Strategies to enhance DNA vaccine efficacy against emerging arboviruses: lessons from ZIKA and Chikungunya viruses.

The mosquito-borne arboviruses Zika and Chikungunya pose serious health threats worldwide, often co-circulating and causing co-infections in Aedes endemic regions. While no licensed vaccine is currently available for Zika virus (ZIKV), recent progress has led to the approval of vaccines for Chikungunya virus (CHIKV) in few countries, highlighting the need for improved vaccine strategies. Zika virus (ZIKV) presents unique challenges, including its neurotropism, association with congenital abnormalities, and potential for sexual and vertical transmission, whereas Chikungunya virus (CHIKV) is cha

Zika Virus Outbreak - Bangladesh, September-December 2024.

Zika virus infection is transmitted to humans primarily through the bite of infected Aedes species mosquitoes. Although most Zika virus disease cases are mild or asymptomatic, severe neurologic complications can occur. Infection during pregnancy can result in severe congenital anomalies. In Bangladesh, Zika virus was first detected in an archived specimen from 2014; subsequently, five cases of Zika virus disease were identified in 2023. In September 2024, in response to identification of a confirmed Zika virus disease case in Bangladesh's capital, Dhaka, in a woman aged 29 years who was initia

cGAS‑STING pathway in digestive tract cancer: From molecular mechanisms to therapeutic applications (Review).

BACKGROUND: Idiopathic lymphoplasmacytic rhinitis (LPR) is a common inflammatory disorder of the nasal cavity in dogs due to unknown etiology. It is characterised by non-specific clinical signs, including nasal discharge, epistaxis and breathing problems. Diagnosis is usually based on the histopathologic identification of infiltrating plasmocytes and lymphocytes in the nasal mucosa and the exclusion of other underlying diseases. Treatment strategies include glucocorticoids, non-steroidal anti-inflammatory drugs, antibiotics and antifungal medications. The aim of this study was to evaluate the

Neglected arboviruses in South America: clinical manifestations and diagnostic challenges.

In South America, the arboviruses that are most noted are those that lead to dengue, Zika, and chikungunya. Nevertheless, there are additional arboviruses that trigger outbreaks, yet do not receive adequate attention, including Oropouche virus (OROV), Mayaro virus (MAYV), Saint Louis Encephalitis virus, Madariaga virus (MADV), and Rocío virus (ROCV). A rapid review was conducted using the databases PubMed, LILACS, Scopus, and Google Scholar. This analysis focused on studies that outline the clinical signs and diagnostic methods of neglected arboviruses impacting human populations in South Amer

Interplay between flaviviruses and the interferon response: mechanisms of immune evasion and therapeutic implications.

The genus Orthoflavivirus encompasses a group of medically significant arthropod-borne viruses, such as dengue virus (DENV), Japanese encephalitis virus West Nile virus Zika virus (ZIKV), and others, which pose persistent global public health threats. The host interferon (IFN) system constitutes a pivotal first line of defense against these viral infections. However, orthoflaviviruses have evolved a remarkable array of sophisticated strategies to antagonize both the induction and signaling pathways of type I and III IFNs. This review systematically summarizes the mechanisms by which orthoflavi

Development of a Reverse Transcription Recombinase Polymerase Amplification CRISPR/Cas12a Assay for Visual and Highly Specific Identification of Zika Virus.

Zika virus (ZIKV), a single-stranded positive-sense RNA virus of the Flaviviridae family (Flavivirus genus), causes acute febrile illness and severe congenital anomalies. Serological cross-reactivity with Dengue virus (DENV) and Chikungunya virus (CHIKV) complicates diagnosis, underscoring the urgency of developing specific point-of-care tests for early detection, outbreak mitigation, and reduced misdiagnosis risks. This study established and optimized an RPA-CRISPR/Cas12a assay for the rapid, visual, and highly specific detection of ZIKV. Primers and crRNAs targeting the highly conserved caps

Marine-Derived Natural Products Against Flaviviruses: Mechanisms, Evidence, and Future Directions.

Although flaviviruses, including DENV, ZIKV and JEV, remain important causes of febrile, congenital, and neurological diseases, treatment options remain largely supportive, with limited availability of virus-specific antiviral therapies. Marine organisms and marine-derived microorganisms produce chemically distinct antiviral materials, including sulfated polysaccharides, terpenoids, alkaloids, peptides, cyclodepsipeptides, and polyketides. However, their activities range from preliminary extract-level inhibition to direct biochemical target validation, making mechanistic comparison difficult.

A Clinical Review of Congenital and Perinatal Infections: From Womb to Follow-Up.

The acronym "TORCH" was originally introduced in 1974 by Dr Nahmias to collectively describe the most common pathogens responsible for congenital and perinatal infections, including Toxoplasma gondii, others, rubella virus, cytomegalovirus (CMV), and herpes simplex virus. With advances in diagnostic capabilities and expanded epidemiologic surveillance, the list of recognized congenital pathogens has grown to include syphilis, varicella-zoster virus, Zika virus, HIV, parvovirus B19, and lymphocytic choriomeningitis virus. While the TORCH acronym continues to be widely used in clinical and educa

Understanding histotripsy ablation safety: a systematic review of pre-clinical and clinical adverse events.

BACKGROUND: Facet joint tropism is asymmetry in orientation of the bilateral facets. Some studies have shown that tropism may increase the risk of disc degeneration and herniations, as well as degenerative spondylolisthesis (DS). It remains controversial whether tropism is a pre-existing developmental phenomena or secondary to progressive remodeling of the joint structure due to degenerative changes. As such, the following study addressed the occurrence of tropism of the lower lumbar spine (i.e. L3-S1) in a degenerative spondylolisthesis patient model. METHODS: An international, multi-center

Development of Universal Primer Sets for Zika Virus Envelope Gene Amplification and Sequencing.

The Zika virus (ZIKV) remains an important public health concern owing to its association with neurological disorders and congenital abnormalities. Genetically, ZIKV is classified into two major lineages-African and Asian-and continues to evolve, with evidence of genetic diversification following its introduction into the Americas. The study of the envelope (E) gene of ZIKV is a key tool for understanding viral biology, phylogenetic analysis, and developing antiviral drugs and vaccine candidates. However, few studies have focused on designing primers capable of efficiently detecting and amplif

Assessment of Affordances in the Home Environment and Neurodevelopment of Children With Congenital Zika Syndrome.

BACKGROUND: Congenital Zika syndrome (CZS) is characterized by brain abnormalities caused by in utero exposure to the Zika virus (ZIKV), resulting in significant neurodevelopmental, motor and visual impairments in affected children. These impairments hinder their ability to engage in social interactions and explore their surroundings. However, personalized and continuous sensory stimulation, particularly within the home environment, may play a crucial role in supporting motor and visual development in these children. Therefore, this study aimed to explore the relationship between sensory stimu

Infectivity in full-term placenta of Zika viruses with different lipid profiles.

Among flaviviruses, Zika virus (ZIKV) is the only arbovirus officially recognized as a teratogenic agent, as a consequence of its ability to infect and cross the placental barrier causing congenital malformation in the fetus. While many studies have focused on understanding ZIKV pathogenesis during pregnancy, the viral mechanisms affecting fetal development remain largely unclear. In this study, we investigated ZIKV virulence in placental trophoblasts, using viruses with distinct lipid profiles. Firstly, we propagated a ZIKV strain belonging to the Asian lineage in either mammalian or mosquito

A low pre-existing anti-NS1 humoral immunity to DENV is associated with microcephaly development after gestational ZIKV exposure.

BACKGROUND: Gestational Zika virus (ZIKV) infection is associated with the development of congenital Zika syndrome (CZS), which includes microcephaly and fetal demise. The magnitude and quality of orthoflavivirus-specific humoral immunity have been previously linked to the development of CZS. However, the role of ZIKV NS1-specific humoral immunity in mothers and children with prenatal ZIKV exposure and CZS remains undefined. In addition, considering that most of the at-risk population lives in dengue virus (DENV)-endemic areas, it is not clear what is the association between pre-existing DENV

Ubiquitin-like modifier-activating enzyme 1 interacts with Zika virus NS5 and promotes viral replication in the infected cell.

Translation errors, impaired folding or environmental stressors (e.g. infection) can all lead to an increase in the presence of misfolded proteins. These activate cellular responses to their removal, including intracellular protein degradation activities. Protein ubiquitylation is involved in two major degradation pathways, the ubiquitin-proteasome system and selective autophagy. In humans, the ubiquitin-like modifier-activating enzyme 1 (UBA1) is the primary E1 enzyme in the ubiquitin conjugation cascade. Viruses have evolved to exploit protein degradation pathways to complete their infection

Association between congenital Zika syndrome and hospitalizations during early childhood: a nationwide cohort study.

OBJECTIVES: Congenital Zika Syndrome (CZS) has been linked to a wide spectrum of abnormalities. However, differences in hospitalization patterns between children with and without CZS have not yet been investigated. METHODS: We compared rates of hospital admissions for all and specific diseases, proportions of admission causes, and total length of hospital stay (LOS) between children with CZS and those without the syndrome. Adjusted incidence rate ratios (aIRR) and aLOS were estimated using negative binomial regression. RESULTS: Compared to those without CZS, the aIRR for all-cause hospitaliz

Autophagy-Mediated Downregulation of AXL and TIM-1 Promotes Sustained Zika Virus Infection.

Zika virus (ZIKV) infection can lead to a variety of clinical outcomes, including severe congenital abnormalities. The phosphatidylserine (PS) receptors AXL and TIM-1 are recognized as critical entry factors for ZIKV in vitro. However, it remains unclear if and how ZIKV regulates these receptors during infection. In this study, we investigated AXL and TIM-1 expression in human alveolar basal epithelial A549 cells, glioblastoma U87 cells, and embryonic stem cells-derived trophoblast following ZIKV infection. We found that both the Asian strain FSS13025 and the African strain MR766 of ZIKV downr

Modelling the impact and cost-effectiveness of upcoming Zika virus vaccines on congenital Zika syndrome.

BACKGROUND: Zika virus (ZIKV) continues to circulate in Southeast Asia following the 2015-2016 global epidemic, posing an ongoing risk of importation and disease spread for Singapore, a tropical city-state in the region. The virus remains a threat to pregnant women and their fetuses due to the risk of Congenital Zika Syndrome (CZS). Vaccines currently in development offer hope for reducing ZIKV infections and CZS cases. METHODS: We developed an agent-based compartmental model, incorporating Singapore's demographic dynamics, to simulate ZIKV epidemic trajectories and project the national disea

PAV-05 Naphthoquinone Potently Inhibit Zika Virus Replication in Infected Cells.

BACKGROUND: Zika (ZIKV) is a virus transmitted by mosquitoes that can cause Guillain- Barré syndrome and congenital malformations like microcephaly. Given its explosive resurgence and the resulting epidemics in 2016, the search for effective antiviral drugs has become absolutely necessary. METHODS: In this study, we examined the potential of naphthoquinone derivatives that have a sulfonamide or sulfonate group to inhibit ZIKV replication in primary cultured neurons and in Vero cells. RESULTS: In our in vitro studies, we found that PAV05 had low cytotoxicity with a CC50 of 329 μM ±3.6 for Ver
