PURPOSE: To evaluate early changes of chorioretinal scars and optic disk in congenital Zika syndrome (CZS).
METHODS: This prospective cohort study included 35 eyes of children with CZS who presented with chorioretinal scars, which were assessed by retinal imaging at diagnosis and at 1 year's follow-up. ImageJ software was used to measure the chorioretinal scar area. A comparison board was created to classify the lesions with regard to pigmentation. The vertical (v) and horizontal (h) cup:disk ratios (CDRs) were rated, and the CDR means at baseline and follow-up were analyzed. Ocular ultrasoun

The placenta plays a critical role during pregnancy, enabling maternal-fetal exchange while serving as a barrier against pathogens. However, some viruses, including human cytomegalovirus (HCMV) or Zika virus (ZIKV), can cross this barrier and cause congenital infections, leading to severe fetal outcomes such as growth restriction, impairment of brain development, or fetal demise. Understanding how viruses infect and disseminate through the placenta is therefore essential. To investigate viral vertical transmission in a physiologically relevant context, an ex vivo model using human placental ex

Viral infections can significantly affect the physiopathology of reproductive organs, leading to fertility problems, reducing the success rates of assisted reproductive technologies, and negatively impacting pregnancy. This review aims to summarize the current evidence on viral pathogens that are either suspected or confirmed to play a role in reproductive medicine and their effects on early pregnancy. For instance, viral hepatitis and human immunodeficiency virus can decrease sperm quality. Human papilloma virus infection in men appears to cause infertility, while herpesviruses pose a greater

Transplacental infections such as cytomegalovirus, Zika virus, toxoplasmosis, and rubella, especially if acquired in the perinatal period, are well-known causes of congenital anomalies. However, their phenotypic overlap with certain genetic syndromes can pose significant diagnostic challenges during fetal autopsy and prenatal counseling. With an objective to review and highlight the clinical and pathological similarities between congenital infections and genetic syndromes, and emphasizing diagnostic pitfalls in fetal autopsy and the importance of integrated approaches, a comprehensive literatu

Congenital infections are an ongoing relevant cause of congenital and perinatal abnormalities since effective vaccination is not available for many diseases. Moreover, climate change, mutational evolution of pathogens, or their vectors associated with intense human traveling predispose to outbreaks and modification of the geographic distribution of traditionally limited infectious diseases. Here, we review the so-called TORCH infections (toxoplasmosis, others, rubella, cytomegalovirus, herpes) with a special emphasis on the arboviral infections (zika, dengue, chikungunya, oropouche). Although

The current century has been associated with the outbreaks of emerging and re-emerging viral infections such as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), monkeypox virus (MPXV), and Zika virus (ZIKV). Although their common clinical manifestations are currently well-known, the urogenital system is gaining attention as a significant, though often underappreciated, anatomical site for emerging and re-emerging viral infections, disease development, and transmission. The present review aims to comprehensively discuss the urogenital complications linked to the infections caused b

The previous literature shows mixed conclusions regarding the risk of adverse perinatal outcomes in pregnant women with symptomatic chikungunya, dengue, and Zika. We investigated this topic using a linked population-based Brazilian cohort from 2015 to 2020. The study included 6,993,395 live births. Among these, 6066 ( < 0.1%) mothers were notified with chikungunya during pregnancy, 19,022 (0.3%) with dengue, and 8396 (0.1%) with Zika. Symptomatic maternal chikungunya was associated with an increased risk of preterm birth (Hazard ratio: 1.10, 95%CI 1.01-1.22), low Apgar score 5' (1.44, 1.14-1.8

Arboviral infections in paediatric populations present unique challenges due to distinct pathophysiological mechanisms influenced by developmental and immunological differences. Commonly implicated arboviruses include dengue virus (DENV), Zika virus (ZIKV), chikungunya virus (CHIKV), West Nile virus (WNV), and yellow fever virus (YFV). These viruses exhibit specific tropisms, targeting organs such as the central nervous system (CNS), liver, and vasculature. Immune responses in children, characterised by an underdeveloped adaptive system and enhanced innate immunity, can exacerbate inflammation

BACKGROUND: During the Zika virus (ZIKV) epidemic in Brazil, regional disparities in yellow fever (YF) vaccination coverage and microcephaly incidence led to the hypothesis that maternal YF vaccination could protect against ZIKV infection and microcephaly.
METHODS: This case-cohort study was conducted in Campo Grande, Brazil, from 2018 to 2022 and included children with confirmed in utero ZIKV exposure (2015-2018) and a matched control group, totaling 129 mother-child dyads. Maternal blood samples were collected to assess ZIKV exposure and neutralizing YF vaccine-induced antibody (NAb) titers

Zika virus (ZIKV) is typically mild in humans but can cause severe congenital defects when contracted during pregnancy. Chimeric live-attenuated vaccine candidate YF-ZIK previously showed protective efficacy against lethal infection and developmental abnormalities in mice after a single dose. Here we demonstrate that YF-ZIK is safe, induces antiviral immunity, and protects rhesus macaques against high-dose experimental challenge. A single subcutaneous dose elicits neutralizing antibodies within 7-14 days, boosted by a second dose at 4 weeks. Passive serum transfer protects AG129 mice, supporti

Mosquito-borne flaviviruses, such as Zika virus (ZIKV) and dengue virus (DENV), cause diverse severe clinical manifestations including fever, rash, hepatitis, arthralgia, and congenital anomalies. Here, we identified a host factor, the adaptor protein complex 1 gamma 1 subunit (AP1G1), which plays an important role in both ZIKV and dengue virus 2 (DENV2) infections. We explored the role of AP1G1 in ZIKV and DENV2 infections using CRISPR/Cas9 gene editing technology and RNA interference (RNAi) techniques. Knockout or silencing of AP1G1 decreases the replication of ZIKV and DENV2 in multiple hum

BACKGROUND: Zika virus (Orthoflavivirus zikaense), a mosquito-borne virus in the family Flaviviridae and genus Orthoflavivirus, has garnered international attention due to its neurological and congenital impacts. Although endemic to Africa, its presence in Nigeria remains poorly understood and often overshadowed by other febrile illnesses such as malaria and dengue. This review synthesizes peer-reviewed literature published between 2015 and 2025 to explore the epidemiology, diagnostic challenges, and public health implications of ZIKV in Nigeria.

Zika virus is spread to human populations primarily by Aedes aegypti mosquitoes, and Zika virus disease has been linked to a number of developmental abnormalities and miscarriages, generally coinciding with infection during early pregnancy. In this paper, we propose a new mathematical model for the transmission of Zika and study a range of control strategies to reduce the incidence of affected pregnancies in an outbreak. While most infectious disease models primarily focus on measures of the spread of the disease, our model is formulated to estimate the number of affected pregnancies throughou

The 2015-2016 Zika virus (ZIKV) epidemic underscored the severe consequences of congenital Zika syndrome (CZS) and the broader challenges posed by neurotropic flaviviruses. As key mediators of cytotoxic immunity, CD8 T cells play a crucial and multifaceted role in ZIKV pathogenesis. While essential for controlling viral replication, their infiltration into the central nervous system (CNS)-an immune-privileged site-raises potential concerns regarding immunopathology. This review explores the dual roles of CD8 T cells during ZIKV infection, emphasizing both their antiviral functions and their po

Infection by the Zika virus (ZIKV) during pregnancy can cause congenital Zika syndrome (CZS) or other central nervous system conditions in infants, underscoring the importance of understanding the role of ZIKV-induced placental damage in the development of CZS. Here, we established a ZIKV-infected pregnant mouse model and examined the pathological changes in the placenta following ZIKV infection. We found that ZIKV infection induces severe placental inflammation associated with trophoblast PANoptosis. Specifically, ZIKV activates RIG-I and recruits ASC, caspase-1, NLRP3, caspase-8, and RIPK1 t

Intracranial calcifications (ICCs) are a characteristic neuropathological feature of several congenital viral infections, including Zika virus (ZIKV), cytomegalovirus (CMV), and lymphocytic choriomeningitis virus (LCMV). These lesions are linked to severe neurodevelopmental outcomes, such as microcephaly, epilepsy, and cognitive deficits, yet the mechanisms underlying their formation and resolution remain unclear. ICCs are thought to arise from an imbalance in osteogenic and osteolytic signaling in the developing brain. Recent work implicates pericytes as key targets of ZIKV, capable of osteog

West Nile virus (WNV), Zika virus (ZIKV), and Dengue virus (DENV) are vector-borne diseases endemic in tropical and subtropical countries around the world. Their incidence has been growing in recent years and they are becoming increasingly relevant even in non-endemic areas, representing a significant public health problem worldwide. Globalization and climate change have led to drastic changes and radical transformations in the development of an ecosystem capable of sustaining the life cycle of viruses even in urban environments, where transmission occurs within an entirely susceptible populat
