Zika virus (ZIKV) is a global public health concern, mainly due to the congenital and neurological syndromes associated with the disease. The similarity of clinical symptoms to those of other flavivirus infections makes clinical diagnosis particularly challenging. Additionally, developing specific and sensitive diagnostic assays remains difficult because of the high antigenic similarity among these arboviruses, and effective immunotherapeutic treatments are still limited. In this study, four human anti-ZIKV single-chain variable fragment (scFv) antibodies were isolated by phage display through

Zika virus (ZIKV) is an arbovirus associated with neurological complications such as neonatal microcephaly and fetal anomalies, collectively referred to as congenital Zika syndrome. In the context of co-circulation with other arboviruses, it remains unclear whether pre-infection with a virus could be a risk factor for increased severity of ZIKV-induced disease. Here, we aimed to investigate whether there is differential gene expression in offspring of ZIKV-infected mothers pretreated with panflavivirus (4G2) at subneutralizing concentrations. Twelve weeks after birth, the offspring of these mo

INTRODUCTION: The 2015-2016 Zika virus (ZIKV) pandemic revealed a pathogenic potential, including Guillain-Barré syndrome and Congenital Zika Syndrome (CZS). Despite the decline in incidence, the potential for future circulation and the absence of specific antivirals underscore continued scientific interest in prophylactic vaccine development.
OBJECTIVE: To synthesize evidence from clinical trials regarding the safety and preliminary immunogenicity of current ZIKV vaccine candidates.
METHODS: This systematic review followed PRISMA guidelines (CRD420251056463). Searches were performed in MEDL

Zika virus (ZIKV) is a Flavivirus that has spread globally since its initial reporting. Transmitted to humans via multiple mosquito species, its typical symptoms (e.g., Guillain-Barre syndrome [GBS] and neonatal microcephaly) pose severe public health challenges. Based on existing research, the pathogenic characteristics of ZIKV are largely shaped by host molecular pathways, endowing the virus with an enhanced ability toward specific host cell types. On the other hand, ZIKV achieves intracellular survival and increased pathogenicity by evading the human immune system. Host innate immune respon

A "go-bag" assumes an emergency with a beginning, a peak, and an end. This editorial asks what a family needs to keep for the years after that ending - not to survive the emergency, but to survive its administrative afterlife. Drawing on a decade of research with families living with congenital Zika syndrome in Northeast Brazil, and on Alagoas's compensation law, we examine how states convert the diagnostic limits of an emergency into an evidentiary burden borne by individual families. A decade after exposure, families are asked to reconstruct causation from medical fragments never designed to

Zika virus (ZIKV) is an arthropod-borne Orthoflavivirus that caused a major outbreak in Central and South America in 2015-16. During that outbreak, ZIKV infection during pregnancy was associated with high rates of adverse outcomes, including miscarriage and a spectrum of neurological birth defects collectively termed congenital Zika syndrome (CZS). Ten years later, there are still no licensed ZIKV vaccines, underscoring the need for robust experimental models to evaluate vaccine efficacy against congenital disease. Here, we evaluated a highly pathogenic nonhuman primate pregnancy model for its

Zika virus (ZIKV) is a mosquito-borne flavivirus that caused an epidemic in the Americas in 2015-2016, accompanied by severe neurological manifestations, including congenital Zika syndrome and Guillain-Barré syndrome. Our previous data demonstrate that the protein level of karyopherin α6 (KPNA6), a transport factor in nucleocytoplasmic trafficking, increases in ZIKV-infected cells, and that KPNA6 depletion reduces ZIKV replication. Here, we found that ZIKV infection led to the relocation of KPNA6 to the perinuclear region. KPNA6 was observed to partially co-localize with double-stranded RNA, a

In recent years, Zika virus infections have spread geographically across various territories, including French Polynesia, the South Pacific, and South America, with a significant impact on Brazil. Severe Zika virus infections can lead to neurological complications, such as Guillain-Barré syndrome in adults. Furthermore, infection during pregnancy can result in microcephaly and other congenital brain malformations in the fetus. Aedes aegypti cohabits with humans and is the principal vector responsible for transmitting Zika virus to humans. Recent advances have elucidated the molecules involved

Background and objectives Zika virus (ZIKV) is a flavivirus transmitted by the bite of infected Aedes mosquito. In 2015-16, Brazil reported cases of ZIKV virus infection followed by Guillain-Barre syndrome and congenital birth defects. India has reported ZIKV virus infections sporadically since 2016, without adverse events. This prompted us to conduct this in-silico investigation and identify reasons for this variation. The objective was to study ZIKV envelope protein (E-protein) to identify possible mutations and their potential role in virus entry into the host cell. Methods Using multiple s

Oropouche virus (OROV), an emerging arbovirus of the genus Orthobunyavirus, has become a growing public health concern following its recent expansion across the Americas and its potential to cause severe clinical outcomes in maternal and child health. Although Oropouche fever has classically been described as an acute self-limited febrile illness, accumulating evidence indicates that OROV is associated with meningitis and fatal infection, as well as evidence of vertical transmission associated with adverse fetal outcomes, including microcephaly and other congenital abnormalities, spontaneous a

BACKGROUND: Oropouche virus (OROV), a neglected arbovirus of the Orthobunyavirus genus within the Peribunyaviridae family, has historically been confined to South and Central America. Recent epidemiological changes, including geographic expansion, increasing incidence, exported travel-associated cases, and reports of neurological and congenital complications, have raised concern regarding its potential for wider dissemination. Although no confirmed cases have been reported in Asia to date, future emergence remains biologically and ecologically plausible due to interconnected virological, ecolo

BACKGROUND: Zika virus (ZIKV), known for its alarming health consequences, is primarily transmitted by infected Aedes spp. mosquitoes but can also spread through sexual contact and from mother to child during pregnancy. While most infections are mild or asymptomatic, ZIKV has been linked to severe neurological disorders, such as Guillain-Barré syndrome (GBS) in adults and congenital Zika syndrome (CZS) in infants. While the substantial economic burden and the range of these disorders have been described, the potential long-term effects and the impact on quality of life (QoL) remain inadequatel

Zika virus (ZIKV) infection during pregnancy is associated with the development of Congenital Zika syndrome. The ZIKV epidemic in 2015 in South America exhibited an increased incidence of microcephaly cases in infants born to mothers who were infected during pregnancy, and infection in adults led to Guillain-Barré Syndrome. A growing body of literature demonstrates that the vertical transmission of ZIKV results in placental infection and injury. Further, ZIKV is transmitted to the fetus, specifically inducing apoptosis in neuronal progenitor cells, resulting in the development of microcephaly.

Arboviral infections, including West Nile virus (WNV), dengue virus (DENV), chikungunya virus (CHIKV), Zika virus (ZIKV), and, to a lesser extent, Oropouche virus (OROV), pose a heterogeneous and evolving challenge for pediatric health in Europe. The increasing presence of competent vectors and international travel has resulted in both imported infections and, for selected arboviruses, documented autochthonous transmission events. Although pediatric cases are often underrecognized, children may develop severe manifestations, including neuroinvasive disease and congenital infection. Available d

Viral infections during pregnancy can lead to several adverse outcomes, including miscarriage, stillbirth, intrauterine growth restriction, and neonatal complications, which may manifest congenital malformations and organ dysfunction. Infants who exhibit symptoms following maternal infection tend to have poorer health outcomes compared to their asymptomatic counterparts. Various viruses are known to cause birth defects, with the most common being cytomegalovirus (CMV), rubella virus, hepatitis B and C viruses, herpes simplex viruses 1 and 2, human herpesvirus 6 (HHV-6), Zika virus, and human i

The maternal-fetal interface represents a dynamic immunological frontier where pregnancy outcomes are determined by the delicate balance between host defense and microbial pathogenesis. Vertical transmission of pathogens across the placental barrier can lead to devastating consequences including fetal loss, stillbirth, prematurity, and congenital anomalies. The classic TORCH acronym encompasses Toxoplasma gondii, Other agents, Rubella virus, Cytomegalovirus, and Herpes simplex virus, though emerging viruses including Zika virus and SARS-CoV-2 have expanded this spectrum. Maternal infection with Zika virus (ZIKV), dengue virus (DENV), or chikungunya virus (CHIKV) may increase the risk of adverse pregnancy and foetal outcomes, but evidence credibility varies. The aim of this study is to evaluate and compare the strength of evidence from systematic reviews and meta-analyses on these arboviruses and pregnancy outcomes. PubMed, Web of Science, and Cochrane Library from inception to December 31, 2025 were searched. Two reviewers independently screened studies, extracted data, and assessed methodological quality using AMSTAR 2. Evidence credibility was graded as high

Zika virus (ZIKV) infection represents a critical threat to maternal-fetal health due to its ability to cross the placental barrier, converting this normally protective organ into a direct target of viral infection. This study examines the mechanisms by which ZIKV infects placental cells, including trophoblasts and Hofbauer macrophages, through the engagement of specific cellular receptors such as AXL and T-cell immunoglobulin and mucin (TIM), while simultaneously evading host immune defenses. Although the placental immune system employs interferon signaling and innate immune cells to restrict

This scoping review examines the neurological effects of maternal flavivirus infection, specifically Zika virus (ZIKV), Dengue virus (DENV), and Yellow Fever virus (YFV), on fetal brain development. Inclusion criteria focused on studies involving maternal infection and neurodevelopmental outcomes in offspring up to five years of age. The review found that ZIKV, especially the African Strain, exhibited neuroviolence and pronounced fetal brain abnormalities. Despite the presence of transplacental maternal transfer, these antibodies did not fully prevent congenital malformations, including microc

Zika virus (ZIKV), a member of the Flaviviridae family, causes significant public health concerns through congenital Zika syndrome and Guillain-Barré syndrome, yet no effective anti-ZIKV drugs or vaccines are available. To address this critical need, we conducted phenotypic, cytopathic effect-based high-throughput screening followed by medicinal chemistry optimization and discovered novel benzamide anti-ZIKV leads. Current best compounds demonstrated superior potency (EC50 values 40-400 nM, CC50 > 50 µM) compared to NITD-008, the most potent known anti-ZIKV agent. Time-of-addition assays, resi
