Kinetics of neutralizing and binding antibody responses following Zika virus infection during pregnancy: A nested analysis of participants from the microcephaly epidemic research group (MERG) and Zika in infants and pregnancy (ZIP) cohorts.

ZIKV infection during pregnancy can result in Congenital Zika Syndrome (CZS) in the newborn. Nevertheless, diagnosing ZIKV infection during pregnancy remains challenging, and the kinetics of the serological response to acute infection remains insufficiently reported in the literature. Using samples collected longitudinally from two cohorts of pregnant women in Brazil, this study aims to estimate the timeframe to reach anti-ZIKV immunoglobulin (IgM) negativity. Additionally, it seeks to describe the positivity of IgM, IgG3, and neutralizing antibodies based on the interval between symptom onset

Evaluation of Effectiveness of Autocidal Gravid Ovitraps for Preventing Zika Virus Infection, Puerto Rico, USA.

Aedes aegypti mosquitoes drive arboviral outbreaks in tropical regions. Zika virus (ZIKV), linked to congenital and neurologic complications, caused a major outbreak in Puerto Rico, USA, in 2016, infecting ≈26% of the population. Autocidal gravid ovitraps (AGOs), pesticide-free devices targeting gravid Ae. aegypti mosquitoes, have been shown to reduce transmission of another arbovirus, chikungunya. During March-May 2017, we conducted a household-based serosurvey in 4 demographically similar communities in southeastern Puerto Rico, 2 with long-term AGO deployment (≈85% coverage) and 2 without,

Zika virus infection induces ultrastructural alterations, mitochondrial dysfunction and oxidative stress in human trophoblast HTR-8/SVneo.

BACKGROUND: Zika fever gained importance in Brazil in 2015 due to its association with congenital syndrome. Although Zika virus (ZIKV) crosses the placenta and infects the fetus, its pathogenesis remains incompletely understood. This study investigated the effects of ZIKV infection in HTR-8/SVneo trophoblast cells. METHODS: Cells were infected with ZIKV (MOI 0.1, 0.2, or 1) or Mock control for 24 or 48 hours. Infection rate and viability were assessed by immunofluorescence and flow cytometry. Ultrastructural changes were analyzed by transmission electron microscopy. Mitochondrial membrane pot

Zika Virus-Induced Metabolic Reprogramming Drives Lipid Droplet Biogenesis, Promoting Viral Replication and Ocular Pathogenesis.

Zika virus (ZIKV) remains a significant global public health threat due to its association with severe neurological and ocular abnormalities, including microcephaly and congenital glaucoma in infants. Viruses often exploit host metabolic programs, such as energy and lipid metabolism, to support their replication. However, how ZIKV-driven metabolic reprogramming affects the anterior segment of the eye, especially trabecular meshwork (TM) cells, remains poorly defined. In this study, we investigated the roles of AMP-activated protein kinase (AMPK) signaling, fatty acid (FA) metabolism, and lipid

Discordant imaging findings in dizygotic twins with known exposure to maternal ZIKV infection during initial stages of pregnancy.

Zika virus (ZIKV) is an arbovirus commonly transmitted to humans through the bite of an infected Aedes mosquito. Clinically, ZIKV is known to cause a wide range of symptoms, from febrile rash to severe neurological conditions, with congenital microcephaly being one of the most notable. Herein, we present a case of concurrent ZIKV infection in dizygotic twins, where only one twin developed microcephaly and severe encephalic malformations, while the other exhibited normal encephalic architecture. Our findings shed light on the complex interplay between ZIKV infection and the manifestation of neu

Ten years of Zika in Brazil: achievements, challenges and perspectives.

Zika virus (ZIKV) infections have impacted public health in Brazil since 2015, primally due to Congenital Zika Virus Syndrome (CZVS) cases, which may lead to microcephaly and other clinical manifestations, such as hearing and visual impairments. More than ten years after the first diagnosis of Zika in Brazil, some progress has been made and the epidemiological scenario has improved considerably. However, despite these advances, more than a thousand ZIKV infections are reported annually in Brazil and new CZVS cases continue to be observed. Herein, we performed a retrospective and prospective an

Ten Years of Congenital Zika Syndrome: From Outbreak to a Decade of Clinical, Therapeutic, and Preventive Advances in a Tropical Disease Context.

A decade has elapsed since the first recognized cluster of congenital anomalies associated with Zika virus (ZIKV) was reported in Brazil in 2015, culminating in the formal delineation of Congenital Zika Syndrome (CZS) as a specific pattern of birth defects. This narrative review examines the ten-year trajectory of CZS as a tropical infectious disease, from its initial emergence and public health emergency declaration by the World Health Organization (WHO) in February 2016, through evolving epidemiological, clinical, and scientific understanding. CZS is characterized by a spectrum of severe neu

Vitamin A Modulates AHR Signaling and Restricts Zika Virus Replication in Human Retinal Pigment Epithelial Cells: Insights from Molecular Modeling and Antiviral Assays.

Zika virus (ZIKV) is an emerging flavivirus associated with congenital malformations and ocular complications, representing a significant public health concern. Retinal pigment epithelium (RPE) cells play a key role in maintaining retinal integrity and represent a primary target of ZIKV infection, making them a relevant model for studying host-virus interactions. In this study, we evaluated the antiviral activity of fat- and water-soluble vitamins against ZIKV in hTERT RPE-1 (hRPE1) cells. Particularly, vitamin A was identified as the compound that most effectively inhibited viral replication.

The anatomy of a public health crisis: Household responses over the course of the Zika epidemic in Brazil.

In 2015, Brazil experienced an epidemic caused by the Zika virus. We use hundreds of millions of administrative records to document household responses to the first public health alert linking the Zika virus to the risk of congenital disease for those in utero. We study two margins of behavior: risk avoidance (avoiding pregnancy), and risk mitigation during pregnancy (ultrasounds and abortions). On risk avoidance, we find a 7% reduction in pregnancies post-alert, a response triggered immediately after the alert. On risk mitigation, we find a 9% increase in the use of ultrasounds in the first t

Marine-derived ascofuranone as a novel inhibitor of Zika virus with therapeutic potential.

The Zika virus (ZIKV), a mosquito-borne Flaviviridae Orthoflavivirus, poses global health risks through its association with severe congenital and neurological complications. Despite urgent needs, no approved therapies or vaccines exist against ZIKV infection. This therapeutic gap has accelerated drug discovery efforts, where marine-derived compounds show particular promise. Their structural uniqueness and antiviral potential position marine ecosystems as vital sources for novel anti-ZIKV agents. In this study, a natural product of marine fungi, ascofuranone (ASC) demonstrated prominent antivi

Immune mechanisms of congenital Zika syndrome.

Zika virus (ZIKV) infection during pregnancy can adversely affect mother and child. ZIKV can vertically transmit from mother to fetus, especially during early pregnancy, to cause congenital Zika syndrome (CZS), which includes microcephaly, ocular and musculoskeletal defects, and other disorders. Whereas human clinical studies have provided crucial insights into how ZIKV affects pregnancy, animal models have uncovered mechanisms of fetal disease. This Review discusses fetal immunological factors that contribute to or protect from CZS, as well as the interplay between maternal and fetal immunity

Zika virus as an oncolytic therapy.

Glioblastoma (GBM) is a common, aggressive, primary brain tumor. New therapies are needed to improve outcomes in patients with this disease. One such approach which has shown preclinical promise is the use of genetically engineered Zika virus for oncolytic virotherapy. Zika virus is an enveloped, positive-sense, single-stranded RNA virus and is spread by mosquitoes of the Aedes genus. Most infections with Zika virus in adults are asymptomatic. However, primary infection with Zika virus early during pregnancy can cause fetal congenital defects. This phenomenon, termed congenital Zika syndrome,

Comparing Adverse Health Outcomes in Infants Born to Women Monoinfected with Zika Virus or Infected with Both Zika and Chikungunya Viruses during Pregnancy: A Prospective Cohort Study in Pernambuco, Brazil (2015-2017).

The association between Zika virus (ZIKV) infection in pregnant women and congenital defects is well-established. However, the consequences of infection with both ZIKV and Chikungunya virus (CHIKV) during pregnancy remain poorly understood. The aim for the present study was to estimate and compare the risk of adverse outcomes in infants born to mothers infected with ZIKV alone and those born to mothers infected with both ZIKV and CHIKV during pregnancy. A cohort study was conducted, following pregnant women with rash who were reported to the Strategic Health Surveillance Information Center of

Zika Virus in Pregnancy: Maternal Infection and Fetal Consequences.

The Zika virus has emerged as a major public health concern due to its potential to cause serious complications, particularly in pregnant women and their unborn children. Transmitted primarily by infected Aedes mosquitoes, the virus is prevalent in tropical and subtropical regions. Recent outbreaks in Pune, Maharashtra, underscore the need for vigilant preventive strategies. While most infected individuals remain asymptomatic, symptomatic cases may present with fever, rash, and joint pain. The primary concern is the risk of congenital abnormalities, notably microcephaly, associated with matern

Zika Virus Inoculation During Pregnancy Impaired Maternal Care and Altered Prolactin and Corticosterone Levels in Rats.

Zika virus (ZIKV) is an Orthoflavivirus known for its teratogenicity and for causing congenital Zika syndrome (CZS) in the offspring, but recent studies suggest a negative impact on maternal brain health and behavior following gestational ZIKV inoculation. This behavior, crucial for the offspring's neurodevelopment, is regulated by the hypothalamic-pituitary-adrenal (HPA) axis and prolactin. This study investigated the effect of gestational ZIKV inoculation on maternal care in Wistar rats and its relationship with prolactin and corticosterone levels. Fifty-six females were inoculated intraperi

Generation and characterization of an anti ITGB4 monoclonal antibody that blocks Zika virus entry.

INTRODUCTION: Zika virus (ZIKV) causes congenital disease and neurological complications, yet no approved vaccines or antivirals are available. Integrin β4 (ITGB4) has been identified as an entry receptor for ZIKV. METHODS: Recombinant human ITGB4 ectodomain was expressed using a baculovirus-insect cell system and used to immunize BALB/c mice. Hybridoma screening identified monoclonal antibody 7C3. Binding affinity to ITGB4 was determined by biolayer interferometry. Epitope relationships between 7C3 and the previously reported anti-ITGB4 antibody 13H10 were assessed by competitive binding ass

Using Paulo Freire's philosophy to share power: early insights from planning for the co-production of knowledge in research.

BACKGROUND: In recent decades, there has been a growing consensus that incorporating patient, carer, community and public participation is important. How this is accomplished remains more contested. Co-production of knowledge in research is described as one potential approach; however, its theory and practice often remain unclear. The requirement of sharing power disrupts traditional research dynamics and can be challenging in many settings. Global South epistemologies, which can be interpreted as offering a framework for understanding power sharing and empowerment, hold the potential to aid c

Morphological analysis of the hippocampus in rats exposed to Zika virus on embryonic day 9: Evidence of male vulnerability.

BACKGROUND: Congenital Zika syndrome (CZS) encompasses severe neurodevelopmental anomalies, however the impact of infection during early gestation - a critical window for neural tube closure - remains poorly understood. This study investigated some acute effects of Zika virus (ZIKV) inoculation during early gestation (gestational day 9, G9) on the hippocampus of neonate rats (postnatal day 1, PND1), focusing on volumetric changes and microglial morphology with emphasis on sex-specific differences. METHODS: Pregnant Wistar rats were intraperitoneally injected with ZIKV (1×106 PFU) or vehicle c

Impact of Day/Night Cycle Temperature Regimes on Zika Virus Replication in Mosquito Cells and Implications for Transmission Potential.

ZIKV is an emerging mosquito-borne pathogen with the unique capability to cause severe congenital abnormalities. Mosquito-borne viruses are reliant on an optimal temperature within the mosquito host to spread to humans. Few studies explore how day/night temperature variations impact virus replication within the mosquito. Most laboratory studies are conducted at constant temperatures, or with very minimal temperature oscillations. In consequence, ZIKV transmission models and risk maps utilize biological data derived from constant temperature studies. We sought to determine if the use of viral r
