homeostasis, plasma membrane and cell wall integrity, stress response, oxygen concentration, temperature, and metabolic adaptation, in addition to its role in generating energy. Consequently, mitochondrial fitness is essential for the pathogenicity of various organisms, including fungi. Cryptococcus neoformans is a fungal pathogen responsible for over 180,000 HIV-related deaths each year. In this study, we analyzed C. neoformans metabolic plasticity when grown with non-fermentable carbon sources

Roles of Mutation, Ploidy, and Recombination in Adaptive Evolution in Two Divergent Model Yeasts.

Genetic variation underlies the capacity of populations to adapt, yet what drives how this variation is generated and maintained in natural populations remains poorly understood. Fundamental processes such as mutation, ploidy, and recombination are known to shape genetic variation and adaptive potential but are typically studied in isolation and under controlled laboratory conditions. How these processes act together under varying environmental conditions to structure genetic variation across complex natural populations remains unresolved. In yeasts, these processes are dependent on reproducti

Strain-specific variation in virulence and host-immune modulation across the Cryptococcus gattii species complex.

Cryptococcus gattii is an environmental yeast capable of causing severe infections in immunocompetent hosts. Its virulence mechanisms remain less characterized than those of C. neoformans. This study aimed to evaluate the expression of major virulence factors and the immunomodulatory potential of eight C. gattii strains representing five genotypes and multiple sequence types (STs). Virulence traits-including capsule thickness, melanin and laccase production, phospholipase, hemolysin, urease, and DNase activities-were assessed using standardized in vitro assays. In vivo pathogenicity was evalua

In vitro effects of IFN-γ, IL-4 and IL-27 on human bronchial epithelial cells infected with Cryptococcus neoformans sensu stricto.

BACKGROUND: The innate immune response and cytokine milieu in airway mucosa, mediated by bronchial epithelial cells, are critical in determining susceptibility or protection against cryptococcosis. In experimental models, Th2 and Th1 responses are linked to susceptibility and protection, respectively, while the roles of other cytokines remain less understood. AIMS: To evaluate the in vitro effects of IL-4, IFN-γ, and IL-27 (100ng/mL) on human bronchial epithelial cells (BEAS-2B) infected with a strain of Cryptococcus neoformans sensu stricto (multiplicities of infection [MOI] 1-100). mGem: Progress on the development of conjugate vaccines for Cryptococcus neoformans infections.

Cryptococcus neoformans is an important fungal pathogen that causes cryptococcosis, a life-threatening disease predominantly affecting immunocompromised individuals. Current estimates suggest that cryptococcosis is responsible for over 150,000 deaths annually, highlighting the need for the development of effective preventive measures. Early research utilizing polysaccharide filtrates (CneF) laid the groundwork for polysaccharide-containing vaccines, demonstrating partial protection in murine infection models. Further vaccine constructs made with purified extracellular polysaccharides (EPS) con

Cryptococcus neoformans infection presenting as a mediastinal mass in an immunocompetent child with parrot exposure: a case report and literature review.

Cryptococcus neoformans typically causes pulmonary or central nervous system (CNS) infections, but mediastinal mass as its primary manifestation is rare-especially in immunocompetent children with pet parrot exposure. This study reports a 7-year-old girl who presented with recurrent fever and a mediastinal mass secondary to Cryptococcus neoformans infection, with a 5-month history of daily contact with parrot feces. Conventional diagnostic tests (e.g., fungal culture, serology) were negative, and the diagnosis was confirmed by targeted metagenomic next-generation sequencing (tNGS) of bronchoal

Current opinion on the potential role of liposomal amphotericin B in underexplored clinical scenarios.

SUMMARYThe increasing incidence of invasive fungal infections caused by resistant Candida species and molds presents a growing clinical challenge, particularly in immunocompromised patients. While echinocandins and azoles remain the cornerstone of antifungal therapy, their use may be limited by increasing reports of resistance, tolerance, drug-drug interactions, and hepatotoxicity. Liposomal amphotericin B (L-AmB), a broad-spectrum antifungal agent with a more favorable safety profile than conventional amphotericin formulations, may offer a valuable alternative in selected high-risk scenarios.

Antifungal activity of emodin against the Cryptococcus neoformans/Cryptococcus gattii complexes: an effect on the enzyme superoxide dismutase.

The Cryptococcus neoformans/Cryptococcus gattii species complexes include encapsulated yeasts that cause cryptococcosis. Emodin has demonstrated antimicrobial activity. This study evaluated emodin's antifungal activity against both planktonic cells and mature biofilms of the Cryptococcus neoformans/Cryptococcus gattii complexes, and its interaction with superoxide dismutase (SOD). Planktonic susceptibility was assessed by broth microdilution (CLSI; M27-A4). Mature biofilms were treated with emodin (128-1024 µg mL-1) and evaluated by crystal violet staining for biomass and the [3-(4,5-dimethylt

Dysbiosis of the Gut-Lung Axis and Its Immune Correlates During Pulmonary Cryptococcus neoformans Infection.

Cryptococcus neoformans is a major fungal pathogen responsible for life-threatening meningitis, especially in immunocompromised individuals. Although the gut-lung axis is known to regulate immune responses in respiratory infections, its role in cryptococcosis remains unclear. This study aimed to define the dynamic changes in the gut and lung microbiota and their relationship with host immunity during C. neoformans infection. Using a mouse model, we found that pulmonary infection induced significant dysbiosis in both the lung and gut microbiota, marked by decreased beneficial commensals and inc

Human Dectin-1- and Dectin-2-targeted DectiSomes are effective against diverse pathogenic fungi.

Annually, aspergillosis, candidiasis, cryptococcosis, and mucormycosis result in approximately 1,500,000, 650,000, 120,000, and 59,000 deaths, respectively. Mortality rates among patients receiving antifungal drug treatment range from 30% to 90%. Therefore, there is an urgent need to improve the efficacy of antifungal drug therapies against infections by these high-priority fungal diseases. Aspergillus fumigatus, Candida albicans, Cryptococcus neoformans, and Rhizopus delemar are the most common causative pathogens. We have previously developed DectiSomes, which are liposomes loaded with antif

GM-CSF orchestrates monocyte and granulocyte responses to Cryptococcus gattii.

Cryptococcus gattii is an emerging fungal pathogen that is acquired through the respiratory tract and causes invasive infections in both immunocompromised and otherwise healthy people. Many of these apparently immunocompetent patients are subsequently found to have autoantibodies against the pleiotropic cytokine GM-CSF. In this study, we investigated the potential role of GM-CSF (or CSF2) in the host response to C. gattii using a murine model of infection. Interestingly, infected Csf2-/- mice were found to have significantly improved survival and decreased lung fungal burden compared to wild t
